Prognosis for patients with amyotrophic lateral sclerosis: development and validation of a personalised prediction model
Henk Jan Westeneng,Thomas P. A. Debray,Anne E. Visser,Ruben P A van Eijk,James Rooney,Andrea Calvo,Sarah Martin,Christopher J McDermott,Alexander G. Thompson,Susana Pinto,Xenia Kobeleva,Angela Rosenbohm,Beatrice Stubendorff,Helma Sommer,Bas M. Middelkoop,Annelot M. Veldink,Leonard H. van den Berg +47 more
TL;DR: An externally validated model is developed to predict survival without tracheostomy and non-invasive ventilation for more than 23 h per day in European patients with ALS and could be applied to individualised patient management, counselling, and future trial design.
Journal ArticleDOI
Neurofilament markers for ALS correlate with extent of upper and lower motor neuron disease.
Koen Poesen,Maxim De Schaepdryver,Beatrice Stubendorff,Benjamin Gille,Petra Muckova,Sindy Wendler,Tino Prell,Thomas M. Ringer,Heidrun Rhode,Olivier Stevens,Kristl G. Claeys,Goedele Couwelier,Ann D'Hondt,Nikita Lamaire,Petra Tilkin,Dimphna Van Reijen,Sarah Gourmaud,Nadin Fedtke,Bianka Heiling,Matthias Rumpel,A. Rödiger,A. Gunkel,Otto W. Witte,Claire Paquet,Rik Vandenberghe,Julian Grosskreutz,Philip Van Damme +26 more
TL;DR: This study provides Class II evidence that elevated concentrations of CSF pNfH and NfL can accurately identify patients with ALS and shows an added value as diagnostic biomarkers for ALS, whereas the prognostic value of pNFH andNfL warrants further investigation.
Journal ArticleDOI
Safety and efficacy of rasagiline as an add-on therapy to riluzole in patients with amyotrophic lateral sclerosis: a randomised, double-blind, parallel-group, placebo-controlled, phase 2 trial
Albert C. Ludolph,Joachim Schuster,Johannes Dorst,Luc Dupuis,Jens Dreyhaupt,Jochen H. Weishaupt,Jan Kassubek,Ulrike Weiland,Susanne Petri,Thomas Meyer,Julian Grosskreutz,Berthold Schrank,Matthias Boentert,Alexander Emmer,Andreas Hermann,Daniel Zeller,Johannes Prudlo,Andrea Sylvia Winkler,Torsten Grehl,Michael T. TL;DR: There was no difference between groups in the primary outcome of survival, although post-hoc analysis suggested that rasagiline might modify disease progression in patients with an initial slope of Amyotrophic Lateral Sclerosis Functional Rating Scale Revised greater than 0·5 points per month at baseline.
Journal ArticleDOI
Assessment of pulmonary function in amyotrophic lateral sclerosis: when can polygraphy help evaluate the need for non-invasive ventilation?
Tino Prell,Thomas M. Ringer,Kara Wullenkord,Philipp Garrison,A. Gunkel,Beatrice Stubendorff,Otto W. Witte,Julian Grosskreutz +7 more
TL;DR: In patients not reporting dyspnoea and having an FVC of >75%, nocturnal hypoventilation was observed in nearly every second patient, suggesting polygraphy does not provide useful additional information if the FVC is already <75% as predicted, but in patients with more or less normal lung function parameters or where lung spirometry cannot perform adequately, it can provide sufficient evidence for the need of NPPV.
Journal ArticleDOI
Down-regulation of purinergic P2X7 receptor expression and intracellular calcium dysregulation in peripheral blood mononuclear cells of patients with amyotrophic lateral sclerosis
Jingyu Liu,Tino Prell,Beatrice Stubendorff,Silke Keiner,Thomas M. Ringer,A. Gunkel,Vedrana Tadić,Nadine Goldhammer,Ayse Malci,Otto W. Witte,Julian Grosskreutz +10 more
TL;DR: The results confirm the hypothesis that Ca(2+) abnormalities in ALS are measurable in immune cells and suggest that the reduction of P2X7 receptor expression on PBMCs leads to intracellular calcium dysregulation.
