Abstract: A coelomycete with characters resembling the asexual morphs in the family Botryosphaeriaceae was isolated from a fallen leaf of an orchid collected in Thailand. Morphological and phylogenetic analyses placed the strain in Neoscytalidium. Phylogenetic relationships among Neoscytalidium species were inferred by analyzing internal transcribed spacers and large subunit of rRNA sequence data and indicate that our strain is a new species, which is introduced and illustrated herein as Neoscytalidium orchidacearum sp. nov.

Abstract: Nitroaromatic and nitroamine compounds such as 2,4,6-trinitrotoluene (TNT) are teratogenic, cytotoxic, and may cause cellular mutations in humans, animals, plants, and microorganisms. Microbial-based bioremediation technologies have been shown to offer several advantages against the cellular toxicity of nitro-organic compounds. Thus, the current study was designed to evaluate the ability of Trichoderma viride to degrade nitrogenous explosives, such as TNT, by microbiological assay and Gas chromatography-mass spectrometry (GC-MS) analysis. In this study, T. viride fungus was shown to have the ability to decompose, and TNT explosives were used at doses of 50 and 100 ppm on the respective growth media as a nitrogenous source needed for normal growth. The GC/MS analysis confirmed the biodegradable efficiency of TNT, whereas the initial retention peak of the TNT compounds disappeared, and another two peaks appeared at the retention times of 9.31 and 13.14 min. Mass spectrum analysis identified 5-(hydroxymethyl)-2-furancarboxaldehyde with the molecular formula C6H6O3 and a molecular weight of 126 g·mol-1 as the major compound, and 4-propyl benzaldehyde with a formula of C10H12O and a molecular weight of 148 g mol-1 as the minor compound, both resulting from the biodegradation of TNT by T. viride. In conclusion, T. viride could be used in microbial-based bioremediation technologies as a biological agent to eradicate the toxicity of the TNT explosive. In addition, future molecular-based studies should be conducted to clearly identify the enzymes and the corresponding genes that give T. viride the ability to degrade and remediate TNT explosives. This could help in the eradication of soils contaminated with explosives or other toxic biohazards.

Abstract: Biomaterials developed using sustainable methods and non-toxic solvents have been effectively applied as eco-friendly, sustainable reaction medium and catalysts for biological applications. This study describes the development of e-caprolactone-citric acid nanomicelles using deep eutectic solvents (DESs) in the form of choline chloride and citric acid. The solvent formation was confirmed by FT-IR and 1H NMR spectroscopy. The highest polycondensation yield of e-caprolactone (e-cp) with citric acid (CA) poly(e-cp-co-CA) was obtained in 1 : 2 DES medium. Camptothecin (CPT), used as a model anticancer agent, was encapsulated by poly(e-cp-co-CA) micelles, which act as drug carriers. Micelle formation was determined by fluorescence analysis using pyrene (CMC = 0.5 mg mL−1). The self-assembled CPT-loaded poly(e-cp-co-CA) micelles were characterized using FT-IR, XRD, SEM and TEM techniques. The size of the nanomicelles was observed to be ∼270 nm using dynamic light scattering (DLS) techniques. The encapsulation efficiency and in vitro profiles of CPT in poly(e-cp-co-CA) micelles were investigated by UV spectrometry at λmax of 350 nm. The antiproliferative effect of the encapsulated CPT, which was investigated in A549 cells, suggested that e-cp-co-CA may be a promising anticancer drug carrier.

Abstract: Muyocopron species are associated with a wide variety of plant substrates worldwide and presently 57 species epithets are listed in Index Fungorum. Species in this genus form distinctive black, dull, rounded regions on the surface of plants and the genus is probably polyphyletic. The present study clarifies the phylogenetic placement of Muyocopron and related species, using fresh tropical collections from northern Thailand. Three Muyocopron species are characterized based on analyses of combined LSU and SSU sequence datasets. Phylogenetic analyses indicate that Muyocopron species form a distinct lineage with the Dyfrolomycetales and Acrospermales lineages. The new order Muyocopronales with three new Muyocopron species is introduced based on its distinct phylogeny and unique morphological characteristics. The taxonomy and phylogenetic relationships of tropical Muyocopron species are reappraised with suggestions for future work.

Abstract: : The unpolarized absorption and circular dichroism spectra of the fundamental vibrational transitions of the chiral molecule, 4-methyl-2-oxetanone, are calculated ab initio. Harmonic force fields are obtained using Density Functional Theory (DFT), MP2, and SCF methodologies and a 5S4P2D/3S2P (TZ2P) basis set. DFT calculations use the Local Spin Density Approximation (LSDA), BLYP, and Becke3LYP (B3LYP) density functionals. Mid-IR spectra predicted using LSDA, BLYP, and B3LYP force fields are of significantly different quality, the B3LYP force field yielding spectra in clearly superior, and overall excellent, agreement with experiment. The MP2 force field yields spectra in slightly worse agreement with experiment than the B3LYP force field. The SCF force field yields spectra in poor agreement with experiment.The basis set dependence of B3LYP force fields is also explored: the 6-31G* and TZ2P basis sets give very similar results while the 3-21G basis set yields spectra in substantially worse agreements with experiment. jg

Abstract: Fusarium species, particularly Fusarium graminearum and F. culmorum, are the main cause of trichothecene type B contamination in cereals. Data on the distribution of Fusarium trichothecene genotypes in cereals in Europe are scattered in time and space. Furthermore, a common core set of related variables (sampling method, host cultivar, previous crop, etc.) that would allow more effective analysis of factors influencing the spatial and temporal population distribution, is lacking. Consequently, based on the available data, it is difficult to identify factors influencing chemotype distribution and spread at the European level. Here we describe the results of a collaborative integrated work which aims (1) to characterize the trichothecene genotypes of strains from three Fusarium species, collected over the period 2000–2013 and (2) to enhance the standardization of epidemiological data collection. Information on host plant, country of origin, sampling location, year of sampling and previous crop of 1147 F. graminearum, 479 F. culmorum, and 3 F. cortaderiae strains obtained from 17 European countries was compiled and a map of trichothecene type B genotype distribution was plotted for each species. All information on the strains was collected in a freely accessible and updatable database (www.catalogueeu.luxmcc.lu), which will serve as a starting point for epidemiological analysis of potential spatial and temporal trichothecene genotype shifts in Europe. The analysis of the currently available European dataset showed that in F. graminearum, the predominant genotype was 15-acetyldeoxynivalenol (15-ADON) (82.9%), followed by 3-acetyldeoxynivalenol (3-ADON) (13.6%), and nivalenol (NIV) (3.5%). In F. culmorum, the prevalent genotype was 3-ADON (59.9%), while the NIV genotype accounted for the remaining 40.1%. Both, geographical and temporal patterns of trichothecene genotypes distribution were identified.
