Abstract: Summary. The allergenicity and antigenicity of various airborne fungi isolated from the atmosphere of Riyadh were studied. Protein nitrogen contents were estimated and found to range from 0.9 mg ml−1 for Cladosporium to 2.1 mg ml−1 for Aspergillus extracts. Sodium dodecyl sulphate–polyacrylamide gel electrophoresis analysis for those extracts exhibited a number of protein bands of higher molecular weight between 13 and 80 kDa for Alternaria, Ulocladium, Penicillium, Aspergillus and Cladosporium. Extracts in both aqueous and lyophilized forms were sterilized and tested for diagnostic skin prick test in 100 consecutive patients having bronchial asthma and allergic rhinitis. Overall, 13% of patients reacted positively to fungal extracts, revealing allergic sensitization to these fungi. These findings necessitate further investigation as regards the purification and characterization of these local extracts for better diagnostic use in patients in Saudi Arabia. Zusammenfassung. Es wurde die Allergenitat und Antigenitat mehrerer luftburtiger Pilze untersucht, die aus der Luft von Riad isoliert worden waren. Hierzu wurden Pilzextrakte hergestellt, deren Proteinstickstoffgehalt zwischen 0.9 mg ml−1 bei Cladosporium und 2.1 mg ml−1 bei Aspergillus lag. Die SDS–PAGE-Analyse zeigte eine Anzahl von Proteinbanden hoheren Molekulargewichts zwischen 13 und 80 kDa fur Alternaria, Ulocladium, Penicillium, Aspergillus und Cladosporium. Die sowohl wassrigen wie lyophilisierten Extrakte wurden sterilisiert und an 100 unausgewahlten Patienten mit Bronchialasthma und allergischer Rhinitis im Pricktest getestet. Ingesamt 13% der Patienten reagierten auf die Extrakte positiv, was fur eine allergische Sensibilisierung gegen diese Pilze spricht.

Abstract: In efforts to improve plant productivity and enhance defense mechanisms against biotic and abiotic stresses, endophytic bacteria have been used as an alternative to chemical fertilizers and pesticides. In the current study, 25 endophytic microbes recovered from plant organs of Triticum aestivum L. (wheat) were assessed for biotic (phyto-fungal pathogens) and abiotic (salinity, drought, and heavy metal) stress tolerance. Among the recovered isolates, BPR-9 tolerated maximum salinity (18% NaCl), drought (15% PEG-6000), and heavy metals (µg mL−1): Cd (1200), Cr (1000), Cu (1000), Pb (800), and Hg (30). Based on phenotypic and biochemical characteristics, as well as 16S rDNA gene sequencing, endophytic isolate BPR-9 was recognized as Priestia aryabhattai (accession no. OM743254.1). This isolate was revealed as a powerful multi-stress-tolerant crop growth promoter after extensive in-vitro testing for plant growth-promoting attributes, nutrient (phosphate, P; potassium, K; and zinc, Zn) solubilization efficiency, extracellular enzyme (protease, cellulase, amylase, lipase, and pectinase) synthesis, and potential for antagonistic activity against important fungal pathogens viz. Alternaria solani, Rhizoctonia solani, Fusarium oxysporum, and Ustilaginoidea virens. At elevated salt levels, increases were noted in indole-3-acetic acid; siderophores; P, K, and Zn-solubilization; ACC deaminase; and ammonia synthesized by Priestia aryabhattai. Additionally, under in-vitro plant bioassays, wheat seedlings inoculated with P. aryabhattai experienced superior growth compared to non-inoculated seedlings in high salinity (0–15% NaCl) environment. Under NaCl stress, germination rate, plant length, vigor indices, and leaf pigments of wheat seedlings significantly increased following P. aryabhattai inoculation. Furthermore, at 2%-NaCl, B. aryabhattai greatly and significantly (p ≤ 0.05) decreased relative leaf water content, membrane damage, and electrolyte leakage compared with the non-inoculated control. Catalase, superoxide dismutase, and peroxidase activity increased by 29, 32, and 21%, respectively, in wheat seedlings exposed to 2% NaCl and inoculated with the bacteria. The present findings demonstrate that endophytic P. aryabhattai strains might be used in the future as a multi-stress reducer and crop growth promoter in agronomically important crops including cereals.

Abstract: During our studies of palm fungi in Thailand we identified a new species of Barriopsis on a petiole of Archontophoenix alexandrae, which we introduce herein as B. archontophoenicis. The new species is compared with other species in the genus Barriopsis and differs in its epapillate ostiole and smaller ascospores. Phylogenetic analyses of combined ITS, LSU, SSU and TEF1-alpha sequence data also show the species to be distinct.

Abstract: : The unpolarized absorption and circular dichroism spectra of the fundamental vibrational transitions of the chiral molecule, 4-methyl-2-oxetanone, are calculated ab initio. Harmonic force fields are obtained using Density Functional Theory (DFT), MP2, and SCF methodologies and a 5S4P2D/3S2P (TZ2P) basis set. DFT calculations use the Local Spin Density Approximation (LSDA), BLYP, and Becke3LYP (B3LYP) density functionals. Mid-IR spectra predicted using LSDA, BLYP, and B3LYP force fields are of significantly different quality, the B3LYP force field yielding spectra in clearly superior, and overall excellent, agreement with experiment. The MP2 force field yields spectra in slightly worse agreement with experiment than the B3LYP force field. The SCF force field yields spectra in poor agreement with experiment.The basis set dependence of B3LYP force fields is also explored: the 6-31G* and TZ2P basis sets give very similar results while the 3-21G basis set yields spectra in substantially worse agreements with experiment. jg

Abstract: Fusarium species, particularly Fusarium graminearum and F. culmorum, are the main cause of trichothecene type B contamination in cereals. Data on the distribution of Fusarium trichothecene genotypes in cereals in Europe are scattered in time and space. Furthermore, a common core set of related variables (sampling method, host cultivar, previous crop, etc.) that would allow more effective analysis of factors influencing the spatial and temporal population distribution, is lacking. Consequently, based on the available data, it is difficult to identify factors influencing chemotype distribution and spread at the European level. Here we describe the results of a collaborative integrated work which aims (1) to characterize the trichothecene genotypes of strains from three Fusarium species, collected over the period 2000–2013 and (2) to enhance the standardization of epidemiological data collection. Information on host plant, country of origin, sampling location, year of sampling and previous crop of 1147 F. graminearum, 479 F. culmorum, and 3 F. cortaderiae strains obtained from 17 European countries was compiled and a map of trichothecene type B genotype distribution was plotted for each species. All information on the strains was collected in a freely accessible and updatable database (www.catalogueeu.luxmcc.lu), which will serve as a starting point for epidemiological analysis of potential spatial and temporal trichothecene genotype shifts in Europe. The analysis of the currently available European dataset showed that in F. graminearum, the predominant genotype was 15-acetyldeoxynivalenol (15-ADON) (82.9%), followed by 3-acetyldeoxynivalenol (3-ADON) (13.6%), and nivalenol (NIV) (3.5%). In F. culmorum, the prevalent genotype was 3-ADON (59.9%), while the NIV genotype accounted for the remaining 40.1%. Both, geographical and temporal patterns of trichothecene genotypes distribution were identified.
