Abstract: An undescribed Nia species was recorded from intertidal decayed wood of Avicennia marina collected from Red Sea mangroves in Saudi Arabia. Nia lenicarpa sp. nov. is characterized by smooth basidiomes, growing singly or in groups of 2 to 10 confluent basidiomes, with a three-layered, thick peridium and 4-spored basidia. Nia lenicarpa differs from N. vibrissa by having smaller basidiomes without peridial hairs but with thick peridia, larger basidia and basidiospores. Phylogenetic analyses of SSU and LSU rDNA placed N. lenicarpa in a monophyletic clade with two N. vibrissa clades. There are no molecular data available for the other two Nia species; N. epidermoidea and N. globispora. The morphology of N. vibrissa from driftwood collected from a beach in Japan is illustrated. Vertical sections of basidiomes of the Japanese material showed the presence of an outer peridium that is formed by the tips of the peridial hairs which was not reported before. This supports the presence of several cryptic species in the N. vibrissa complex.

Abstract: Aspergillus species are regularly involved in human broncho-pulmonary diseases, mainly in immunocompromised patients. The essential oils extracted from three different plants were tested for their inhibitory effect on the growth of five pathogenic Aspergilli including Aspergillus flavus, Aspergillus fumigatus, Aspergillus niger, Aspergillus versicolor and Aspergillus terreus which are pathogens with clinical importance. The results of the antimicrobial tests showed that the isolated essential oils inhibited the growth of Aspergillus species to varying degrees. The Ocimum basilicum oil completely inhibited the mycelial growth and spore germination of A. terreus and A. fumigatus at 200 and 250 ppm. In addition, this oil showed a very strong antifungal activity against the mycelial growth and spore germination of A. flavus and A. versicolor with 97.7, 97.5, 91.2 and 93.5%, respectively, when compared with control. The essential oil extracted from Eucalyptus globulus also exhibited significant antifungal activity against the Aspergillus species tested. This oil completely inhibited the mycelial growth and spore germination of A. fumigatus. Conversely, Nigella sativa oil exhibited moderate inhibitory activity against all the tested Aspergillus species. The present study supports the proposition that plant essential oils may have a role in both pharmaceutical and agricultural practices.

Abstract: Transition metal oxide NPs have delivered wide applications in various fields. Therefore, in this study, a novel fungus, Alternaria sp. (NCBI Accession No: MT982648) was isolated and characterized from the vicinity of medicinal plants. Eventually, in this method extracted proteins from isolated fungus were utilized to synthesize highly biocompatible zinc nanoparticles (ZnO NPs). The various physical techniques including UV-visible spectroscopy, TEM, HR-TEM, XRD, DLS, zeta potential, and FTIR were used to characterize particles. The UV-visible absorption (λMax) and binding energy for the as-synthesized particles were found to be 329 nm and 3.91 eV, respectively. Further, the polydispersed particles were revealed to have regular crystallinity with hexagonal wurtzite phase of ZnO with the spacing of ~2.46 Å under XRD and HR-TEM. The average size of a particle under TEM was found to be ~18 nm. The evaluation of various surface functional groups of particles was done by FTIR. The average hydrodynamic diameter of particles was found to be ~57 d. nm with 0.44 particle distribution index whereas the nanoemulsion stability was explained by Zeta potential (-9.47 mV). These particles were found to exhibit potential antibacterial and anticancer activities. They were found to be bactericidal against S. abony (MIC 5.73 μg/mL); B. pumilis (MIC 6.64 μg/mL); K. pneumonia (MIC 14.4 μg/mL); E. coli (MIC 8.7 μg/mL); B. subtilis (MIC 5.63 μg/mL) and S. aureus (MIC 12.04 μg/mL). Further, they are also found to be concentration-dependent anticancer and inhibited the growth of A549 cells (IC50-65.3 μg/mL) whereas they were found to demonstrate no any cytotoxicity against NRK normal kidney cell line. The internalization of particles into the nucleus (i.e., nuclear fragmentation and DNA damage) was confirmed by DAPI staining. The intracellular particles were found to generate excessive ROS. Further, the anticancer potential was also estimated by noticing a hike in oxidative stress parameters, cell viability, cell morphology, and change in mitochondrial membrane potential. We effectively synthesized potentially potent antibacterial and anticancer novel bioengineered ZnO NPs.

Abstract: : The unpolarized absorption and circular dichroism spectra of the fundamental vibrational transitions of the chiral molecule, 4-methyl-2-oxetanone, are calculated ab initio. Harmonic force fields are obtained using Density Functional Theory (DFT), MP2, and SCF methodologies and a 5S4P2D/3S2P (TZ2P) basis set. DFT calculations use the Local Spin Density Approximation (LSDA), BLYP, and Becke3LYP (B3LYP) density functionals. Mid-IR spectra predicted using LSDA, BLYP, and B3LYP force fields are of significantly different quality, the B3LYP force field yielding spectra in clearly superior, and overall excellent, agreement with experiment. The MP2 force field yields spectra in slightly worse agreement with experiment than the B3LYP force field. The SCF force field yields spectra in poor agreement with experiment.The basis set dependence of B3LYP force fields is also explored: the 6-31G* and TZ2P basis sets give very similar results while the 3-21G basis set yields spectra in substantially worse agreements with experiment. jg

Abstract: Fusarium species, particularly Fusarium graminearum and F. culmorum, are the main cause of trichothecene type B contamination in cereals. Data on the distribution of Fusarium trichothecene genotypes in cereals in Europe are scattered in time and space. Furthermore, a common core set of related variables (sampling method, host cultivar, previous crop, etc.) that would allow more effective analysis of factors influencing the spatial and temporal population distribution, is lacking. Consequently, based on the available data, it is difficult to identify factors influencing chemotype distribution and spread at the European level. Here we describe the results of a collaborative integrated work which aims (1) to characterize the trichothecene genotypes of strains from three Fusarium species, collected over the period 2000–2013 and (2) to enhance the standardization of epidemiological data collection. Information on host plant, country of origin, sampling location, year of sampling and previous crop of 1147 F. graminearum, 479 F. culmorum, and 3 F. cortaderiae strains obtained from 17 European countries was compiled and a map of trichothecene type B genotype distribution was plotted for each species. All information on the strains was collected in a freely accessible and updatable database (www.catalogueeu.luxmcc.lu), which will serve as a starting point for epidemiological analysis of potential spatial and temporal trichothecene genotype shifts in Europe. The analysis of the currently available European dataset showed that in F. graminearum, the predominant genotype was 15-acetyldeoxynivalenol (15-ADON) (82.9%), followed by 3-acetyldeoxynivalenol (3-ADON) (13.6%), and nivalenol (NIV) (3.5%). In F. culmorum, the prevalent genotype was 3-ADON (59.9%), while the NIV genotype accounted for the remaining 40.1%. Both, geographical and temporal patterns of trichothecene genotypes distribution were identified.
