The review demonstrates the progress in the chemistry of calix[4]resorcinarenes over the last decade. Special attention is paid to their synthesis, conformational behaviour, functionalization and ability to serve as cavitands and capsules, to form monolayers and host–guest complexes. Practical applications of calix[4]resorcinarenes and their derivatives in various fields are surveyed.

Chemistry of calix[4]resorcinarenes

A novel class of antibiotics was isolated from cultures of Streptomyces coeruleorubidus strain AB 1183F-64. The antimicrobial activity of the pacidamycins is selective against Pseudomonas aeruginosa. The various congeners are nucleoside peptides which differ in the terminal amino acid residues. The structures were determined using MS-MS and 2D NMR techniques.

We report a new, single-stage method for the synthesis of 2-arylpyrrolidines, based on an acid-catalyzed reaction of 2-naphthol with γ-ureidoacetals, enabling the preparation of the target compounds under mild conditions, while avoiding costly reagents and catalysts.

N-(4,4-diethoxybutyl)pyrimidin-2-amine undergoes acid-catalyzed intramolecular cyclisation in the presence of phenols leading to the formation of 2-(2-arylpyrrolidin-1-yl)pyrimidine derivatives. Target compounds were obtained with moderate to good yields by operationally simple one-step procedure.

Facile synthesis of 2-(2-arylpyrrolidin-1-yl)pyrimidines via acid-catalyzed reaction of N-(4,4-diethoxybutyl)pyrimidin-2-amine with phenols

A new method based on the acid-catalyzed reaction of pyridoxal with polyfunctional phenols has been developed for the synthesis of potentially biologically active 1-aryl-6-methyl-1,3-dihydrofuro[3,4- c ]pyridin-7-ols. Advantages of this method include a good yield of the target furopyridines and the use of readily available hydrochloric acid as a catalyst. The possibility of extending the proposed approach to nu­cleo­philes other than phenols was demonstrated by the synthesis of the known biologically active compound 1-(5-hydroxy-3-methyl-1-phenyl-1 H -pyrazol-4-yl)-6-methyl-1,3-dihydrofuro[3,4- c ]pyridin-7-ol hydrochloride (TM2002).
