Journal ArticleDOI
Epilepsy, hyperalgesia, impaired memory, and loss of pre- and postsynaptic GABA(B) responses in mice lacking GABA(B(1))
Valerie Schuler,Christian Lüscher,Christophe Blanchet,Norman Klix,Gilles Sansig,Klaus Klebs,Markus Schmutz,Jakob Heid,Clive Gentry,Laszlo Urban,Alyson Fox,Will P.J.M Spooren,Anne-Lise Jaton,Jean-Marie Vigouret,Mario F. Pozza,Peter H. Kelly,Johannes Mosbacher,Wolfgang Froestl,Edgar Käslin,Reinhard Korn,Serge Bischoff,Klemens Kaupmann,Herman van der Putten,Bernhard Bettler +23 more
TL;DR: It is demonstrated that Balb/c mice lacking the GABA(B(1)) subunit are viable, exhibit spontaneous seizures, hyperalgesia, hyperlocomotor activity, and memory impairment, and casts doubt on the existence of proposed receptor subtypes.
Journal ArticleDOI
Pain related behaviour in two models of osteoarthritis in the rat knee.
Janet Fernihough,Clive Gentry,Marzia Malcangio,Alyson Fox,John Rediske,Theodore C. Pellas,Bruce L. Kidd,Stuart Bevan,Janet Winter +8 more
TL;DR: An analysis of pain related behaviours in two models of OA in the rat: partial medial meniscectomy and iodoacetate injection provides a basis for studies on the mechanisms of pain in OA, and for development of novel therapeutic analgesics.
Journal ArticleDOI
siRNA relieves chronic neuropathic pain
Gabriele Dorn,Sadhana Patel,Glen Wotherspoon,Maja Hemmings-Mieszczak,Jane Barclay,Francois Natt,Pierre Martin,Stuart Bevan,Alyson Fox,Pam Ganju,William Leonard Wishart,Jonathan Hall +11 more
TL;DR: It is shown that RNAi can block a pathophysiological pain response and provide relief from neuropathic pain in a rat disease model by down regulating an endogenous, neuronally expressed gene.
Journal ArticleDOI
VR1 protein expression increases in undamaged DRG neurons after partial nerve injury.
TL;DR: Examination of the expression of the sensory neuron‐specific cation channel Vanilloid Receptor 1 (VR1), an important transducer of noxious stimuli, in three models of nerve injury in the rat found that VR1‐IR persisted in the transected sciatic nerve proximal to the lesion, despite its down‐regulation in the damaged neuronal somata.
Journal ArticleDOI
The VR1 Antagonist Capsazepine Reverses Mechanical Hyperalgesia in Models of Inflammatory and Neuropathic Pain
Katharine Walker,Laszlo Urban,Stephen J. Medhurst,Sadhana Patel,Mohanjit Panesar,Alyson Fox,Peter McIntyre +6 more
TL;DR: The data show that VR1 antagonists have antihyperalgesic activity in animal models of chronic inflammatory and neuropathic pain, and illustrate species differences in the in vivo pharmacology of VR1 that correlate with differences in pharmacology previously seen in vitro.
