Journal ArticleDOI
Type IIA topoisomerase inhibition by a new class of antibacterial agents.
Benjamin D. Bax,Pan F. Chan,Drake S. Eggleston,Drake S. Eggleston,Andrew P. Fosberry,Daniel R. Gentry,Fabrice Gorrec,Fabrice Gorrec,Ilaria Giordano,Michael M. Hann,Alan Joseph Hennessy,Martin Hibbs,Jianzhong Huang,Emma J. Jones,Jo J. Jones,Kristin K. Brown,Ceri J. Lewis,Earl May,Earl May,Martin R. Saunders,Onkar M. P. Singh,Claus Spitzfaden,Carol Shen,Anthony Shillings,Andrew J. Theobald,Alexandre Wohlkonig,Alexandre Wohlkonig,Neil D. Pearson,Michael N. Gwynn +28 more
TL;DR: This work provides new insights into the mechanism of topoisomerase action and a platform for structure-based drug design of a new class of antibacterial agents against a clinically proven, but conformationally flexible, enzyme class.
Journal ArticleDOI
Structural basis of DNA gyrase inhibition by antibacterial QPT-1, anticancer drug etoposide and moxifloxacin.
Pan F. Chan,Velupillai Srikannathasan,Jianzhong Huang,Haifeng Cui,Andrew P. Fosberry,Minghua Gu,Michael M. Hann,Martin Hibbs,Paul Homes,Karen A. Ingraham,Jason Pizzollo,Carol Shen,Anthony Shillings,Claus Spitzfaden,Robert Tanner,Andrew J. Theobald,Robert A. Stavenger,Benjamin D. Bax,Michael N. Gwynn +18 more
TL;DR: The first co-crystal structures of the antibacterial QPT-1 and the anticancer drug etoposide with Staphylococcus aureus DNA gyrase are reported, showing binding at the same sites in the cleaved DNA as the fluoroquinolone moxifloxacin.
Journal ArticleDOI
Crystallization and initial crystallographic analysis of covalent DNA-cleavage complexes of Staphyloccocus aureus DNA gyrase with QPT-1, moxifloxacin and etoposide
Velupillai Srikannathasan,Alexandre Wohlkonig,Anthony Shillings,Onkar M. P. Singh,Pan F. Chan,Jianzhong Huang,Michael N. Gwynn,Andrew P. Fosberry,Paul Homes,Martin Hibbs,Andrew J. Theobald,Claus Spitzfaden,Benjamin D. Bax +12 more
TL;DR: A G/T mismatch introduced at the ends of the DNA duplexes facilitated the crystallization of slightly asymmetric complexes of the inherently flexible DNA-cleavage complexes of S. aureus DNA gyrase.
Journal ArticleDOI
Discovery and Biochemical Characterization of Plasmodium Thioredoxin Reductase Inhibitors from an Antimalarial Set
Andrew J. Theobald,Iván Caballero,Isabel Coma,Gonzalo Colmenarejo,Concepción Cid,Francisco-Javier Gamo,Martin Hibbs,Amy L Bass,Daniel Thomas +8 more
TL;DR: High-purity, full-length, recombinant native enzyme from four Plasmodium species, and thioredoxin substrates from P. falciparum and Rattus norvegicus are produced using a unique, high-throughput, in vitro native substrate assay.
Journal ArticleDOI
High-yield production and characterization of biologically active GST-tagged human topoisomerase IIα protein in insect cells for the development of a high-throughput assay
Praveen Kumar Singh,Pan F. Chan,Martin Hibbs,Maria-Jesus Vazquez,Delfina C. Segura,Daniel Thomas,Andrew J. Theobald,Kathleen T. Gallagher,Namir J. Hassan +8 more
TL;DR: A method for the rapid over-expression and purification of active GST-tagged human topoisomerase IIα using the baculovirus mediated insect cell expression system is reported and validated the enzyme and the high-throughput fluorescence assay as tools for inhibitor identification and selectivity studies.
