Laccase immobilized on functionalized cellulose nanofiber/alginate composite hydrogel for efficient bisphenol A degradation from polluted water

Polyphenolic materials are a class of fascinating and versatile bioinspired materials for biointerfacial engineering. In particular, due to the presence of active chemical groups, a series of unique physicochemical properties become accessible and tunable of the as-prepared polyphenolic platforms, which could delicately regulate the cell activities via cell-material contact-dependent interactions. More interestingly, polyphenols could also affect the cell behaviors via cell-material contact-independent manner, which arise due to their intrinsically functional characteristics (e.g., antioxidant and photothermal behaviors). As such, a comprehensive understanding on the relationship between material properties and desired biomedical applications, as well as the underlying mechanism at the cellular and molecular level would provide material design principles and accelerate the lab-to-clinic translation of polyphenolic platforms. In this review, we firstly give a brief overview of cell hallmarks governed by surrounding cues, followed by the introduction of polyphenolic material engineering strategies. Subsequently, a detailed discussion on cell-polyphenols contact-dependent interfacial interaction and contact-independent interaction was also carefully provided. Lastly, their biomedical applications were elaborated. We believe that this review could provide guidances for the rational material design of multifunctional polyphenols and extend their application window.

Versatile polyphenolic platforms in regulating cell biology.

Synthetic polymers (SyPs) have found many relevant applications niches in biomedical engineering. Their mechanical properties, defined chemical structure, batch to batch consistency are attractive features that render them superior (at least in some senses) to natural polymers. However, most SyPs must be functionalized in order to properly serve for an intended biological application. Here we describe recent strategies used to functionalize SyPs for tissue engineering and related applications. We review functionalization strategies to promote cell–polymer interactions, to direct cell fate, and to induce extra-cellular matrix (ECM) (or tissue) remodeling. Besides chemical functionalization, we describe a selection of methods (i.e., casting and particle leaching, thermally induced phase separation, electrospinning, gas foaming, and 3D printing) that have been used in recent literature to modify the architecture/topography of scafolds made of SyPs. We also review recent literature on SyPs functionalization to impart antimicrobial or conductive character and to engineer actuators for tissue engineering applications. Finally, we briefly discuss some of the trends on the engineering of SyPs (i.e., bioinspiration, 3D bioprinting, nucleic-acid-based platforms) that are currently reshaping tissue engineering.

The aim of the current work was to study the physicochemical properties and biological activity of different types of porous granules containing silver or gallium ions. Firstly, hydroxyapatites powders doped with Ga3+ or Ag+ were synthesized by the standard wet method. Then, the obtained powders were used to fabricate ceramic microgranules (AgM and GaM) and alginate/hydroxyapatite composite granules (AgT and GaT). The ceramic microgranules were also used to prepare a third type of granules (AgMT and GaMT) containing silver or gallium, respectively. All the granules turned out to be porous, except that the AgT and GaT granules were characterized by higher porosity and a better developed specific surface, whereas the microgranules had very fine, numerous micropores. The granules revealed a slow release of the substituted ions. All the granules except AgT were classified as non-cytotoxic according to the neutral red uptake (NRU) test and the MTT assay. The obtained powders and granules were subjected to various antibacterial test towards the following four different bacterial strains: Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa and Escherichia coli. The Ag-containing materials revealed high antibacterial activity.

Antibacterial and Cytotoxicity Evaluation of New Hydroxyapatite-Based Granules Containing Silver or Gallium Ions with Potential Use as Bone Substitutes

Polyester (PET) prostheses are commonly used in reconstructive vascular surgery. The most serious complication after implantation is early or late infection of the graft. Therefore, there is high demand to protect prosthesis against bacterial adhesion and biofilm development. For this reason, in this work PET prostheses were first coated by highly adhesive polycatecholamine layer. The grafts were then coupled with gentamicin and studied in relation to morphological and structural properties, biological safety (contact with blood, reaction of vascular endothelial cells (HUVEC), Danio rerio fish), drug release and antibacterial activity. Among two tested catecholamine monomers, L-DOPA was found to be more effective precursor in this process than dopamine. For L-DOPA, assistance of Cu2+, Mg2+ and Na+ ions seems to increase the amount of further immobilized drug. Coated prostheses exhibited greater human endothelial cell proliferation increase and lower cytotoxic effect than uncoated. The modification reduced the hemolysis observed for pristine commercial graft and limited the rate of abnormalities in D. rerio larvae, confirming the safety of the proposed modification. The coating allowed to double the amount of immobilized antibiotic in comparison with uncoated graft which resulted in increased antibacterial activity and reduced bacterial adhesion against 4 bacterial strains prevalent in biomaterials infections. Overall, poly(L-DOPA)-coatings deposited on PET vascular grafts can effectively functionalize these prostheses for higher safety in biomedical applications. 

Polycatecholamine and gentamicin as modifiers for antibacterial and blood-biocompatible polyester vascular prostheses

Multipurpose Fe3O4@APTES–Ag heterostructures for mutual heat generation, SERS probing, and antimicrobial activity were fabricated using a three-step process. Silver metallic particles were precipitated on a thin silica shell that served as an interlayer with Fe3O4 nanocubes. The structural properties were studied by means of the powder X-ray diffraction technique, and selected area electron diffraction. Particle size, distribution, and morphology were evaluated using transmission electron microscopy, while element mapping was performed using the STEM-EDS technique. The presence of the silica shell and the effectiveness of the Ag reduction were checked by FTIR-ATR spectroscopy. The heat generation ability was studied by using AMF and NIR contactless external stimulations working separately and simultaneously. We demonstrated that the dual mode stimulation leads to a SAR (specific absorption rate) of 1000 W g−1 with the predominant role of the mechanism associated with the light interaction. The SERS effect was recorded with the use of the R6G standard molecule showing high capability of the heterostructures for Raman signal augmentation. Fe3O4 nanocubes decorated with Ag particles have shown antibacterial activity against P. aeruginosa. The Fe3O4@APTES–Ag presents promising potential as a multipurpose platform for biological applications ranging from photomagnetic therapies, to analytical probes exploiting the SERS effect and antibacterial activity.
