Screening barley varieties tolerant to drought stress based on tolerant indices

Considering the importance of salinity stress and genotype screening under stress conditions, the current study evaluated faba bean genotypes in response to saline stress and identified those that were tolerant and determined the influential ratio of each yield component on seed yield under both conditions. As a result, 12 faba bean genotypes were tested under 2 levels of salt stress (100 mM and 200 mM) and a control. The study was analyzed with multivariate (descriptive, ANOVA, PCA, biplot, cluster analysis, and indices) analysis techniques to determine the tolerance level of each genotype. Similarly, the cluster analysis results reported that faba bean genotypes were divided into two groups under the control and 100 mM salinity levels; however, the 200 mM salinity level recorded three groups of faba bean genotypes, showing that salinity stress may limit phenotypic variability among faba bean genotypes. The descriptive analysis results showed a wide range of diversity among the studied characteristics under control and salinity stress conditions. The number of seeds/plants recorded a significant association with plant height (cm) (PH), stomatal conductance (SC), days to flowering (DF), the number of pods, and seed weight (g) (SW); however, an insignificant association was recorded with leaf temperature (LT), fresh weight (g) (FW), Na+, K+, and Na/K ratio. The first three principal components (PCs) represent 81.45% of the variance among the studied traits. The most significant characteristics that contributed the most to the diversity were (PH, leaf area, SPAD reading, stomatal conductance, DF, number of pods/plants, number of seeds/pods, SW, K, and total chlorophyll content); however, the significant genotypes (Hassawi-2, Sakha, ILB-4347, Misr-3, FLIP12501FB) were present in PC1 under both conditions. The results predicted that Hassawi-2, ILB-4347, Sakha, Misr-3, and Flip12501FB were the significant (tolerant) genotypes. However, FLIP12504FB represents a sensitive genotype based on its final grain yield. The results of the indices also recorded significant index correlations with grain yield, demonstrating that these indices are effective tools for screening faba bean-tolerant genotypes under salinity stress conditions.

Agronomical and Physiological Responses of Faba Bean Genotypes to Salt Stress

Salicylic acid (SA) has been shown to ameliorate drought stress. However, physiological and biochemical mechanisms involved in drought stress tolerance induced by SA in plants have not been well understood. Thus, this study aimed to study the role of SA application on enzymatic and non-enzymatic antioxidants, photosynthetic performance, and plant growth in A. chilensis plants subjected to moderate drought stress. One-year-old A. chilensis plants were subjected to 100% and 60% of field capacity. When plants reached moderate drought stress (average of stem water potential of −1.0 MPa, considered as moderate drought stress), a single SA application was performed on plants. Then, physiological and biochemical features were determined at different times during 14 days. Our study showed that SA application increased 13.5% plant growth and recovered 41.9% AN and 40.7% gs in drought-stressed plants on day 3 compared to drought-stressed plants without SA application. Interestingly, SOD and APX activities were increased 85% and 60%, respectively, in drought-stressed SA-treated plants on day 3. Likewise, SA improved 30% total phenolic content and 60% antioxidant capacity in drought-stressed A. chilensis plants. Our study provides insight into the SA mechanism to tolerate moderate drought stress in A. chilensis plants.

Salicylic Acid Improves Antioxidant Defense System and Photosynthetic Performance in Aristotelia chilensis Plants Subjected to Moderate Drought Stress

Superoxide dismutase (SOD) family in durum wheat: promising candidates for improving crop resilience

The main objectives of this study were to find the possible structural association between the activity of enzymatic antioxidants and the grain yield of triticale plants as well as identifying the genotypic variability which might be effective on this association. Accordingly, expression levels of superoxide dismutase (SOD) isozymes (Mn-SOD, Cu/Zn-SOD, and Fe-SOD) were appraised to distinguish any possible relationship between SOD expression and drought resistance of triticale. A novel analytical method for distinguishing elite genotypes based on measured features was proposed. Additionally, a new programing based on SAS-language (IML) was introduced to estimate the genetic parameters rooted from combined ANOVA model (linear mixed model), which is capable of being used in any field study other than the current one. Thirty genotypes of triticale were studied under normal and drought stress conditions during 6 years (three different locations). Accordingly, based on the results of genetic variability, heatmap analysis, biplot graph, and clustering technique, two genotypes with the highest genetic distance were selected to appraise the differential expression profiling of three SOD isozyme in shoot and root organs. Field experiments and bioinformatics results showed that superoxide dismutase (SOD) was the most influential antioxidant in resistance of triticale to drought stress; therefore, it could be used as an indirect selection index in early stages to distinguish resistant genotypes to drought stress. Additionally, Mn-SOD and Fe-SOD showed roughly similar expression levels for both genotypes under drought stress. However, Cu/Zn-SOD expression level was higher in root and shoot of the tolerant genotype than the susceptible genotype. Heatmap analysis that is applied for the first time to screen suitable genotypes, showed to be highly capable of distinguishing elite genotypes and pointing out the proper features for selection criteria. Bioinformatics results indicated that SOD is more important than other enzymatic antioxidant for being considered as selection criteria or candidate gene for transgenic purposes. Based on expressional results, Mn-SOD announced as a general isozyme that is probably highly expressed in most of the species, while, Cu/Zn-SOD was introduced as a genotype specific isozyme that is likely more expressed in tolerant genotypes
