Copper-mediated coupling reactions and their applications in natural products and designed biomolecules synthesis.

Ferrostatin-1 (Fer-1) inhibits ferroptosis, a form of regulated, oxidative, nonapoptotic cell death. We found that Fer-1 inhibited cell death in cellular models of Huntington’s disease (HD), periventricular leukomalacia (PVL), and kidney dysfunction; Fer-1 inhibited lipid peroxidation, but not mitochondrial reactive oxygen species formation or lysosomal membrane permeability. We developed a mechanistic model to explain the activity of Fer-1, which guided the development of ferrostatins with improved properties. These studies suggest numerous therapeutic uses for ferrostatins, and that lipid peroxidation mediates diverse disease phenotypes.

Predicting the electronic structure of extended organic molecules constitutes an important fundamental task of modern chemistry. Studies of electronic excitations, charge-transfer, energy-transfer, and isomerization of conjugated systems form the basis for our understanding of the photophysics and photochemistry of complex molecules1-3 as well as organic nanostructures and supramolecular assemblies.4,5 Photosynthesis and other photochemical biological processes that constitute the basis of life on Earth involve assemblies of conjugated chromophores such as porphyrins, chlorophylls, and carotenoids.6-8 Apart from the fundamental interest, these studies are also closely connected to numerous important technological applications.9 Conjugated polymers are primary candidates for new organic optical materials with large nonlinear polarizabilities.10-19 Potential applications include electroluminescence, light emitting diodes, ultrafast switches, photodetectors, biosensors, and optical limiting materials.20-27 Optical spectroscopy which allows chemists and physicists to probe the dynamics of vibrations and electronic excitations of molecules and solids is a powerful tool for the study of molecular electronic structure. The theoretical techniques used for describing spectra of isolated small molecules are usually quite different from those of molecular crystals, and many intermediate size systems, such as clusters and polymers, are not readily described by the methods developed for either of these limiting cases.28

In this work, studies on the arylation of anilines derived from dehydroabietic acid, the main component of disproportionated rosin, are presented. The redox properties of the new diarylamines were investigated by cyclic voltammetry, and their free radical scavenging activity was tested by reduction of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. Three of the diarylamines with lower oxidation potential proved to be as active as isopropyldiphenylamine (IPPD) and superior to tert-butylhydroxytoluene (BHT), both commercially available synthetic antioxidants.

The demand for novel effective antioxidant-based drugs has led to the synthesis and evaluation of the antioxidant potential in several molecules derived from natural compounds. In this work the in vitro antioxidant activity of an abietic acid-derived catechol (methyl 11,12-dihydroxyabietate-8,11,13-trien-18-oate, MDTO) was evaluated. This substance, possessing important biological properties, is similar to carnosic acid, a natural antioxidant from rosemary or sage leaves. Aiming to understand the antioxidant activity of MDTO, the energetics of its O-H bond, using time-resolved photoacoustic calorimetry (TR-PAC), was investigated. On energetic grounds it is predicted that MDTO is a good free radical scavenger, although its activity is lower than that of quercetin, a very effective antioxidant, which was used for comparison. In agreement with these predictions, the DPPH(*) and ABTS(*+) radical scavenging activities are lower than those of quercetin. In addition, MDTO also reacts with HOCl, a powerful proinflammatory oxidant produced by activated neutrophils, and protects liposomes against iron-ascorbate-induced oxidation. The discussion of these results foresees potential applications of MDTO as an antioxidant.

Antioxidant activity of a catechol derived from abietic acid

New [60] and [70]fullerene–coumarin dyads were prepared by covalently linking a coumarin dye to a fullerene (C60 or C70)
				via a 1,3-dipolar cycloaddition reaction of azomethine ylides or by a cyclopropanation reaction of fullerenes with malonate derivatives. These dyads were spectroscopically characterized and their fluorescence properties studied. Fluorescence quenching of the coumarin fluorophore in the dyads results from efficient dipole–dipole resonance energy transfer from the coumarin moiety to the fullerene moiety.
