Conceptual density functional theory.

Efficient synthetic methods required to assemble complex molecular arrays include reactions that are both selective (chemo-, regio-, diastereo-, and enantio-) and economical in atom count (maximum number of atoms of reactants appearing in the products). Methods that involve simply combining two or more building blocks with any other reactant needed only catalytically constitute the highest degree of atom economy. Transition metal-catalyzed methods that are both selective and economical for formation of cyclic structures, of great interest for biological purposes, represent an important starting point for this long-term goal. The limited availability of raw materials, combined with environmental concerns, require the highlighting of these goals.

The atom economy--a search for synthetic efficiency

A new iron(III) halide-promoted aza-Prins cyclization between γ,δ-unsaturated tosylamines and aldehydes provides six-membered azacycles in good to excellent yields. The process is based on the consecutive generation of γ-unsaturated-iminium ion and further nucleophilic attack by the unsaturated carbon−carbon bond. Homoallyl tosylamine leads to trans-2-alkyl-4-halo-1-tosylpiperidine as the major isomer. In addition, the alkyne aza-Prins cyclization between homopropargyl tosylamine and aldehydes gives 2-alkyl-4-halo-1-tosyl-1,2,5,6-tetrahydropyridines as the only cyclic products. The piperidine ring is widely distributed throughout Nature, e.g., in alkaloids,1 and is an important scaffold for drug discovery, being the core of many pharmaceutically significant compounds.2,3 The syntheses of these type of compounds have been extensively studied in the development of new drugs containing six-membered-ring heterocycles.4 Reactions between N-acyliminium ions and nucleophiles, also described as amidoalkylation or Mannich-type condensations, have been frequently used to introduce substituents at the R-carbon of an amine.5 There are several examples that involve an intramolecular attack of a nucleophilic olefin into an iminium cation for the construction of a heterocyclic ring system.6 Traditionally, the use of hemiaminals or their derivatives as precursors of N-acyliminium intermediates has been a common two-step strategy in these reactions.6a Among this type of cyclization is the aza-Prins cyclization,7 which uses alkenes as intramolecular nucleophile. fumagillin. The biological activity and novel structure of fumagillin have stimulated the interest of several groups in synthesis. However, the complications of asymmetry (six centers) and highly reactive functionality have conspired to prevent solution of the p r ~ b l e m . ~ The goal of this synthesis was (*)-fumagillol (2), since this should be easily convertible to fumagillin. In fact, treatment of (-)-fumagillol, readily available by degradation of fumagilliq6 with 1 equiv of methyllithium, followed by addition of this solution to 5 equiv of decatetraenedioyl chloride in tetrahydrofuran at 78', gave fumagillin, identical with natural fumagillin by thin-layer chromatographic (tlc), infrared (ir), nuclear magnetic resonance (nmr), and mass spectral comparison. The demonstration of the first total synthesis of (&)fumagillol proceeded as follows. Alkylation of methyl acetoacetate with 3,3-dimethylallyl bromide' followed by deacetylation using the procedure of Ritter and KanieckP gave methyl 5-methyl-4-hexenoate (3),9 bp 47-50' (5 mm) (57 % yield). Oxidation of the olefin 3 with selenium dioxide in 97 % aqueous 1,2-dimethoxyethane for 8 hr at reflux gave stereoselectively in 41 %

Crambines A (7) (eight steps, 22%). B (45d) (eight steps, 19%), C1 (98) (seven steps, 21%), and C2 (9a) (seven steps, 27%) have been synthesized expediently and stereospecifically by a biomimetic route from methyl acetoacetate. Aminodihydropyrimidines 39 and 40 are formed efficiently from enone ester 36 by a two-step procedure involving addition of O-methylisourea to give methoxydihydropyrimidine 37 followed by displacement of the methoxy group of 37 with ammonia. Hydrogenolysis of 40a and 40d afford crambines C2 and C1, respectively. Mesylation of the alcohol of 39a or 40a followed by Et 3 N-catalyzed cyclization and hydrogenolysis affords crambine A (7)
