Abstract: The effect of exposure to sub-lethal concentrations of cypermethrin, a synthetic pyrethroid pesticide, on biochemical parameters of muscle, blood and enzyme activities in brain, liver and kidney of the Indian major carp, Labeo rohita was studied. The sub-lethal exposure studies were done for up to 45 days at 1/10 and 1/50 of 96 h LC(50) of cypermethrin. The 96 h LC(50) was found to be 0.139 ppm. RNA levels decreased while DNA levels were elevated. Acid phosphatase was unchanged while alkaline phosphatase was depleted. Brain acetylcholinesterase activity was decreased significantly (P<0.05) over a period of 45 days at both cypermethrin concentrations. Lactate dehydrogenase activity in brain and liver was elevated, but inhibited in kidney. Succinate dehydrogenase and ATPase activities were depleted in brain, kidney and liver. There was a decrease in serum protein level over control at both concentrations of the pyrethroid. Blood glucose level and total leucocytes were elevated compared with controls at either concentration from day 15 to day 45. Haemoglobin percentage and total erythrocytes decreased in both sub-lethal concentrations. Extracts of the herb Datura stramonium were effective in countering the toxicity of this pesticide. Our data suggest that sub-lethal exposure of cypermethrin alters the biochemical, haematological parameters and enzymes of organs tissue and exert stress on the fish. Plant extracts may be useful in counteracting some of these effects.

Abstract: Summary The aim of this study was to evaluate dietary dosages of garlic on the immune response and disease resistance against infections due to the opportunistic pathogen Aeromonas hydrophila in Labeo rohita fingerlings. Garlic, Allium sativum, was incorporated into the diets (0%, 0.1%, 0.5% and 1.0%) of rohu, L. rohita fingerlings (10 ± 2 g). Every 20 days, different biochemical (serum total protein, albumin, globulin, albumin : globulin ratio, blood glucose), haematological (WBC, RBC and haemoglobin content) and immunological (superoxide anion production, lysozyme activity and serum bactericidal activity) parameters were evaluated. Superoxide anion production, lysozyme, serum bactericidal, serum protein and albumin were enhanced in garlic treated groups compared with the control group. After 60 days, fish were challenged with A. hydrophila and mortality (%) was recorded up to day 10 post-challenge. Survival decreased in control group (57%) up to day 10 after infection. However, this was increased in the garlic treatment group, i.e. 85% survivability in the 1 g garlic kg−1 (B group) and 5 g garlic kg−1 (C group), and 71% survivability in the 10 g garlic kg−1 (D group), respectively. These results indicate that Allium sativum stimulates the immunity and makes L. rohita more resistant to infection by A. hydrophila.

Abstract: The innate immune system of fish is considered to be the first line of defence against a broad spectrum of pathogens and is more important for fish as compared with mammals. Lysozyme level or activity is an important index of innate immunity of fish and is ubiquitous in its distribution among living organisms. It is well documented that fish lysozyme possess lytic activity against both Gram-positive bacteria and Gram-negative bacteria. It is also known to be opsonic in nature and activates the complement system and phagocytes. It is present in mucus, lymphoid tissue, plasma and other body fluids of freshwater and marine fish. It is also expressed in a wide variety of tissues. Lysozyme activity has been shown to vary depending on the sex, age and size, season, water temperature, pH, toxicants, infections and degree of stressors. Here, we review our current understanding of different types of lysozyme and their expression and its role in fish innate immune system.
