A potent Cas9-derived gene activator for plant and mammalian cells
TL;DR: A new potent dCas9–TAD, named d Cas9–TV, is developed through plant cell-based screens that confers far stronger transcriptional activation of single or multiple target genes than the routinely used dCas 9–VP64 activator in both plant and mammalian cells.
Journal ArticleDOI
SARS-CoV-2 drives JAK1/2-dependent local complement hyperactivation.
Bingyu Yan,Tilo Freiwald,Tilo Freiwald,Daniel Chauss,Luopin Wang,Erin E. West,Carmen Mirabelli,Charles J. Zhang,Eva-Maria Nichols,Nazish Malik,Richard Gregory,Marcus Bantscheff,Sonja Ghidelli-Disse,Martin Kolev,Tristan Frum,Jason R. Spence,Jonathan Z. Sexton,Konstantinos D. Alysandratos,Konstantinos D. Wobus,Arian Laurence,Michail S. Lionakis,Claudia Kemper,Claudia Kemper,Behdad Afzali,Majid Kazemian +35 more
TL;DR: In this paper, the authors found that the complement system was one of the intracellular pathways most highly induced by SARS-CoV-2 infection in lung epithelial cells.
Journal ArticleDOI
Autocrine vitamin D signaling switches off pro-inflammatory programs of TH1 cells.
Daniel Chauss,Tilo Freiwald,Tilo Freiwald,Reuben McGregor,Reuben McGregor,Bingyu Yan,Luopin Wang,Estefania Nova-Lamperti,Dhaneshwar Kumar,Dhaneshwar Kumar,Zonghao Zhang,Heather L. Teague,Erin E. John,Paul Lavender,Claudia Kemper,Claudia Kemper,Giovanna Lombardi,Nehal N. Mehta,Nichola Cooper,Michail S. Lionakis,Arian Laurence,Majid Kazemian,Behdad Afzali +33 more
TL;DR: This paper showed that complement triggers contraction of helper T responses by inducing intrinsic expression of the vitamin D (VitD) receptor and the VitD-activating enzyme CYP27B1, permitting T cells to both activate and respond to VitD.
Journal ArticleDOI
Integrated pan-cancer map of EBV-associated neoplasms reveals functional host-virus interactions
Srishti Chakravorty,Bingyu Yan,Chong Wang,Luopin Wang,Joseph Taylor Quaid,Chin Fang Lin,Scott D. Briggs,Joydeb Majumder,D. Alejandro Canaria,Daniel Chauss,Gaurav Chopra,Matthew R. Olson,Bo Zhao,Behdad Afzali,Majid Kazemian +14 more
TL;DR: A comprehensive map of the host cell-pathogen interactome in EBV-associated cancers is presented, uncover novel points of interaction between a common oncovirus and the human genome and identify novel regulatory nodes and druggable targets for individualized EBV and cancer-specific therapies.
Journal ArticleDOI
Host-Virus Chimeric Events in SARS-CoV-2-Infected Cells Are Infrequent and Artifactual.
Bingyu Yan,Srishti Chakravorty,Carmen Mirabelli,Luopin Wang,Jorge L Trujillo-Ochoa,Daniel Chauss,Dhaneshwar Kumar,Dhaneshwar Kumar,Michail S. Lionakis,Matthew R. Olson,Christiane E. Wobus,Behdad Afzali,Majid Kazemian +12 more
TL;DR: The findings indicate that HVC events observed in RNA-sequencing libraries from SARS-CoV2 infected cells are extremely rare and are likely artifacts arising from either random template switching of reverse-transcriptase and/or sequence alignment errors.
