Publisher Summary This chapter describes chemical shift standards and pointing out some of the necessary corrections and experimental recommendations that need to be followed so that the NMR community can exploit chemical shift information to the fullest. Chemical shift analysis holds much promise for the field of protein NMR. There are two very different chemical shift standards are being used has led to the appearance of several very different tables for random coil 13 C chemical shifts, which has already caused a good deal of confusion within the NMR community. In particular case, chemical shift analysis was applied to a small, flexible peptide to both localize and quantify helix content as a function of TFE concentration. By monitoring the TFE-induced displacement of assigned a- 13 C resonances, the extent of the relative chemical shift change could be correlated with the intensity of the helical CD signal. These chemical shift plots are reported to provide qualitative details on the flexibility of the molecule and the presence of possible nonnative conformers.

Chemical shifts as a tool for structure determination.

Dissected Nucleus-Independent Chemical Shift Analysis of π-Aromaticity and Antiaromaticity.

Accepting a commission to review progress in the subject embodied in our title is, perhaps, to take up something of a ‘poisoned chalice’. One of us, contributing on this same topic more than 20 years ago at the 1979 International Symposium on Aromaticity in Dubrovnik, wrote1 “A cynic would say that there are actually only two difficulties in discussing the subject of ‘aromaticity’ and ‘ring currents’sdeciding what is meant by ‘ring current’, and assigning a meaning to the term ‘aromaticity’!” That comment, though ostensibly facetious, had serious intent: it did encapsulate, with only a modicum of exaggeration, the problems that inherently beset any assessment such as the one attempted at Dubrovnik1 and in the present review. At the heart of the matter lies the undeniable fact that neither ring currents nor aromaticity are physical observables. Nevertheless, the intervening period has seen the ring-current idea, at least, become generally less controversial and more accepted than it once was. At the time of our opening quotation, one of us and Haigh had just published an exhaustive review2 of the ring-current concept covering the period up to about 1980sthe end of what might now be regarded as the era of semiempirical calculations in this field.2 This review2 (1979/1980) concluded that “the ‘ring current’ picture has proved itself ... to have great power in rationalising, at least qualitatively, the magnetic properties of π-electron systems. It is so pictorial that one can almost feel what is happening when a [conjugated] molecule is subjected to a magnetic field. Whatever advances the future may bring, it may be that the favourite habitat of the ‘ring current’ will be that in which it was born and brought up, namely, that of semi-empirical π-electron theory”. In other words, these authors were sanguine that, at the time (ca. 1980), the ring-current idea was gently coming to the end of its natural, useful life. However, as recently as 1997, when reviewing progress concerning the status of the ring-current model during the decade and a half or so after 1980s a period in this field that we have dubbed3 ‘the ab initio era’sthe present authors3 were initially somewhat surprised to find themselves concluding that “the ‘ring-current’ idea has well survived the first 15 years of the ab initio era.” Lazzeretti’s subsequent magnum opus4 on ring currents has more than confirmed this. By contrast, the sheer fact that in 2001sthe very first year of the 21st centurysthe American Chemical Society has seen fit to run this particular issue of Chemical Reviews shows that the concept of Aromaticity is as elusive as it ever was (see section II). There are three reasons for our not feeling obliged or inclined to present, in this review, an exhaustive, systematic, or historical critique of the ring-current concept itself. First, as we have just claimed, the idea of a ring current seems more secure now than it was 20 years ago, and it would appear that less apology or justification is needed for invoking it. Aromaticity and ring currents.

The solution structure of a synthetic 36-residue polypeptide comprising the C-terminal cellulose binding domain of cellobiohydrolase I (CT-CBH I) from Trichoderma reesei was investigated by nuclear magnetic resonance (NMR) spectroscopy. The 1H NMR spectrum was completely assigned in a sequential manner by two-dimensional NMR techniques. A large number of stereospecific assignments for beta-methylene protons, as well as ranges for the phi, psi, and chi 1 torsion angles, were obtained on the basis of sequential and intraresidue nuclear Overhauser enhancement (NOE) and coupling constant data in combination with a conformational data base search. The structure calculations were carried out in an iterative manner by using the hybrid distance geometry-dynamical simulated annealing method. This involved computing a series of initial structures from a subset of the experimental data in order to resolve ambiguities in the assignments of some NOE cross-peaks arising from chemical shift degeneracy. Additionally, this permitted us to extend the stereospecific assignments to the alpha-methylene protons of glycine using information on phi torsion angles derived from the initial structure calculations. The final experimental data set consisted of 554 interproton distance restraints, 24 restraints for 12 hydrogen bonds, and 33 phi, 24 psi, and 25 chi 1 torsion angle restraints. CT-CBH I has two disulfide bridges whose pairing was previously unknown. Analysis of structures calculated with all three possible combinations of disulfide bonds, as well as without disulfide bonds, indicated that the correct disulfide bridge pairing was 8-25 and 19-35. Forty-one structures were computed with the 8-25 and 19-35 disulfide bridges, and the average atomic rms difference between the individual structures and the mean structure obtained by averaging their coordinates was 0.33 +/- 0.04 A for the backbone atoms and 0.52 +/- 0.06 A for all atoms. The protein has a wedgelike shape with an amphiphilic character, one face being predominantly hydrophilic and the other mainly hydrophobic. The principal element of secondary structure is made up of an irregular triple-stranded antiparallel beta-sheet composed of residues 5-9 (beta 1), 24-28 (beta 2), and 33-36 (beta 3) in which strand beta 3 is hydrogen bonded to the other two strands.(ABSTRACT TRUNCATED AT 400 WORDS)

Determination of the three-dimensional solution structure of the C-terminal domain of cellobiohydrolase I from Trichoderma reesei. A study using nuclear magnetic resonance and hybrid distance geometry-dynamical simulated annealing.

The free electron model of Pauling is used to calculate the magnetic field around a benzene ring which is rotating rapidly about all axes in an external magnetic field. It is assumed that the π electrons precess in two circular paths, one on each side of the ring, equal in radius to the C–C distance in the benzene ring. The separation of these rings is taken as 1.28 A, which gives a calculated value for the nuclear magnetic resonance shielding value for benzene protons equal to the observed value. The field thus calculated is employed to predict shielding values for other aromatic compounds. Agreement with experiment is in general good, but there are some exceptions.
