Journal ArticleDOI
Causes and prevalence of visual impairment among adults in the United States.
Nathan Congdon,O'Colmain Bj,Caroline C W Klaver,Ronald Klein,Beatriz Munoz,David S. Friedman,John H. Kempen,Hugh R. Taylor,Paul Mitchell +8 more
TL;DR: The prevalence of visual disabilities will increase markedly during the next 20 years, owing largely to the aging of the US population.
Journal ArticleDOI
A common haplotype in the complement regulatory gene factor H (HF1/CFH) predisposes individuals to age-related macular degeneration
Gregory S. Hageman,Don H. Anderson,Lincoln V. Johnson,Lisa S. Hancox,Andrew J Taiber,Lisa I. Hardisty,Jill L. Hageman,Heather Stockman,James D. Borchardt,Karen M. Gehrs,Richard J.H. Theodore Smith,Adam Olsh,Julie Bergeron,Jana Zernant,Joanna E. Merriam,Bert Gold,Michael Dean,Rando Allikmets +26 more
TL;DR: It is shown that factor H (HF1), the major inhibitor of the alternative complement pathway, accumulates within drusen and is synthesized by the retinal pigmented epithelium, implicating HF1 function in the pathogenetic mechanisms underlying AMD.
Journal ArticleDOI
An international classification and grading system for age-related maculopathy and age-related macular degeneration
Alan C. Bird,Neil M. Bressler,Susan B. Bressler,I H Chisholm,Gabriel Coscas,M.D. Davis,P.T.V.M. de Jong,Caroline C W Klaver,Barbara E.K. Klein,Ronald Klein,Philip B. Mitchell,J.P. Sarks,S.H. Sarks,Gisèle Soubrane,Hugh R. Taylor,Johannes R. Vingerling +15 more
TL;DR: A common detection and classification system is needed for epidemiologic studies of age-related maculopathy (ARM) and such a grading scheme for ARM is described in this paper.
Journal ArticleDOI
The Rotterdam Study: Objectives and design update
Albert Hofman,Guy Brusselle,Sarwa Darwish Murad,Cornelia M. van Duijn,Oscar H. Franco,André Goedegebure,M. Arfan Ikram,Caroline C W Klaver,Tamar Nijsten,Robin P. Peeters,Bruno H. Ch. Stricker,Henning Tiemeier,André G. Uitterlinden,Meike W. Vernooij +13 more
TL;DR: The rationale of the study and its design is given, and a summary of the major findings and an update of the objectives and methods are presented.
Journal ArticleDOI

TL;DR: The results support the hypothesis that rare coding variants can pinpoint causal genes within known genetic loci and illustrate that applying the approach systematically to detect new loci requires extremely large sample sizes.
