Abstract: The force-generating mechanism of dynein differs from the force-generating mechanisms of other cytoskeletal motors. To examine the structural dynamics of dynein's stepping mechanism in real time, we used polarized total internal reflection fluorescence microscopy with nanometer accuracy localization to track the orientation and position of single motors. By measuring the polarized emission of individual quantum nanorods coupled to the dynein ring, we determined the angular position of the ring and found that it rotates relative to the microtubule (MT) while walking. Surprisingly, the observed rotations were small, averaging only 8.3°, and were only weakly correlated with steps. Measurements at two independent labeling positions on opposite sides of the ring showed similar small rotations. Our results are inconsistent with a classic power-stroke mechanism, and instead support a flexible stalk model in which interhead strain rotates the rings through bending and hinging of the stalk. Mechanical compliances of the stalk and hinge determined based on a 3.3-μs molecular dynamics simulation account for the degree of ring rotation observed experimentally. Together, these observations demonstrate that the stepping mechanism of dynein is fundamentally different from the stepping mechanisms of other well-studied MT motors, because it is characterized by constant small-scale fluctuations of a large but flexible structure fully consistent with the variable stepping pattern observed as dynein moves along the MT.

The active channel layer includes pentacene formed by thermal treatment of its precursors and operates as a hole carrier. Within the pentacene film are nanoparticles or nanowires of a second material that operate as electron carriers. The electron carrier materials are selected from a group of soluble semiconducting inorganic nanocrystals and nanowires or solube derivatives of fullerene.

Abstract: X-ray atomic pair distribution functions (PDFs) were collected from a range of canonical metallic nanomaterials, both elemental and alloyed, prepared using different synthesis methods and exhibiting drastically different morphological properties. Widely applied shape-tuned attenuated crystal (AC) fcc models proved inadequate, yielding structured, coherent, and correlated fit residuals. However, equally simple discrete cluster models could account for the largest amplitude features in these difference signals. A hypothesis testing based approach to nanoparticle structure modeling systematically ruled out effects from crystallite size, composition, shape, and surface faceting as primary factors contributing to the AC misfit. On the other hand, decahedrally twinned cluster cores were found to be the origin of the AC structure misfits for a majority of the nanomaterials reported here. Abstract: Nanorod diffusion in polymer melts is faster than predicted by the continuum model (CM). Rutherford backscattering spectrometry is used to measure the concentration profile of titanium dioxide (TiO2) nanorods (L = 43 nm, d = 5 nm) in a polystyrene (PS) matrix having molecular weights (M) from 9 to 2000 kDa. In the entangled regime, the tracer diffusion coefficients (D) of TiO2 decrease as the M–1.4, whereas the CM predicts DCM ∼ M–3.0 using the measured zero-shear viscosity of TiO2(1 vol %): PS(M) blends. By plotting D/DCM versus M/Me, where Me is the entanglement molecular weight, diffusion is enhanced by a factor of 10–103 as M/Me increases. The faster diffusion is attributed to decoupling of nanorod diffusion from polymer relaxations in the surrounding matrix, which is facilitated by the nanorod dimensions (i.e., L greater than and d less than the entanglement mesh size, 8 nm).

Abstract: Current research into semiconductor clusters is focused on the properties of quantum dots-fragments of semiconductor consisting of hundreds to many thousands of atoms-with the bulk bonding geometry and with surface states eliminated by enclosure in a material that has a larger band gap. Quantum dots exhibit strongly size-dependent optical and electrical properties. The ability to join the dots into complex assemblies creates many opportunities for scientific discovery.

Abstract: Highly luminescent semiconductor quantum dots (zinc sulfide-capped cadmium selenide) have been covalently coupled to biomolecules for use in ultrasensitive biological detection. In comparison with organic dyes such as rhodamine, this class of luminescent labels is 20 times as bright, 100 times as stable against photobleaching, and one-third as wide in spectral linewidth. These nanometer-sized conjugates are water-soluble and biocompatible. Quantum dots that were labeled with the protein transferrin underwent receptor-mediated endocytosis in cultured HeLa cells, and those dots that were labeled with immunomolecules recognized specific antibodies or antigens.
