Journal ArticleDOI
Antitumor imidazotetrazines. 1. Synthesis and chemistry of 8-carbamoyl-3-(2-chloroethyl)imidazo[5,1-d]-1,2,3,5-tetrazin-4(3 H)-one , a novel broad-spectrum antitumor agent.
Malcolm F. G. Stevens,John A. Hickman,Robert Stone,Neil W. Gibson,Ghouse Unissa Baig,Edward Lunt,Christopher G. Newton +6 more
TL;DR: Compound 7a has curative activity against L-1210 and P388 leukemia and may act as a prodrug modification of the acyclic triazene 5-[3-(2-chloroethyl)triazen-1-yl]imidazole-4-carboxamide (MCTIC), since it ring opens to form the triazenes in aqueous sodium carbonate.
Journal ArticleDOI
Antitumor imidazotetrazines. 14. Synthesis and antitumor activity of 6- and 8-substituted imidazo[5,1-d]-1,2,3,5-tetrazinones and 8-substituted pyrazolo[5,1-d]-1,2,3,5-tetrazinones
Edward Lunt,Christopher G. Newton,Christopher Smith,G. P. Stevens,Malcolm F. G. Stevens,C. G. Straw,Walsh Roger John Aitchison,P. J. Warren,C. Fizames,François Lavelle +9 more
TL;DR: From the limited number of pyrazolotetrazinones reported here, it is suggested that the same conclusions as regards activity also hold true for this ring system.
Journal ArticleDOI
Hypolipidemic 2-[4-(1,1-dimethylethyl)phenyl]-4H-3,1-benzoxazin-4-ones. Structure-activity relationships of a novel series of high-density lipoprotein elevators.
Garry Fenton,Christopher G. Newton,Barry M. Wyman,Philip Bagge,Dron Donald Ian,David Riddell,Graham Jones +6 more
TL;DR: Evidence is presented suggesting that a metabolite or degradation product is responsible for the changes in lipoprotein concentration observed with active molecules of this type.
Journal ArticleDOI
Acyl-CoA:cholesterol O-acyl transferase (ACAT) inhibitors. 1. 2-(Alkylthio)-4,5-diphenyl-1H-imidazoles as potent inhibitors of ACAT.
Neil Victor Harris,Christopher Smith,Ashton Michael John,Bridge Andrew William,R.C. Bush,E. C. J. Coffee,Dron Donald Ian,M. F. Harper,David J. Lythgoe,Christopher G. Newton +9 more
TL;DR: Structural-activity relationships for analogues of 2 demonstrate that the 4,5-diphenyl-1H-imidazole moiety is a pharmacophore for inhibition of rat microsomal ACAT in vitro and produces a significant hypocholesterolemic effect in the cholesterol-fed rat.
