DPPH antioxidant assay revisited

Antioxidant properties have been attributed to the essential oils but a great diversity of results has been reported, mainly attributed to the chemical composition of the essential oils and the diversity of tests used. In fact, several in vitro assays for the determination of antioxidant ability have been used. They can be divided into two main groups: those that evaluate lipid peroxidation and that those that measure free radical scavenging ability. In the present study, a brief description of the mechanism involved in the majority of techniques used for the evaluation of antioxidant activity of essential oils is given. At the same time, the antioxidant activities of some essential oils, measured through several methodologies, are presented and compared. Copyright © 2009 John Wiley & Sons, Ltd.

Antioxidant activity of medicinal and aromatic plants. A review.

This study evaluated, using in vitro assays, the antibacterial, antioxidant, and tyrosinase-inhibition activities of methanolic extracts from peels of seven commercially grown pomegranate cultivars. Antibacterial activity was tested on Gram-positive (Bacillus subtilis and Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli and Klebsiella pneumonia) using a microdilution method. Several potential antioxidant activities, including radical-scavenging ability (RSA), ferrous ion chelating (FIC) and ferric ion reducing antioxidant power (FRAP), were evaluated. Tyrosinase enzyme inhibition was investigated against monophenolase (tyrosine) and diphenolase (DOPA), with arbutin and kojic acid as positive controls. Furthermore, phenolic contents including total flavonoid content (TFC), gallotannin content (GTC) and total anthocyanin content (TAC) were determined using colourimetric methods. HPLC-ESI/MSn analysis of phenolic composition of methanolic extracts was also performed. Methanolic peel extracts showed strong broad-spectrum activity against Gram-positive and Gram-negative bacteria, with the minimum inhibitory concentrations (MIC) ranging from 0.2 to 0.78 mg/ml. At the highest concentration tested (1000 μg/ml), radical scavenging activities were significantly higher in Arakta (83.54%), Ganesh (83.56%), and Ruby (83.34%) cultivars (P 50%) against monophenolase and diphenolase activities at the highest screening concentration. The most active peel extract was the Bhagwa cultivar against monophenolase and the Arakta cultivar against diphenolase with IC50 values of 3.66 μg/ml and 15.88 μg/ml, respectively. High amounts of phenolic compounds were found in peel extracts with the highest and lowest total phenolic contents of 295.5 (Ganesh) and 179.3 mg/g dry extract (Molla de Elche), respectively. Catechin, epicatechin, ellagic acid and gallic acid were found in all cultivars, of which ellagic acid was the most abundant comprising of more than 50% of total phenolic compounds detected in each cultivar. The present study showed that the tested pomegranate peels exhibited strong antibacterial, antioxidant and tyrosinase-inhibition activities. These results suggest that pomegranate fruit peel could be exploited as a potential source of natural antimicrobial and antioxidant agents as well as tyrosinase inhibitors.

Antibacterial, antioxidant and tyrosinase-inhibition activities of pomegranate fruit peel methanolic extract

Cancer is the second leading cause of death in the world. Chemotherapy and radiotherapy are the common cancer treatments. However, the development of adverse effects resulting from chemotherapy and radiotherapy hinders the clinical use, and negatively reduces the quality of life in cancer patients. Natural products including crude extracts, bioactive components-enriched fractions and pure compounds prepared from herbs as well as herbal formulas have been proved to prevent and treat cancer. Of significant interest, some natural products can reduce chemotherapy and radiotherapy-induced oral mucositis, gastrointestinal toxicity, hepatotoxicity, nephrotoxicity, hematopoietic system injury, cardiotoxicity, and neurotoxicity. This review focuses in detail on the effectiveness of these natural products, and describes the possible mechanisms of the actions in reducing chemotherapy and radiotherapy-induced side effects. Recent advances in the efficacy of natural dietary supplements to counteract these side effects are highlighted. In addition, we draw particular attention to gut microbiotan in the context of prebiotic potential of natural products for the protection against cancer therapy-induced toxicities. We conclude that some natural products are potential therapeutic perspective for the prevention and treatment of chemotherapy and radiotherapy-induced side effects. Further studies are required to validate the efficacy of natural products in cancer patients, and elucidate potential underlying mechanisms.

Natural Product Interventions for Chemotherapy and Radiotherapy-Induced Side Effects.

The present review demonstrates that EOs and their components have many functional properties and exert their action in mammals as well as in other organisms (insects, fungi, bacteria and viruses). The synergistic effect of EO components is a promising field that could lead to the optimisation of a given bioactivity. This phenomenon has been observed in many activities, such as those of antimicrobials, antioxidants, analgesics and semiochemicals. EOs are complex mixtures of components that show higher activities than their isolated components; their final activities are due to the combine effects of several minor components. Thus, EOs contain multifunctional components that exert their activities through different mechanisms. EOs and their components may have new applications against various diseases of different origins (cancer, fungal, bacterial or viral), because some of these complex diseases require multiple components and multifunctional therapies.

Bioactivity of Essential Oils and Their Components

The chemical composition of the essential oils obtained from the leaves, the barks of the stem and the root, as well as from the fresh and dried fruits of Xylopia aethiopica, growing in Ghana, was investigated by gas chromatography/mass spectrometry analyses. Kovats indices, mass spectra, and standard compounds were used to identify a total of 93 individual compounds. The monoterpene hydrocarbons formed the main portion in all studied samples. β-Pinene was predominant in all cases, while trans-m-mentha-1(7),8-diene was the main compound in the essential oils of the leaves and the barks of roots and stems. Their potential antioxidant activity was also investigated and found to be significant in scavenging superoxide anion radical.
