Journal ArticleDOI
A Dynamic Protein Interaction Landscape of the Human Centrosome-Cilium Interface.
Gagan D. Gupta,Etienne Coyaud,João Gonçalves,Bahareh A. Mojarad,Yi Liu,Yi Liu,Qianzhu Wu,Qianzhu Wu,Ladan Gheiratmand,David Comartin,David Comartin,Johnny M. Tkach,Sally W.T. Cheung,Mikhail Bashkurov,Monica Hasegan,James D.R. Knight,Zhen-Yuan Lin,Markus Schueler,Friedhelm Hildebrandt,Friedhelm Hildebrandt,Jason Moffat,Anne-Claude Gingras,Anne-Claude Gingras,Brian Raught,Brian Raught,Laurence Pelletier,Laurence Pelletier +26 more
TL;DR: Systematic profiling of proximity interactions combined with functional analysis provides a rich resource for better understanding human centrosome and cilia biology and may be applied to other complex biological structures or pathways.
Journal ArticleDOI
Inhibition of the Mitochondrial Protease ClpP as a Therapeutic Strategy for Human Acute Myeloid Leukemia
Alicia Cole,Zezhou Wang,Etienne Coyaud,Veronique Voisin,Marcela Gronda,Yulia Jitkova,Rachel Mattson,Rose Hurren,Sonja Babovic,Neil MacLean,Ian Restall,Xiaoming Wang,Danny V. Jeyaraju,Mahadeo A. Sukhai,Swayam Prabha,Shaheena Bashir,Ashwin Ramakrishnan,Elisa Leung,Yi Hua Qia,Nianxian Zhang,Kevin R. Combes,Troy Ketela,Fengshu Lin,Walid A. Minden,Connie J. Eaves,Gary D. Bader,Zhenyue Hao,Steven M. Kornblau,Brian Raught,Aaron D. Schimmer,Aaron D. Schimmer +44 more
TL;DR: Mechanistically, it is found that ClpP interacts with mitochondrial respiratory chain proteins and metabolic enzymes, and knockdown of ClPP in leukemic cells inhibited oxidative phosphorylation and mitochondrial metabolism.
Journal ArticleDOI
VAPs and ACBD5 tether peroxisomes to the ER for peroxisome maintenance and lipid homeostasis.
Rong Hua,Derrick Cheng,Etienne Coyaud,Spencer A. Freeman,Erminia Di Pietro,Yuqing Wang,Adriano Vissa,Christopher M. Yip,Gregory D. Fairn,Nancy Braverman,John H. Brumell,William S. Trimble,Brian Raught,Brian Raught,Peter K. Kim +14 more
TL;DR: This study reports that the ER-resident VAMP-associated proteins A and B interact with the peroxisomal membrane protein acyl-CoA binding domain containing 5 (ACBD5) and that this interaction is required to tether the two organelles together, thereby facilitating the lipid exchange between them.
Journal ArticleDOI
A proximity-dependent biotinylation map of a human cell.
Christopher D. Go,Christopher D. Go,James D.R. Knight,Archita Rajasekharan,Bhavisha Rathod,Geoffrey G. Hesketh,Kento T. Abe,Kento T. Abe,Ji-Young Youn,Ji-Young Youn,Payman Samavarchi-Tehrani,Hui Zhang,Lucie Y. Zhu,Evelyn Popiel,Jean-Philippe Lambert,Jean-Philippe Lambert,Etienne Coyaud,Etienne Coyaud,Sally W.T. Cheung,Dushyandi Rajendran,Cassandra J. Wong,Hana Antonicka,Laurence Pelletier,Laurence Pelletier,Alexander F. Palazzo,Eric A. Shoubridge,Brian Raught,Brian Raught,Anne-Claude Gingras,Anne-Claude Gingras +29 more
TL;DR: In this article, a proximity-dependent biotinylation technique was used to define the location of more than 4,000 proteins in a human cell and almost 36,000 proximal interactions between proteins, including those at the interface of the mitochondria and ER.
Journal ArticleDOI
BioID-based Identification of Skp Cullin F-box (SCF)β-TrCP1/2 E3 Ligase Substrates.
Etienne Coyaud,Monika Mis,Estelle M.N. Laurent,Wade H. Dunham,Amber L. Couzens,Mélanie Robitaille,Anne-Claude Gingras,Stephane Angers,Brian Raught,Brian Raught +9 more
TL;DR: The technique presented here represents a complementary approach to more standard IP-MS methods and should be broadly applicable for the identification of substrates for many ubiquitin E3 ligases, and suggests that β-TrCP is an important hub in the cellular stress response.
