Abstract: Sixty-three amide alkaloids, including three new, piperflaviflorine A (1), piperflaviflorine B (2), and sarmentamide D (4), and two previously synthesized ones, (1E,3S)-1-cinnamoyl-3- hydroxypyrrolidine (3) and N-[7′-(4′-methoxyphenyl)ethyl]-2-methoxybenzamide (5), were isolated from the aerial parts of Piper flaviflorum and Piper sarmentosum. Their structures were elucidated by detailed spectroscopic analysis and, in case of 3, by single-crystal X-ray diffraction. Most of the isolates were tested for their antifungal and antibacterial activities. Ten amides (6–15) showed antifungal activity against Cryptococcus neoformans ATCC 90 113 with IC50 values in the range between 4.7 and 20.0 µg/mL.

Abstract: “Long-Dan” and “Qin-Jiao” are two important TCM herbs since ancient times in China. In the Chinese Pharmacopoeia, the dried roots and rhizomes of four species from the genus Gentiana, e.g. Gentiana manshurica, G. scabra, G. triflora and G. rigescens, are recorded under the name of Gentianae Radix et Rhizoma (“Long-Dan” in Chinese), while the other four species from the same genus including G. macrophylla, G. crassicaulis, G. straminea and G. duhurica are recorded and used as the raw materials of Gentianae Macrophyllae Radix (“Qin-Jiao” in Chinese). On the basis of the establishment of a validated HPLC–UV method for quantifying simultaneously, five iridoid glycosides, e.g. loganic acid (1), swertiamarinin (2), gentiopicroside (3), sweroside (4) and 2′-(o,m-dihydroxybenzyl)sweroside (5) have been used successfully as chemical markers for the comparison of the species used as “Long-Dan”, “Qin-Jiao” and their adulterants in the present study. The results suggested that four iridoid glycosides 1–4 commonly existed in both “Long-Dan” and “Qin-Jiao”, while 2′-(o,m-dihydroxybenzyl)sweroside (5) also existed as one of the major components in “Dian-Long-Dan” species. Moreover, the contents of compounds 1–5 were various in different “Long-Dan” and “Qin-Jiao” species. Herein, we profiled and compared three “Long-Dan” species, four “Qin-Jiao” species and five adulterants by applying multivariate statistical techniques to their HPLC data sets to establish the differences and/or similarities. Abstract: The genus Piper is one of the largest genera in the Piperaceae, with most species widely distributed globally, covering all continents. To date, many Piper species have been scientifically investigated for their chemical diversities and interesting broad spectrum of bioactivities, including central nervous system (CNS), pesticidal, antifungal and antibacterial effects. This review systematically summarizes the scaffolds of the alkaloids reported, the major chemicals isolated from Piper spp., and their biological activities. Besides the alkaloids, some neolignans with rearranged skeletons show structural diversities, while the chalcones, flavonoids and kava-pyrones have some potential activities. Herein, the sesquiterpenes and phenolic compounds from Piper species and their bioactivities are also surveyed.

Abstract: Piper species are aromatic plants used as spices in the kitchen, but their secondary metabolites have also shown biological effects on human health. These plants are rich in essential oils, which can be found in their fruits, seeds, leaves, branches, roots and stems. Some Piper species have simple chemical profiles, while others, such as Piper nigrum, Piper betle, and Piper auritum, contain very diverse suites of secondary metabolites. In traditional medicine, Piper species have been used worldwide to treat several diseases such as urological problems, skin, liver and stomach ailments, for wound healing, and as antipyretic and anti-inflammatory agents. In addition, Piper species could be used as natural antioxidants and antimicrobial agents in food preservation. The phytochemicals and essential oils of Piper species have shown strong antioxidant activity, in comparison with synthetic antioxidants, and demonstrated antibacterial and antifungal activities against human pathogens. Moreover, Piper species possess therapeutic and preventive potential against several chronic disorders. Among the functional properties of Piper plants/extracts/active components the antiproliferative, anti-inflammatory, and neuropharmacological activities of the extracts and extract-derived bioactive constituents are thought to be key effects for the protection against chronic conditions, based on preclinical in vitro and in vivo studies, besides clinical studies. Habitats and cultivation of Piper species are also covered in this review. In this current work, available literature of chemical constituents of the essential oils Piper plants, their use in traditional medicine, their applications as a food preservative, their antiparasitic activities and other important biological activities are reviewed.

Abstract: The genus Gentiana comprises approximately 400 species. Many species have a wide range of pharmacological activities and have been used therapeutically for thousands of years. To provide comprehensive guidance, utilization and quality control of Gentiana species, this review presents updated information concerning the recent application and progress of chemical analysis including phytochemical analysis, sample preparation and chemometrics. Detailed and comprehensive data including number of analytes, extraction/separation methods, analytical techniques and chemometrics are shown as corresponding tables. These data illustrate that the development of newly discovered compounds and therapeutic uses, understanding of the structure—activity relationship and establishment of harmonious and effective medicinal herb standards are the direction of advancement in future research.

Abstract: Gentiana, which is one of the largest genera of Gentianoideae, most of which had potential pharmaceutical value, and applied to local traditional medical treatment. Because of the phytochemical diversity and difference of bioactive compounds among species, which makes it crucial to accurately identify authentic Gentiana species. In this paper, the feasibility of using the infrared spectroscopy technique combined with chemometrics analysis to identify Gentiana and its related species was studied. A total of 180 batches of raw spectral fingerprints were obtained from 18 species of Gentiana and Tripterospermum by near-infrared (NIR: 10,000-4000 cm-1) and Fourier transform mid-infrared (MIR: 4000-600 cm-1) spectrum. Firstly, principal component analysis (PCA) was utilized to explore the natural grouping of the 180 samples. Secondly, random forests (RF), support vector machine (SVM), and K-nearest neighbors (KNN) models were built while using full spectra (including 1487 NIR variables and 1214 FT-MIR variables, respectively). The MIR-SVM model had a higher classification accuracy rate than the other models that were based on the results of the calibration sets and prediction sets. The five feature selection strategies, VIP (variable importance in the projection), Boruta, GARF (genetic algorithm combined with random forest), GASVM (genetic algorithm combined with support vector machine), and Venn diagram calculation, were used to reduce the dimensions of the data variable in order to further reduce numbers of variables for modeling. Finally, 101 NIR and 73 FT-MIR bands were selected as the feature variables, respectively. Thirdly, stacking models were built based on the optimal spectral dataset. Most of the stacking models performed better than the full spectra-based models. RF and SVM (as base learners), combined with the SVM meta-classifier, was the optimal stacked generalization strategy. For the SG-Ven-MIR-SVM model, the accuracy (ACC) of the calibration set and validation set were both 100%. Sensitivity (SE), specificity (SP), efficiency (EFF), Matthews correlation coefficient (MCC), and Cohen's kappa coefficient (K) were all 1, which showed that the model had the optimal authenticity identification performance. Those parameters indicated that stacked generalization combined with feature selection is probably an important technique for improving the classification model predictive accuracy and avoid overfitting. The study result can provide a valuable reference for the safety and effectiveness of the clinical application of medicinal Gentiana.
