TL;DR: The accumulation characteristics and potential risk posed by polycyclic aromatic hydrocarbons (PAHs) in soils and vegetables grown in the home garden and agricultural field were investigated and indicated that the carcinogenic risk for the soil and vegetables contaminated with PAHs in the authors' study area for the residents was negligible.

The invention comprehends testing putative treatments with such models, e.g., testing putative chemical compounds that may be pharmaceutically relevant for treatment or gene therapy that may be relevant for treatment, or combinations thereof. The invention allows for the study of genetic diseases and putative treatments to better understand and alleviate a genetic disease or a condition, e.g., autism, autism-spectrum disease or disorder, obsessive compulsive disorder, or psychiatric disorders.

Abstract: Background Recent study demonstrates the comprehensive effects of gut microbiota on complex diseases or traits. However, limited effort has been conducted to explore the potential relationships between gut microbiota and BMD. Methods We performed a polygenetic risk scoring (PRS) analysis to systematically explore the relationships between gut microbiota and body BMD. Significant SNP sets associated with gut microbiota were derived from previous genome-wide association study (GWAS). In total, 2,294 to 5,065 individuals with BMD values of different sites and their genotype data were obtained from UK Biobank cohort. The gut microbiota PRS of each individual was computed from the SNP genotype data for each study subject of UK Biobank by PLINK software. Using computed PRS as the instrumental variables of gut microbiota, Pearson correlation analysis of individual PRS values and BMD values was finally conducted to test the potential association between gut microbiota and target trait. Results In total, 31 BMD traits were selected as outcome to assess their relationships with gut microbiota. After adjusted for age, sex, body mass index, and the first 5 principal components (PCs) as the covariates using linear regression model, pelvis BMD (P = 0.0437) showed suggestive association signal with gut microbiota after multiple testing correction. Conclusion Our study findings support the weak relevance of gut microbiota with the development of BMD.

Abstract: Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease with strong genetic components. To identity novel risk variants for ALS, utilizing the latest genome-wide association studies (GWAS) and eQTL study data, we conducted a genome-wide expression association analysis by summary data-based Mendelian randomization (SMR) method. Summary data were derived from a large-scale GWAS of ALS, involving 12577 cases and 23475 controls. The eQTL annotation dataset included 923,021 cis-eQTL for 14,329 genes and 4732 trans-eQTL for 2612 genes. Genome-wide single gene expression association analysis was conducted by SMR software. To identify ALS-associated biological pathways, the SMR analysis results were further subjected to gene set enrichment analysis (GSEA). SMR single gene analysis identified one significant and four suggestive genes associated with ALS, including C9ORF72 (P value = 7.08 × 10−6), NT5C3L (P value = 1.33 × 10−5), GGNBP2 (P value = 1.81 × 10−5), ZNHIT3(P value = 2.94 × 10−5), and KIAA1600(P value = 9.97 × 10−5). GSEA identified 7 significant biological pathways, such as PEROXISOME (empirical P value = 0.006), GLYCOLYSIS_GLUCONEOGENESIS (empirical P value = 0.043), and ARACHIDONIC_ACID_ METABOLISM (empirical P value = 0.040). Our study provides novel clues for the genetic mechanism studies of ALS.

Abstract: Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated (Cas) systems provide bacteria and archaea with adaptive immunity against viruses and plasmids by using CRISPR RNAs (crRNAs) to guide the silencing of invading nucleic acids. We show here that in a subset of these systems, the mature crRNA that is base-paired to trans-activating crRNA (tracrRNA) forms a two-RNA structure that directs the CRISPR-associated protein Cas9 to introduce double-stranded (ds) breaks in target DNA. At sites complementary to the crRNA-guide sequence, the Cas9 HNH nuclease domain cleaves the complementary strand, whereas the Cas9 RuvC-like domain cleaves the noncomplementary strand. The dual-tracrRNA:crRNA, when engineered as a single RNA chimera, also directs sequence-specific Cas9 dsDNA cleavage. Our study reveals a family of endonucleases that use dual-RNAs for site-specific DNA cleavage and highlights the potential to exploit the system for RNA-programmable genome editing.
