Abstract: Inflammatory bowel disease (IBD) is a complex disease, resulting from abnormal immune response to intestinal tract microbiota in genetically susceptible individuals. Crohn's disease and ulcerative colitis (UC) are the two major types of IBD. Transcriptome-wide association study (TWAS) of IBD was first performed using a large-scale genome-wide association study summary data sets of IBD. The FUSION software was applied for TWAS, considering various tissues and cells. The genes identified by TWAS were then validated by the gene expression profiling data sets of IBD. The functional annotation and potential pathways of common differentially expressed genes were further subjected to gene ontology (GO) and pathway enrichment analysis. Integrative analysis of TWAS and messenger RNA (mRNA) expression data detected several tissues related common genes for UC, such as HLA-DRB1 (PTWAS = 0.024; mRNA expression ratio = 1.700) and TAP2 in colon (P TWAS = 0.047; mRNA expression ratio = 2.170). Further comparing the GO enrichment analysis results of TWAS and mRNA expression data, we identified 11 common GO terms for UC, such as plasma membrane (P value = 5.08 × 10-10 ) in intestinal tissues and immune response (P = 0.001) in peripheral blood. We also detected several common pathways for UC, including cell adhesion molecules (P = 0.003) in intestinal tissues, IBD (P = 0.049) in whole blood and phagosome (P = 0.0003) in peripheral blood. Our study results provide novel clues for understanding the genetic mechanism of IBD.

Abstract: Abstract To investigate the mechanical properties and durability of nano-SiO2, polyvinyl alcohol (PVA) fiber-modified fly ash (FA), and metakaolin (MK)-based geopolymer mortar (FMGM), tests of compressive strength, electrical flux, freeze–thaw cycles, and sulfate dry and wet cycles were conducted. Based on the experimental results, combined with Dunn’s gray correlation theory analysis method, a mathematical analysis of the effect sensitivity of the contents of the four mixtures on the mechanical properties and durability of FMGM was carried out. The method of gray correlation analysis can solve the mathematical problem with partial unclear and uncertain information, and the method requires less data and less computation compared with other mathematical analysis method. The results showed that the gray correlation degree between the content of MK and the electric flux value is higher than the that of other comparison sequence and each reference sequence, while the gray correlation degree between the PVA fiber dosage and the loss rate of compressive strength is lower than that of other comparison sequence and each reference sequence. The influence of the four mixture contents on the compressive strength and mass loss rate was in the following decreasing order: MK and FA, PVA fiber, and nano-SiO2. In addition, the influence of the four material mixture contents on the electric flux value and compressive strength loss rate was consistent in the following decreasing order: MK and FA, nano-SiO2, and PVA fiber. Furthermore, the mechanical properties and durability reached the optimum when the PVA fiber content was 0.6% and the dosage of nano-SiO2 was 1.0%. The results of this study can provide a new method for the analysis and evaluation of mechanical properties and durability of nano-SiO2 and PVA fiber-reinforced FMGM in future.

Abstract: In this paper, a dual position-sensitive detector-based vision measurement system camera is built instead of a traditional CCD camera. The 3D position information for the light point is calculated according to the 2D coordinate information of a certain light point in the space illuminated on the two position-sensitive detector (PSD) photosensitive surfaces, which is used for position detection of the spatial light point. In addition, the positioning model for 2D PSDs with different spot sizes in the Gaussian spot mode is derived by the mathematical model of Lucovsky's differential equation for a PSD. For the nonlinear distortion of the PSD, a nonlinear error calibration method using a particle swarm combined with a back propagation neural network is proposed to correct the errors in the measured values through the relationship between the input and output values, to obtain the predicted value that approximates the real coordinates. Then, by comparing the influence of different spot sizes on the positioning accuracy, we conclude that the smaller the spot formed by the convergence of the beam under the optical lens, the higher the positioning accuracy. We believe this conclusion can help improve the accuracy of PSD measurements. Finally, a red LED light spot is set up, and the 3D position measurement and error calibration of the light spot is done by dual PSD cameras, which better solves the position detection problem of a space light spot under close-range conditions because it is fast, reliable, and easy to implement. It also provides an effective method to detect the motion trajectory of a moving light spot in space.

Abstract: Erythropoietic protoporphyria (EPP) is a disorder of heme synthesis in which deficient ferrochelatase activity leads to excessive production and biliary excretion of protoporphyrin. The main clinical features—photosensitivity and hepatobiliary disease that may progress to liver failure—are caused by the toxicity of protoporphyrin. Orthotopic liver transplantation is an effective treatment of liver failure caused by EPP. In this report we have described an EPP Chinese man with end-stage liver disease. He was successfully transplanted. A 3-year follow-up study of protoporphyrin levels, liver tests, and liver biopsies showed no EPP recurrence after liver transplantation.

Abstract: Clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated (Cas) systems provide bacteria and archaea with adaptive immunity against viruses and plasmids by using CRISPR RNAs (crRNAs) to guide the silencing of invading nucleic acids. We show here that in a subset of these systems, the mature crRNA that is base-paired to trans-activating crRNA (tracrRNA) forms a two-RNA structure that directs the CRISPR-associated protein Cas9 to introduce double-stranded (ds) breaks in target DNA. At sites complementary to the crRNA-guide sequence, the Cas9 HNH nuclease domain cleaves the complementary strand, whereas the Cas9 RuvC-like domain cleaves the noncomplementary strand. The dual-tracrRNA:crRNA, when engineered as a single RNA chimera, also directs sequence-specific Cas9 dsDNA cleavage. Our study reveals a family of endonucleases that use dual-RNAs for site-specific DNA cleavage and highlights the potential to exploit the system for RNA-programmable genome editing.
