Historically, work on peripheral circadian clocks has been focused on organs and tissues that have prominent metabolic functions, such as the liver, fat, and muscle. In recent years, skin has emerged as a model for studying circadian clock regulation of cell proliferation, stem cell functions, tissue regeneration, aging, and carcinogenesis. Morphologically, skin is complex, containing multiple cell types and structures, and there is evidence for a functional circadian clock in most, if not all, of its cell types. Despite the complexity, skin stem cell populations are well defined, experimentally tractable, and exhibit prominent daily cell proliferation cycles. Hair follicle stem cells also participate in recurrent, long-lasting cycles of regeneration: the hair growth cycles. Among other advantages of skin is a broad repertoire of available genetic tools enabling the creation of cell type-specific circadian mutants. Also, due to the accessibility of skin, in vivo imaging techniques can be readily applied to study the circadian clock and its outputs in real time, even at the single-cell level. Skin provides the first line of defense against many environmental and stress factors that exhibit dramatic diurnal variations such as solar ultraviolet (UV) radiation and temperature. Studies have already linked the circadian clock to the control of UVB-induced DNA damage and skin cancers. Due to the important role that skin plays in the defense against microorganisms, it also represents a promising model system to further explore the role of the clock in the regulation of the body's immune functions. To that end, recent studies have already linked the circadian clock to psoriasis, one of the most common immune-mediated skin disorders. Skin also provides opportunities to interrogate the clock regulation of tissue metabolism in the context of stem cells and regeneration. Furthermore, many animal species feature prominent seasonal hair molt cycles, offering an attractive model for investigating the role of the clock in seasonal organismal behaviors.

The Circadian Clock in Skin: Implications for Adult Stem Cells, Tissue Regeneration, Cancer, Aging, and Immunity

Amphibians and reptiles have evolved with natural lighting cycles. Consequently, alteration of natural variation in diurnal and nocturnal light intensities and spectral properties has the potential to disrupt their physiology, behavior, and ecology. We review the possible effects of night lighting on many species of amphibians and reptiles, noting that few studies of the consequences of artificial lights to amphibians and reptiles have been conducted to date. The one exception is the information available on the negative impacts of artificial lights on hatchling sea turtles, which has received considerable coverage in both scientific and popular media. In many studies that might be relevant, researchers have not recorded the illumination or irradiance at which experiments were conducted. We identify light pollution as a serious threat that should be considered as part of planning and management decisions in the maintenance or conservation of urban areas containing amphibians and reptiles. However, we consider it too early to precisely gauge the effects of artificial night lighting on other taxa found in light-polluted environments or provide specific management recommendations, beyond pointing out th e urgent need for more information.

EffEcts of ArtificiAl Night lightiNg oN AmphibiANs ANd rEptilEs iN UrbAN ENviroNmENts

Artificial light at night (ALAN) is increasing exponentially worldwide, accelerated by the transition to new efficient lighting technologies. However, ALAN and resulting light pollution can cause unintended physiological consequences. In vertebrates, production of melatonin—the “hormone of darkness” and a key player in circadian regulation—can be suppressed by ALAN. In this paper, we provide an overview of research on melatonin and ALAN in vertebrates. We discuss how ALAN disrupts natural photic environments, its effect on melatonin and circadian rhythms, and different photoreceptor systems across vertebrate taxa. We then present the results of a systematic review in which we identified studies on melatonin under typical light-polluted conditions in fishes, amphibians, reptiles, birds, and mammals, including humans. Melatonin is suppressed by extremely low light intensities in many vertebrates, ranging from 0.01–0.03 lx for fishes and rodents to 6 lx for sensitive humans. Even lower, wavelength-dependent intensities are implied by some studies and require rigorous testing in ecological contexts. In many studies, melatonin suppression occurs at the minimum light levels tested, and, in better-studied groups, melatonin suppression is reported to occur at lower light levels. We identify major research gaps and conclude that, for most groups, crucial information is lacking. No studies were identified for amphibians and reptiles and long-term impacts of low-level ALAN exposure are unknown. Given the high sensitivity of vertebrate melatonin production to ALAN and the paucity of available information, it is crucial to research impacts of ALAN further in order to inform effective mitigation strategies for human health and the wellbeing and fitness of vertebrates in natural ecosystems.

Light Pollution, Circadian Photoreception, and Melatonin in Vertebrates

The pineal gland of poikilothermic vertebrates originates as an evagination from the diencephalic roof between the habenular and the posterior commissures, and associates with a parapineal organ to form the so-called pineal complex. The pinealocytes may be photosensitive, secretory or intermediate cells between both. Melatonin, the indoleamine secreted by the pineal, exhibits a circadian secretory rhythm that conveys environmental information to the organism. The peak melatonin secretion occurs during the night, although there are a few examples of an increase in indoleamine secretion during the day. Melatonin is also synthesized in other sites such as the retina, and it has been found in many invertebrates and unicellular organisms. The rhythmic secretory pattern of melatonin is responsible for many biological rhythms exhibited by lower vertebrates. These rhythms are abolished by pinealectomy in some species, but not in others, suggesting the existence of an extra-pineal pacemaker. The photoperiod and the temperature (especially in reptiles) are the main environmental factors affecting the secretory rhythm of melatonin. Poikilothermic vertebrates exhibit a circadian rhythmic color change, with nocturnal blanching, usually related to melatonin secretion. In amphibians, melatonin exhibits a potent skin lightening activity. However, in fishes and reptiles the melatonin effects vary with the species, the developmental stage, and the pigment cell location. Melatonin also exerts inhibitory or excitatory activity on the amphibian reproductive system, regulation of circadian locomotory activity in reptiles, and modulation of the amphibian metamorphosis. Melatonin has also a modulatory effect on the response of target cells to different hormones and high concentrations or prolonged exposure to the indoleamine may cause autodesensitization in various tissues. Binding sites of melatonin have been detected in the central nervous system and peripheral tissues of various vertebrates. The relative potencies of melatonin analogues demonstrated two subtypes of melatonin receptors (ML-1 and ML-2). A transmembrane melatonin receptor has been cloned from Xenopus laevis melanophores; it belongs to the family of the G protein-coupled receptors and exhibits 85% homology with the mammalian nervous system receptor. Melatonin binding sites in the nucleus of many cell types and its potent intracellular anti-oxidant action suggest mechanisms of action other than through the G-protein coupled receptor.

Comparative aspects of the pineal/melatonin system of poikilothermic vertebrates

Influences of photoperiod, temperature and melatonin were examined on development and color patterns of nymphs, and sternum color and reproductive fate of adults in Halyomorpha halys (brevis). Short-day accelerated nymphal development, whereas long-day accelerated reproductive maturation. The two types of photoperiodic responses at different stages may help maintain the univoltinism of this species in the field, assuring the right timing for diapause and reproduction. The pronotum of fifth instar nymphs reared under LD 11:13 shows a brown-marbled color pattern darker, with less creamy-yellowish speckles, than that of nymphs reared under LD 16:8. Short-day-reared fifth instar nymphs, which are destined to diapause in adults, had shorter white stripes on the pronotum, smaller body size, less frequent feeding and more lipid accumulation than the long-day-reared. The longer the exposure to LD 13:11 at 20 degrees C during the nymphal stage, the greater the expression of short-day associated characteristics observed in the fifth nymphal instar and adult stage. Melatonin orally administered (500 microg/ml) to insects under LD 16:8 produced slightly reduced body size, suppressed feeding, extended nymphal period and increased accumulation of lipid compared to the untreated bugs, but did not affect the incidence of diapause in females. In males, melatonin retarded gonadal development under LD16:8. These results may suggest a possible role of melatonin for the control of seasonal polyphenism and development.

Effects of photoperiod, temperature and melatonin on nymphal development, polyphenism and reproduction in Halyomorpha halys (Heteroptera: Pentatomidae).

Melanophores were studied in tadpoles of the South African clawed toad, Xenopus laevis, during the first week after hatching (stages 46-49) at 25 degrees C. The tadpoles had melanophores with dispersed melanosomes in the light and punctate melanophores in the dark in LD 12:12. The melanophores remained punctate in constant dark and the melanosomes remained dispersed in constant light. Lights-out (in the light-time of LD 12:12) caused the melanophores to become punctate, which occurred more quickly than the dispersion of melanosomes, which commenced when the lights were turned on (in the dark-time of LD 12:12). Melanophores with dispersed melanosomes in tadpoles (in constant light) became punctate in response to a series of melatonin concentrations (0.2-5 ng/ml) in their bathing water irrespective of the time of day melatonin was administered. An image-analysis technique for assessing melanophore responses was tested.
