Troyanskaya,Howard Bussey,Gary D. Bader,Anne-Claude Gingras,Quaid Morris,Philip M. Kim,Chris A. Kaiser,Chad L. Myers,Brenda J. Andrews,Charles Boone +52 more
TL;DR: A network based on genetic interaction profiles reveals a functional map of the cell in which genes of similar biological processes cluster together in coherent subsets, and highly correlated profiles delineate specific pathways to define gene function.
Journal ArticleDOI
Systematic exploration of essential yeast gene function with temperature-sensitive mutants
Zhijian Li,Franco J. Vizeacoumar,Sondra Bahr,Jingjing Li,Jonas Warringer,Frederick S. Vizeacoumar,Renqiang Min,Benjamin VanderSluis,Jeremy Bellay,Michael Devit,James A. Fleming,Andrew D. Stephens,Julian Haase,Zhen Yuan Lin,Anastasia Baryshnikova,Hong Lu,Zhun Yan,Ke Jin,Sarah L. Barker,Alessandro Datti,Alessandro Datti,Guri Giaever,Corey Nislow,Chris Bulawa,Chad L. Myers,Michael Costanzo,Anne-Claude Gingras,Zhaolei Zhang,Anders Blomberg,Kerry Bloom,Brenda J. Andrews,Charles Boone +31 more
TL;DR: This mutant collection should facilitate a wide range of systematic studies aimed at understanding the functions of essential genes, including roles for cohesin and condensin genes in spindle disassembly.
Journal ArticleDOI
Essential Gene Profiles in Breast, Pancreatic, and Ovarian Cancer Cells
Richard Marcotte,Kevin R. Brown,Fernando Suarez,Azin Sayad,Konstantina Karamboulas,Paul M. Krzyzanowski,Fabrice Sircoulomb,Mauricio Medrano,Yaroslav Fedyshyn,Judice L. Y. Koh,Dewald van Dyk,Bohdana Fedyshyn,Marianna Luhova,Glauber C. Brito,Franco J. Vizeacoumar,Frederick S. Vizeacoumar,Alessandro Datti,Dahlia Kasimer,Alla Buzina,Patricia Mero,Christine M. Misquitta,Josee Normand,Maliha Haider,Troy Ketela,Jeffrey L. Wrana,Robert Rottapel,Benjamin G. Neel,Jason Moffat +27 more
TL;DR: This study identified essential gene profiles in 72 breast, pancreatic, and ovarian cancer cell lines using a lentiviral shRNA library targeting ~16,000 genes and a newly developed, dynamic scoring approach, to facilitate the systematic identification of drivers, unanticipated synthetic lethal relationships, and functional vulnerabilities of these tumor types.
Journal ArticleDOI
Interaction landscape of membrane - protein complexes in Saccharomyces cerevisiae
Mohan Babu,James Vlasblom,Shuye Pu,Xinghua Guo,Chris Graham,Björn D. M. Bean,Helen E. Burston,Franco J. Vizeacoumar,Jamie Snider,Sadhna Phanse,Vincent Fong,Yuen Yi C. Tam,Michael Davey,Olha Hnatshak,Navgeet Bajaj,Shamanta Chandran,Thanuja Punna,Constantine Christopolous,Victoria Wong,Analyn Yu,Gouqing Zhong,Joyce Li,Igor Stagljar,Elizabeth Conibear,Shoshana J. Wodak,Andrew Emili,Jack Greenblatt +26 more
TL;DR: A complete survey of putative integral, peripheral and lipid-anchored MPs from Saccharomyces cerevisiae was reported, which revealed unexpected physical associations underlying the membrane biology of eukaryotes and delineates the global topological landscape of the membrane interactome.
Book ChapterDOI
Synthetic genetic array (SGA) analysis in Saccharomyces cerevisiae and Schizosaccharomyces pombe.
Anastasia Baryshnikova,Michael Costanzo,Scott J. Dixon,Franco J. Vizeacoumar,Chad L. Myers,Brenda J. Andrews,Charles Boone +6 more
TL;DR: An array-based approach automates yeast genetic analysis in general and can be easily adapted for a number of different genetic screens or combined with high-content screening systems to quantify the activity of specific reporters in genome-wide sets of single or more complex multiple mutant backgrounds.
