Background: The total flavonoids from ethanol extract of the aerial part of Ziziphora clinopodioides Lam. (Lamlaceae) (Xintahua) showed protective activities against rat acute myocardial ischemia in rats. This study aims to isolate acacetin, a flavonoid, from the aerial part of Z. clinopodioides, to develop an HPLC method for its detection, and to evaluate its protective effects on neonatal rat cardiomyocytes. Methods: Sephadex LH-20 silicagel and pillar layer chromatography silica gel were applied for the isolation and purification of acacetin and its structure was elucidated on the basis of 1 H and 13 C NMR spectroscopy. The content of acacetin in Z. clinopodioides collected from three different origins was determined by HPLC. The neonatal rat cardiomyocytes were isolated and cultured in vitro to establish a hypoxia/reoxygenation injury model. The viability of cardiomyocytes was measured by the MTT method. Changes of malondialdehyde (MDA) content in the medium were also determined. Results: The acacetin content in various batches of Z. clinopodioides ranged from 45.50 to 47.41 μg/g. Acacetin of 25, 10, 5 μg/mL significantly decreased the MDA content in a model of hypoxia/reoxygenation injury (P < 0.001, P< 0.001 and P = 0.033, respectively). Conclusions: Acacetin protects neonatal cardiomyocytes from the damage induced by hypoxia/reoxygenation stress through reduction of lipid peroxidation and enhancement of the antioxidant activity.

Protective effects of acacetin isolated from Ziziphora clinopodioides Lam. (Xintahua) on neonatal rat cardiomyocytes

The volatile organic compounds from flowers, leaves, and stems of Dendranthema indicum var. aromaticum, obtained through a static headspace technique, were analyzed by gas chromatography-mass spectrometry (GC-MS) and accurate mass measurement. The qualitative approach, comprising accurate mass measurement, retention index, and mass spectral search, was utilized to identify compounds. A total of 162 components were identified, representing 97.55–98.72% of the volatiles of individual samples. The principal chemical components in flowers were bornyl acetate (15.40%), α-phellandrene (14.18%), p-cymene (9.64%), camphor (9.54%), β-linalool (8.61%), and α-thujone (7.06%). In leaves, the main components were p-cymene (20.42%), bornyl acetate (20.41%), α-phellandrene (13.67%), and β-linalool (5.46%). Analysis of volatile organic compounds from Dendranthema indicum var. aromaticum by headspace gas chromatography-mass spectrometry and accurate mass measurement.

Dendranthema indicum var. aromaticum is a new species with strong fragrance and is used as a herbal medicine by Chinese folks. The abundant flavonoids play important roles in its pharmacological activities. In this study, an ultrasound-assisted method was used to extract total flavonoids (TF) from D. indicum var. aromaticum by response surface methodology. A quadratic model was developed to optimize the extraction conditions, whose accuracy was verified by statistic analysis. Ethanol and acetic acid at the volume ratio of 70% : 2% were selected as the extract solvent. The optimized extraction conditions were as follows: extraction time, 40 min; solid/liquid ratio, 1 : 23 g/mL; and temperature, 60°C. This is the first report of an efficient and easy-operating method for TF extraction from D. indicum var. aromaticum. Besides, this study provides reference for future pharmacological research on D. indicum var. aromaticum and extraction of bioactive components from other herbs.

Optimization of Ultrasound-Assisted Extraction of Total Flavonoids from Dendranthema indicum var. aromaticum by Response Surface Methodology.

Chrysanthemum indicum var. aromaticum is an important resource plant for breeding excellent aromatic varieties of ornamental and medicinal chrysanthemums. However, the biosynthesis and regulation mechanisms of typically aromatic ingredients in C. indicum var. aromaticum are poorly understood, which restricted the utilization of this plant. Here, the transcriptome of C. indicum var. aromaticum was investigated and comparative analysis was conducted with the other two closely related chrysanthemum taxa C. indicum and C. nankingense. The global transcriptome profiles showed clear difference of gene expression patterns between the three chrysanthemums, of which 52 flavonoid biosynthesis-associated genes, 157 transcription factors, and 24 flavonoid Uridine diphosphate (UDP) -glucuronosyl and -glucosyltransferase genes were significantly differentially expressed. Furthermore, 22 differential expression genes (DEGs) in relation to the terpenoid backbone biosynthesis and 183 DEGs related to terpenoid diversification were identified. Of which, 14 genes involved into TS/CYP gene cluster showed higher expression in CIA than those in CI or CN. These results increased our understanding of flavonoid diversification and aromatic ingredients accumulation in plants, and will also help improve resource conservation and sustainable utilization of the important resource plant C. indicum var. aromaticum.
