Osteoprotegerin: A Novel Secreted Protein Involved in the Regulation of Bone Density
William S. Simonet,David L. Lacey,Colin R. Dunstan,Michael J. Kelley,Ming-Shi Chang,R Lüthy,Hung Q. Nguyen,S Wooden,L Bennett,Tom Boone,Grant Shimamoto,Margaret L. DeRose,Robin Elliott,Anne Colombero,Hong-Lin Tan,Geraldine Trail,John K. Sullivan,E. Davy,N. Bucay,L Renshaw-Gegg,T.M Hughes,Dave Hill,W Pattison,Patricia Shehan Campbell,S Sander,Gwyneth Van,John E. Tarpley,P Derby,Richard Lee,William J. Boyle +29 more
TL;DR: Data show that OPG can act as a soluble factor in the regulation of bone mass and imply a utility for OPG in the treatment of osteoporosis associated with increased osteoclast activity.
Journal ArticleDOI
OPGL is a key regulator of osteoclastogenesis, lymphocyte development and lymph-node organogenesis
Young-Yun Kong,Hiroki Yoshida,Ildiko Sarosi,Hong-Lin Tan,Emma Timms,Casey Capparelli,Sean Morony,Antonio J. Oliveira-dos-Santos,Gwyneth Van,Annick Itie,Wilson Khoo,Andrew Wakeham,Colin R. Dunstan,David L. Lacey,Tak W. Mak,William J. Boyle,Josef M. Penninger +16 more
TL;DR: OPGL is a new regulator of lymph-node organogenesis and lymphocyte development and is an essential osteoclast differentiation factor in vivo.
Journal ArticleDOI
Activated T cells regulate bone loss and joint destruction in adjuvant arthritis through osteoprotegerin ligand
Young-Yun Kong,Ulrich Feige,Iidiko Sarosi,Brad Bolon,Anna Tafuri,Sean Morony,Casey Capparelli,Ji Li,Robin Elliott,Susan McCabe,Tom Wong,G Campagnuolo,Erika Moran,Earl R. Bogoch,Gwyneth Van,Linh T. Nguyen,Pamela S. Ohashi,David L. Lacey,Eleanor N. Fish,William J. Boyle,Josef M. Penninger,Josef M. Penninger +21 more
TL;DR: The results show that both systemic and local T-cell activation can lead to OPGL production and subsequent bone loss, and they provide a novel paradigm for T cells as regulators of bone physiology.
Journal ArticleDOI
TRAF6 deficiency results in osteopetrosis and defective interleukin-1, CD40, and LPS signaling
Mark A. Lomaga,Wen Chen Yeh,Ildiko Sarosi,Gordon S. Duncan,Caren Furlonger,Alexandra Ho,Sean Morony,Casey Capparelli,Gwyneth Van,Stephen Kaufman,Annette Van Der Heiden,Annick Itie,Andrew Wakeham,Wilson Khoo,Takehiko Sasaki,Zhaodan Cao,Josef M. Penninger,Christopher J. Paige,David L. Lacey,Colin R. Dunstan,William J. Boyle,David V. Goeddel,Tak W. Mak +22 more
TL;DR: It is reported that mice deficient in TNF receptor-associated factor 6 (TRAF6) are osteopetrotic with defects in bone remodeling and tooth eruption due to impaired osteoclast function, and it is demonstrated that TRAF6 is crucial not only in IL-1 and CD40 signaling but also, surprisingly, in LPS signaling.
Journal ArticleDOI
RANK is the intrinsic hematopoietic cell surface receptor that controls osteoclastogenesis and regulation of bone mass and calcium metabolism
Ji Li,Ildiko Sarosi,Xiao-Qiang Yan,Sean Morony,Casey Capparelli,Hong-Lin Tan,Susan McCabe,Robin Elliott,Sheila Scully,Gwyneth Van,Stephen J. Kaufman,Shao-Chieh Juan,Yu Sun,John E. Tarpley,Laura Martín,Kathleen Christensen,James McCabe,Paul J. Kostenuik,Hailing Hsu,Frederick A. Fletcher,Colin R. Dunstan,David L. Lacey,William J. Boyle +22 more
TL;DR: Data indicate that RANK is the intrinsic cell surface determinant that mediates osteoprotegerin ligand effects on bone resorption and remodeling as well as the physiological and pathological effects of calciotropic hormones and proresorptive cytokines.
