The Genome of M. Acetivorans Reveals Extensive Metabolic and Physiological Diversity

James E. Galagan, +76 more
- 01 Apr 2002 -
TL;DR: The complete genome sequence of an acetate-utilizing methanogen, Methanosarcina acetivorans C2A, is reported, which indicates the likelihood of undiscovered natural energy sources for methanogenesis, whereas the presence of single-subunit carbon monoxide dehydrogenases raises the possibility of nonmethanogenic growth.
Journal ArticleDOI

Genetic analysis of the archaeon Methanosarcina barkeri Fusaro reveals a central role for Ech hydrogenase and ferredoxin in methanogenesis and carbon fixation

TL;DR: Results obtained from growth experiments, cell suspension experiments, and enzyme activity measurements in cell extracts provide compelling evidence for essential functions of Ech and a 2[4Fe-4S] ferredoxin in the metabolism of M. barkeri.
Journal ArticleDOI

Directed Mutagenesis and Plasmid-Based Complementation in the Methanogenic Archaeon Methanosarcina acetivorans C2A Demonstrated by Genetic Analysis of Proline Biosynthesis

TL;DR: The method employs homologous recombination-mediated gene replacement and was used to construct a variety of proline auxotrophs with mutations in the proABC locus, the first use of directed mutagenesis in Methanosarcina acetivorans C2A.
