Photodynamic therapy involves administration of a tumor-localizing photosensitizing agent, which may require metabolic synthesis (i.e., a prodrug), followed by activation of the agent by light of a specific wavelength. This therapy results in a sequence of photochemical and photobiologic processes that cause irreversible photodamage to tumor tissues. Results from preclinical and clinical studies conducted worldwide over a 25-year period have established photodynamic therapy as a useful treatment approach for some cancers. Since 1993, regulatory approval for photodynamic therapy involving use of a partially purified, commercially available hematoporphyrin derivative compound (Photofrin) in patients with early and advanced stage cancer of the lung, digestive tract, and genitourinary tract has been obtained in Canada, The Netherlands, France, Germany, Japan, and the United States. We have attempted to conduct and present a comprehensive review of this rapidly expanding field. Mechanisms of subcellular and tumor localization of photosensitizing agents, as well as of molecular, cellular, and tumor responses associated with photodynamic therapy, are discussed. Technical issues regarding light dosimetry are also considered.

Photodynamic Therapy

In the choice of an estimator for the spectrum of a stationary time series from a finite sample of the process, the problems of bias control and consistency, or "smoothing," are dominant. In this paper we present a new method based on a "local" eigenexpansion to estimate the spectrum in terms of the solution of an integral equation. Computationally this method is equivalent to using the weishted average of a series of direct-spectrum estimates based on orthogonal data windows (discrete prolate spheroidal sequences) to treat both the bias and smoothing problems. Some of the attractive features of this estimate are: there are no arbitrary windows; it is a small sample theory; it is consistent; it provides an analysis-of-variance test for line components; and it has high resolution. We also show relations of this estimate to maximum-likelihood estimates, show that the estimation capacity of the estimate is high, and show applications to coherence and polyspectrum estimates.

Infrared spectroscopy of proteins.

A significant modification to the additive all-atom CHARMM lipid force field (FF) is developed and applied to phospholipid bilayers with both choline and ethanolamine containing head groups and with both saturated and unsaturated aliphatic chains. Motivated by the current CHARMM lipid FF (C27 and C27r) systematically yielding values of the surface area per lipid that are smaller than experimental estimates and gel-like structures of bilayers well above the gel transition temperature, selected torsional, Lennard-Jones and partial atomic charge parameters were modified by targeting both quantum mechanical (QM) and experimental data. QM calculations ranging from high-level ab initio calculations on small molecules to semiempirical QM studies on a 1,2-dipalmitoyl-sn-phosphatidylcholine (DPPC) bilayer in combination with experimental thermodynamic data were used as target data for parameter optimization. Update of the CHARMM All-Atom Additive Force Field for Lipids: Validation on Six Lipid Types

We have used the pH-induced self-assembly of a peptide-amphiphile to make a nanostructured fibrous scaffold reminiscent of extracellular matrix. The design of this peptide-amphiphile allows the nanofibers to be reversibly cross-linked to enhance or decrease their structural integrity. After cross-linking, the fibers are able to direct mineralization of hydroxyapatite to form a composite material in which the crystallographic c axes of hydroxyapatite are aligned with the long axes of the fibers. This alignment is the same as that observed between collagen fibrils and hydroxyapatite crystals in bone.

Self-assembly and mineralization of peptide-amphiphile nanofibers

Fourier transform infrartd (FTIR) spectroscopy is an established tool for the structural character- ization of proteins. However, many potential pitfalls exist for the unwary investigator. Determination of protein secondary structure by Fourier transform infrared spectroscopy: a critical assessment.

New insight into protein secondary structure from resolution-enhanced infrared spectra.

The general theory of Fourier self-deconvolution, i.e., spectral deconvolution using Fourier transforms and the intrinsic line-shape, is developed. The method provides a way of computationally resolving overlapped lines that can not be instrumentally resolved due to their intrinsic linewidth. Examples of the application of the technique to synthetic and experimental infrared spectra are presented, and potential applications are discussed. It is shown that lines in spectra having moderate signal/noise ratios (∼1000) can readily be reduced in width by a factor of 3. The method is applicable to a variety of spectroscopic techniques.
