Abstract: Examining advances in the class of natural and non-natural piperidine azasugars, important therapeutic agents and potent glycosidase inhibitors, this review looks at syntheses applying olefin metathesis as a highly efficient key step and gateway strategy for discovery of better iminosugar leads for treatment of widespread affections like viral and metabolic diseases. Amply illustrated is how ring-closing metathesis (RCM and RCEYM), promoted by commercial ruthenium alkylidene catalysts, manage to construct the common tetrahydropyridine core while cross-metathesis (CM), starting from this generic scaffold, provides general access to families of novel azasugars. Special consideration is given to high-profile iminosugar drugs of this class (1-deoxynojirimycin and congeners, adenophorine, fagomine, isofagomine and some of their N- and C-substituted analogues) stressing upon newest trends for enhancing biological activity and modulating previously unexploited targets in multispecific therapies.

Abstract: The synthesis of complexes of the general formula Cl2Ru(SIMes)(L)(3-phenylinden-1-ylidene) (5, L = PCy3; 6, L = py; and 7, L = PPh3) from Cl2Ru(PR3)2(3-phenylinden-1-ylidene) (2a, R = Ph; 2b, R = Cy) is reported. This family of olefin metathesis catalysts was fully characterized (1H, 13C and 31P NMR spectroscopy and elemental analysis) and provided excellent activity in the ring-opening metathesis polymerization of 1,5-cyclooctadiene and the ring-closing metathesis of diethyl diallylmalonate. Comparison with the corresponding benzylidene-containing catalysts, 1a,c and 8b, established the decisive role of the carbene ligand on the procedure of the reaction and led to the observation of an unusual catalytic phenomenon, here called “self-inhibition”.(© Wiley-VCH Verlag GmbH & Co. Abstract: The fascinating story of olefin (or alkene) metathesis (eq 1) began almost five decades ago, when Anderson and Merckling reported the first carbon-carbon double-bond rearrangement reaction in the titanium-catalyzed polymerization of norbornene. Nine years later, Banks and Bailey reported “a new disproportionation reaction . . . in which olefins are converted to homologues of shorter and longer carbon chains...”. In 1967, Calderon and co-workers named this metal-catalyzed redistribution of carbon-carbon double bonds olefin metathesis, from the Greek word “μeτάθeση”, which means change of position. These contributions have since served as the foundation for an amazing research field, and olefin metathesis currently represents a powerful transformation in chemical synthesis, attracting a vast amount of interest both in industry and academia.
