Water uptake and extrusion by mitochondria in relation to oxidative phosphorylation.

The mitochondrial permeability transition (PT) is a permeability increase of the inner mitochondrial membrane mediated by a channel, the permeability transition pore (PTP). After a brief historical introduction, we cover the key regulatory features of the PTP and provide a critical assessment of putative protein components that have been tested by genetic analysis. The discovery that under conditions of oxidative stress the F-ATP synthases of mammals, yeast, and Drosophila can be turned into Ca2+-dependent channels, whose electrophysiological properties match those of the corresponding PTPs, opens new perspectives to the field. We discuss structural and functional features of F-ATP synthases that may provide clues to its transition from an energy-conserving into an energy-dissipating device as well as recent advances on signal transduction to the PTP and on its role in cellular pathophysiology.

The Mitochondrial Permeability Transition Pore: Channel Formation by F-ATP Synthase, Integration in Signal Transduction, and Role in Pathophysiology

The permeability transition (PT) denotes an increase of the mitochondrial inner membrane permeability to solutes with molecular masses up to about 1500 Da. It is presumed to be mediated by opening of a channel, the permeability transition pore (PTP), whose molecular nature remains a mystery. Here I briefly review the history of the PTP, discuss existing models, and present our new results indicating that reconstituted dimers of the FOF1 ATP synthase form a channel with properties identical to those of the mitochondrial megachannel (MMC), the electrophysiological equivalent of the PTP. Open questions remain, but there is now promise that the PTP can be studied by genetic methods to solve the large number of outstanding problems.

The effect of thyroxine and other substances on the swelling of isolated rat liver mitochondria.

The vesicles of fragmented sarcoplasmic reticulum, i.e. the physiological relaxing factor, remove most of the exchangeable Ca bound to actomyosin and myofibrils. The extent to which they remove Ca and the extent to which they inhibit myofibrillar activity are closely correlated. Previous work has shown that those in vitro reactions of actomyosin with ATP which are equivalent to its contraction, i.e. superprecipitation and a high ATPase activity, require the formation of a Ca-actomyosin complex, and that actomyosin after the removal of most of its bound Ca is inhibited by physiological concentrations of ATP. The evidence now suggests that the factor achieves its relaxing effect through the dissociation of the Ca-actomyosin complex and that it has no other direct influence on the biological activity of actomyosin. Experiments, showing that under similar conditions the vesicles of the factor are capable of reducing the concentration of ionized Ca in the surrounding medium to about 0.02 µM and less, suggest that the factor competes successfully with actomyosin for the available Ca through its mechanism of Ca accumulation.

On the Mechanism of the Relaxing Effect of Fragmented Sarcoplasmic Reticulum

Ethylenediaminetetra-aceticacid and other chelating agents of the same type inhibit the swelling of liver mitochondria and enhance their oxygen consumption. This is probably due to complex formation of the Calcium ions.

Die Mg2+-Abhangigkeit der durch Hexokinase katalysierten Ubertragung eines Phosphatrests von ATP auf Glucose wird mit Hilfe von Metallpuffern untersucht. Die pMg-Aktivitatskurve fallt im pMg-Bereich 4-6 ab. Die Interpretation dieses Befundes fuhrt zum Schluss, dass das aktive Ferment eine Mg-Protein-Verbindung ist, die bei ab- nehmender Magnesiumionenkonzentration in Mg2+ und inaktives Protein dissoziiert.

Es wurde die Wirkung von verschiedenen Komplexbildnern (Komplexon, Glycolkomplexon, ATP, ADP u. a.) und von zweiwertigen Kationen (in erster Linie Calcium und Xagnesium) auf das Enzymsystem der α-Ketoglutarsaure-Oxydation nach Von Korff2 untersucht. Die Aminopolycarbonsauren Komplexon und Glycolkomplexon hemmen die Reaktion schon in der Konzentration von 10−4-m. vollstandig. Ein nachtraglicher Zusatz von Calcium im Uberschuss fuhrt in beiden Fallen zu einer totalen Reaktivierung, wahrend Magnesium nur im Falle der Komplexonhemmung partiell wirksam ist. Bei Berucksichtigung der Komplexbildungskonstanten kann gefolgert werden, dass Magnesium nur indirekt durch Verdrangung des Calciums aus dem Calcium-Komplexon wirksam ist.

Über die Wirkung von Komplexbildnern und Metallkationen auf die enzymatische Oxydation von Ketoglutarsäure

Die Abhangigkeit der Aktivitat einer ADP-spaltenden Phosphatase aus Kartoffeln von der Calciumionenkonzentration wird mit Hilfe von Calciumpuffern untersucht. Die pCa-Aktivitatskurve des Ferments zeigt, dass der Aktivitatsabfall im pCa-Bereich 3,8-5,5 erfolgt. Daraus kann geschlossen werden, dass eine Calcium-Protein-Verbin-dung (eventuell ein Calcium-Protein-ADP-Komplex) und nieht der Calcium-ADP-Komplex die Calciumabhangigkeit der Reaktion bedingt.
