Antiinflammatory Therapy with Canakinumab for Atherosclerotic Disease
Paul M. Ridker,Brendan M. Everett,Tom Thuren,Jean G. MacFadyen,William Chang,Christie M. Ballantyne,Francisco H. Fonseca,Jose C. Nicolau,Wolfgang Koenig,Stefan D. Anker,John J.P. Kastelein,Jan H. Cornel,Prem Pais,Daniel Pella,Jacques Genest,Renata Cifkova,Alberto J. Lorenzatti,Tamás Forster,Zhanna Kobalava,Luminita Vida-Simiti,Marcus Flather,Hiroaki Shimokawa,Hisao Ogawa,Mikael Dellborg,Paulo Roberto Ferreira Rossi,Roland P.T. Troquay,Peter Libby,Robert J. Glynn +27 more
TL;DR: Antiinflammatory therapy targeting the interleukin‐1β innate immunity pathway with canakinumab at a dose of 150 mg every 3 months led to a significantly lower rate of recurrent cardiovascular events than placebo, independent of lipid‐level lowering.
Journal ArticleDOI
Rosuvastatin to prevent vascular events in men and women with elevated C-reactive protein.
Paul M. Ridker,Eleanor Danielson,Jacques Genest,Antonio M. Gotto,Wolfgang Koenig,Peter Libby,Alberto J. Lorenzatti,Jean G. MacFadyen,Børge G. Nordestgaard,James Shepherd,James T. Willerson,Robert J. Glynn +11 more
TL;DR: In this trial of apparently healthy persons without hyperlipidemia but with elevated high-sensitivity C-reactive protein levels, rosuvastatin significantly reduced the incidence of major cardiovascular events.
Journal ArticleDOI
Rosuvastatin to Prevent Vascular Events in Men and Women With Elevated C-Reactive Protein
Paul M. Ridker,Eleanor Danielson,Francisco Antonio Helfenstein Fonseca,Jacques Genest,Antonio M. Gotto,John J.P. Kastelein,Wolfgang Koenig,Peter Libby,Alberto J. Lorenzatti,Jean G. MacFadyen,Børge G. Nordestgaard,James Shepherd,James T. Willerson,Robert J. Glynn +13 more
TL;DR: It is shown that statin therapy reduces high-sensitivity C-reactive protein levels as well as cholesterol, but no prospective trials have directly addressed the issue of whether elevated levels of high-magnifying C- reactive protein are beneficial for apparently healthy people.
Journal ArticleDOI
The metabolic syndrome and cardiovascular risk a systematic review and meta-analysis.
Salvatore Mottillo,Kristian B. Filion,Jacques Genest,Lawrence Joseph,Louise Pilote,Paul Poirier,Stéphane Rinfret,Ernesto L. Schiffrin,Mark J. Eisenberg +8 more
TL;DR: A systematic review and meta-analysis of the cardiovascular risk associated with the metabolic syndrome as defined by the 2001 NCEP and 2004 revised National Cholesterol Education Program definitions found the syndrome is associated with a 2-fold increase in cardiovascular outcomes and a 1.5- fold increase in all-cause mortality.
Journal ArticleDOI
Mutations in ABC1 in Tangier disease and familial high-density lipoprotein deficiency
Angela Brooks-Wilson,Michel Marcil,Susanne M. Clee,Lin-Hua Zhang,Kirsten Roomp,M. Van Dam,Lu Yu,C. Brewer,Jennifer A. Collins,H.O. Molhuizen,O. Loubser,B. F. F. Ouelette,Keith Fichter,K. J. D. Kastelein,Jacques Genest,Michael R. Hayden +25 more
TL;DR: Two Tangier disease families are identified, confirmed 9q31 linkage and the disease locus is refined to a limited genomic region containing the gene encoding the ATP-binding cassette transporter (ABC1), indicating that TD and FHA are allelic.
