Journal ArticleDOI
TL;DR: A wide variety of Platinum(II) complexes have been investigated for antitumor activity against Sarcoma 180 in Swiss white female mice as mentioned in this paper, and the results showed that only neutral complexes exhibit activity while charged species are inactive and relatively nontoxic.

TL;DR: The binding of cis(c)- and trans(t)-Pt(NH3)2Cl2 to DNA at platinum/DNA-nucleotide ratios (Ri) of 0.1 or less has been studied by means of radioactive 195mPt-labeled compounds, suggesting that both compounds should bind to DNA under biological conditions.

TL;DR: A wide range of asymmetric Schiff base ligands containing primary amine functions can be prepared in one step and in high yield by the reaction of VO(OEt) 3 with salicylaldehyde and the appropriate diamine.
Abstract: A wide range of asymmetric Schiff base ligands containing primary amine functions can be prepared in one step and in high yield by the reaction of VO(OEt) 3 with salicylaldehyde and the appropriate diamine. The resulting VO 2 L complexes contain tridentate ligands with phenolate, imine, and amine coordination to the dioxovanadium(V) complex ion. An X-ray structure of one compound ([VO 2 (1,2-pnSAL), 5A, where 1,2-pnSAL=1(N-salicylideneamina)-2-aminopropane) demonstrates that it is a dimer in the solid state with a V 2 O 4 score

TL;DR: Recently, there has been a surge of activity, based on a great deal of mechanistic information, aimed at developing nonclassical platinum complexes that operate via mechanisms of action distinct from those of the approved drugs as mentioned in this paper.
Abstract: The platinum drugs, cisplatin, carboplatin, and oxaliplatin, prevail in the treatment of cancer, but new platinum agents have been very slow to enter the clinic. Recently, however, there has been a surge of activity, based on a great deal of mechanistic information, aimed at developing nonclassical platinum complexes that operate via mechanisms of action distinct from those of the approved drugs. The use of nanodelivery devices has also grown, and many different strategies have been explored to incorporate platinum warheads into nanomedicine constructs. In this Review, we discuss these efforts to create the next generation of platinum anticancer drugs. The introduction provides the reader with a brief overview of the use, development, and mechanism of action of the approved platinum drugs to provide the context in which more recent research has flourished. We then describe approaches that explore nonclassical platinum(II) complexes with trans geometry or with a monofunctional coordination mode, polynuclea...
