Structure-based Design of Prefusion-stabilized SARS-CoV-2 Spikes

TL;DR: Over 100 structure-guided spike variants based upon a previously determined cryo-EM structure of the prefusion SARS-CoV-2 spike are designed and expressed, with the best variant, HexaPro, having six beneficial proline substitutions leading to ∼10-fold higher expression than its parental construct.

Hapalindole/Ambiguine Biogenesis Is Mediated by a Cope Rearrangement, C-C Bond-Forming Cascade.

TL;DR: It is demonstrated that FamD2 and FamC1 act in concert to form the tetracyclic core ring system of the hapalindoles from cis-indole isonitrile and geranyl pyrophosphate through a presumed biosynthetic Cope rearrangement and subsequent 6-exo-trig cyclization/electrophilic aromatic substitution reaction.
