Abstract: Effects of naphthaleneacetic acid (NAA), aminoethoxyvinylglycine (AVG), and sprayable 1-methylcyclopropene (1-MCP) alone or in combination on fruit ethylene production, preharvest fruit drop, fruit quality, and fruit maturation were examined in 'Delicious' apples (Malus ·domestica Borkh.). 1-MCP and AVG + NAA, when applied 15 days before anticipated harvest (DBAH) for untreated control trees, more effectively delayed preharvest fruit drop than AVG or NAA used alone. However, there was no significant difference in ethylene production between fruit treated with 1-MCP or AVG + NAA and those treated by AVG. Two applications of NAA increased fruit ethylene pro- duction and fruit softening, whereas AVG inhibited NAA-enhanced fruit ethylene production and fruit softening. There was no significant difference in fruit ethylene pro- duction, fruit firmness, and fruit drop control between one and two applications of 1-MCP. The concentrations of 1-MCP did not affect the efficacy of 1-MCP when applied 15 DBAH, but high concentration of 1-MCP more effectively delayed preharvest fruit drop than low concentration of 1-MCP when applied 7 DBAH. Both AVG and 1-MCP suppressed expression of 1-aminocyclopropane-1-carboxylate (ACC) synthase gene MdACS1, ACC oxidase gene MdACO1, and polygalacturonase gene MdPG1 in fruit. Ex- pression ofACS5A andMdACO1 but notMdACS1 in fruit abscission zones was decreased by AVG and 1-MCP. 1-MCP more effectively suppressed expression of MdPG2 in fruit abscission zones than AVG alone.

Abstract: The main objective of this study was to investigate the effectiveness of preharvest 1-methylcyclopropene (1-MCP) treatment on the development of soft scald in 'Honeycrisp' apples. In addition, the effects of preharvest 1-MCP on fruit quality at harvest and after storage were examined. For two consecutive years of study, 'Honey- crisp' trees were sprayed preharvest with 1-MCP and fruit were harvested twice during each year. Preharvest 1-MCP treatments had little consistent effect on fruit maturity at the time of harvest. In both years of study, preharvest 1-MCP reduced the incidence of soft scald in 'Honeycrisp' apples after air storage at 0 or 3 8C for 5 or 6 months. Soggy breakdown developed only in the second year of study and high incidences were reduced by preharvest 1-MCP treatments. Preharvest 1-MCP often reduced flesh firmness loss in 'Honeycrisp' during storage, especially during the second year of study, and with 1-MCP application closer to harvest. Malic acid content was often higher in apples with the preharvest 1-MCP spray closer to harvest. Overall, the most important benefit of preharvest 1-MCP treatments on 'Honeycrisp' apples was the reduction in soft scald development. Due to the high potential for substantial fruit losses from this disorder, the use of preharvest 1-MCP sprays on 'Honeycrisp' apples could be very advantageous.

Abstract: The timing of plant organ abscission is modulated by the balance of two hormones, ethylene and auxin, while the mechanism of organ shedding depends on the loss of middle lamella pectin in the abscission zone (AZ). However, the mechanisms involved in sensing the balance of auxin and ethylene and that affect pectin degradation during abscission are not well understood. In this study, we identified two members of the APETALA2/ethylene-responsive factor (AP2/ERF) transcription factor family in rose (Rosa hybrida), RhERF1 and RhERF4 which play a role in petal abscission. The expression of RhERF1 and RhERF4 was influenced by ethylene and auxin, respectively. Reduced expression of RhERF1 or RhERF4 was observed to accelerate petal abscission. Global expression analysis and real-time PCR assays revealed that RhERF1 and RhERF4 modulate the expression of genes encoding pectin-metabolizing enzymes. A reduction in the abundance of pectin epitopes was detected in the AZs of RhERF1 and RhERF4-silenced plants by immunofluorescence microscopy analysis. In addition, RhERF1 and RhERF4 were shown to bind to the promoter of the pectin-metabolizing gene β-GALACTOSIDASE 1 (RhBGLA1), and reduced expression of RhBGLA1 delayed petal abscission. We conclude that during petal abscission, RhERF1 and RhERF4 integrate and coordinate ethylene and auxin signals to modulate pectin metabolism, in part by regulating the expression of RhBGLA1.

Abstract: Harvista™, a sprayable formulation of 1-methylcyclopropene (1-MCP), has recently been developed for preharvest use on horticultural products, whereas SmartFresh™ is a widely used 1-MCP treatment for products after harvest. The effects of Harvista™ on apple fruit ripening when sprayed at different maturities and on expression patterns of ethylene biosynthesis and receptor genes during storage have been investigated. Harvista™ applied to on-tree maturing apple fruit at an average starch pattern index of 2.5 resulted in a higher at-harvest firmness value compared with those treated at a starch pattern index of 1.5 and 3.5. This indicates that the timing of the Harvista™ application is critical. An application of Harvista™ led to better postharvest fruit firmness retention as well as reduced ethylene production. In addition, both preharvest and postharvest 1-MCP treatments resulted in contrasting responses in the expression patterns of two ethylene biosynthesis genes and in differentially suppressing effects on four ethylene receptor genes. Furthermore, the combined application of Harvista™ + SmartFresh™ resulted in greater fruit firmness retention and longer ethylene suppression. The expression profiles of these genes during on-tree fruit maturation prior to Harvista™ application were also characterized. Different regulation patterns of receptor genes could contribute to differential effects by 1-MCP treatments. The potential roles of Harvista™ to manipulate the ripening process as well as the molecular mechanism influencing 1-MCP treatment efficacy are discussed.
