Abstract: A vegetarian diet has been demonstrated to have a profound influence on human metabolism as well as to aid the prevention of several chronic diseases relative to an omnivorous diet. However, there have been no systematic metabolomic studies on all of the biochemical changes induced in human subjects by long-term vegetarianism. In this study, (1)H NMR spectroscopy in combination with multivariate statistical analysis was applied to explore the variability in the metabolic urinary profiles of healthy populations from four groups: lactovegetarian male (VEGMALE), lactovegetarian female (VEGFEMALE), omnivorous male (OMNMALE), and omnivorous female (OMNFEMALE). Differences in metabolic profiles were examined in relation to diet and gender by principal component analysis (PCA) and spectral integrals. It was found that the most influential low molecular weight metabolites responsible for the differences between the diet groups were N-acetyl glycoprotein (NAG), succinate, citrate, trimethylamine-N-oxide (TMAO), taurine, glycine, hippurate, phenylalanine, methylhistidine and formate, whereas for the differences in gender groups the most discriminatory metabolites were NAG, succinate, creatinine, arginine, TMAO, taurine, hippurate, mannitol, phenylalanine, and methylhistidine. The results from the PCA of all four groups indicated that diet plays a greater role in influencing metabolite differences than gender. As an exploration, this work shows the potential of metabolomics when applied to nutritional and physiological studies, and it will aid further studies.

Abstract: Low-resolution images may be acquired in magnetic resonance imaging (MRI) due to limited data acquisition time or other physical constraints, and their resolutions can be improved with super-resolution methods. Since MRI can offer images of an object with different contrasts, e.g., T1-weighted or T2-weighted, the shared information between inter-contrast images can be used to benefit super-resolution. In this study, an MRI image super-resolution approach to enhance in-plane resolution is proposed by exploring the statistical information estimated from another contrast MRI image that shares similar anatomical structures. We assume some edge structures are shown both in T1-weighted and T2-weighted MRI brain images acquired of the same subject, and the proposed approach aims to recover such kind of structures to generate a high-resolution image from its low-resolution counterpart. The statistical information produces a local weight of image that are found to be nearly invariant to the image contrast and thus this weight can be used to transfer the shared information from one contrast to another. We analyze this property with comprehensive mathematics as well as numerical experiments. Experimental results demonstrate that the image quality of low-resolution images can be remarkably improved with the proposed method if this weight is borrowed from a high resolution image with another contrast. Abstract: Taurine is an indispensable amino acid for many fish species and taurine supplementation is needed when plant-based diets are used as the primary protein source for these species. However, there is limited information available to understand the physiological or metabolic effects of taurine on fish. In this study, 1H nuclear magnetic resonance (NMR)-based metabolomic analysis was conducted to identify the metabolic profile change in the fish intestine with the aim to assess the effect of dietary taurine supplementation on the physiological and metabolomic variation of fish, and reveal the possible mechanism of taurine's metabolic effect. Grouper (Epinephelus coioides) were divided into four groups and fed diets containing 0.0%, 0.5%, 1.0%, and 1.5% taurine supplementation for 84 days. After extraction using aqueous and organic solvents, 25 significant taurine-induced metabolic changes were identified. These metabolic changes in grouper intestine were characterized by differences in carbohydrate, amino acid, lipid and nucleotide. The results reflected both the physiological state and growth of the fish, and indicated that taurine supplementation significantly affects the metabolome of fish, improves energy utilization and amino acid uptake, promotes protein, lipid and purine synthesis, and accelerates fish growth.

Abstract: The trichothecene mycotoxin deoxynivalenol (DON) is produced in wheat, barley and corn following infestation by the fungus Fusarium in the field and during storage. Colloquially known as “vomitoxin” because of its emetic effects in pigs, DON has been associated with human gastroenteritis. Since DON is commonly detected in cereal foods, there are significant questions regarding the risks of acute poisoning and chronic effects posed to persons ingesting this trichothecene. A further challenge is how to best manage perceived risks without rendering critical food staples unavailable to an ever-expanding world population. In experimental animal models, acute DON poisoning causes emesis, whereas chronic low-dose exposure elicits anorexia, growth retardation, immunotoxicity as well as impaired reproduction and development resulting from maternal toxicity. Pathophysiologic effects associated with DON include altered neuroendocrine signaling, proinflammatory gene induction, disruption of the growth hormone axis, and altered gut integrity. At the cellular level, DON induces ribotoxic stress thereby disrupting macromolecule synthesis, cell signaling, differentiation, proliferation, and death. There is a need to better understand the mechanistic linkages between these early dose-dependent molecular effects and relevant pathological sequelae. Epidemiological studies are needed to determine if relationships exist between consumption of high DON levels and incidence of both gastroenteritis and potential chronic diseases. From the perspective of human health translation, a particularly exciting development is the availability of biomarkers of exposure (e.g. DON glucuronide) and effect (e.g. IGF1) now make it possible to study the relationship between DON consumption and growth retardation in susceptible human populations such as children and vegetarians. Ultimately, a fusion of basic and translational research is needed to validate or refine existing risk assessments and regulatory standards for this common mycotoxin.

Abstract: Objective The consumption of an agrarian diet is associated with a reduced risk for many diseases associated with a ‘Westernised’ lifestyle. Studies suggest that diet affects the gut microbiota, which subsequently influences the metabolome, thereby connecting diet, microbiota and health. However, the degree to which diet influences the composition of the gut microbiota is controversial. Murine models and studies comparing the gut microbiota in humans residing in agrarian versus Western societies suggest that the influence is large. To separate global environmental influences from dietary influences, we characterised the gut microbiota and the host metabolome of individuals consuming an agrarian diet in Western society. Design and results Using 16S rRNA-tagged sequencing as well as plasma and urinary metabolomic platforms, we compared measures of dietary intake, gut microbiota composition and the plasma metabolome between healthy human vegans and omnivores, sampled in an urban USA environment. Plasma metabolome of vegans differed markedly from omnivores but the gut microbiota was surprisingly similar. Unlike prior studies of individuals living in agrarian societies, higher consumption of fermentable substrate in vegans was not associated with higher levels of faecal short chain fatty acids, a finding confirmed in a 10-day controlled feeding experiment. Similarly, the proportion of vegans capable of producing equol, a soy-based gut microbiota metabolite, was less than that was reported in Asian societies despite the high consumption of soy-based products. Conclusions Evidently, residence in globally distinct societies helps determine the composition of the gut microbiota that, in turn, influences the production of diet-dependent gut microbial metabolites.
