In complex media such as white paint and biological tissue, light encounters nanoscale refractive-index inhomogeneities that cause multiple scattering. Such scattering is usually seen as an impediment to focusing and imaging. However, scientists have recently used strongly scattering materials to focus, shape and compress waves by controlling the many degrees of freedom in the incident waves. This was first demonstrated in the acoustic and microwave domains using time reversal, and is now being performed in the optical realm using spatial light modulators to address the many thousands of spatial degrees of freedom of light. This approach is being used to investigate phenomena such as optical super-resolution and the time reversal of light, thus opening many new avenues for imaging and focusing in turbid media

Controlling waves in space and time for imaging and focusing in complex media

Biological specimens are rife with optical inhomogeneities that seriously degrade imaging performance under all but the most ideal conditions. Measuring and then correcting for these inhomogeneities is the province of adaptive optics. Here we introduce an approach to adaptive optics in microscopy wherein the rear pupil of an objective lens is segmented into subregions, and light is directed individually to each subregion to measure, by image shift, the deflection faced by each group of rays as they emerge from the objective and travel through the specimen toward the focus. Applying our method to two-photon microscopy, we could recover near-diffraction-limited performance from a variety of biological and nonbiological samples exhibiting aberrations large or small and smoothly varying or abruptly changing. In particular, results from fixed mouse cortical slices illustrate our ability to improve signal and resolution to depths of 400 microm.

Adaptive optics via pupil segmentation for high-resolution imaging in biological tissues

In this paper, we present relay-assisted transmission as a powerful fading mitigation tool for free-space optical systems operating in atmospheric turbulence channels. We study both serial (i.e., multi-hop transmission) and parallel (i.e., cooperative diversity) relaying encoupled with amplify-and-forward and decode-and-forward modes. We consider an aggregated channel model which takes into account both path-loss and turbulence-induced log-normal fading. Since fading variance is distance-dependent in free-space optical systems, relay-assisted transmission takes advantage of the resulting shorter hops and yields significant performance improvements. We derive outage probability of the relaying schemes under consideration which are further confirmed through Monte-Carlo simulations. Our outage probability analysis demonstrates that an impressive performance improvement of 18.5 dB is possible with the use of a single relay at a target outage probability of 10-6.

Relay-Assisted Free-Space Optical Communication

In the field of biomedical optics, optical scattering has traditionally limited the range of imaging within tissue to a depth of one millimetre. A recently developed class of wavefront-shaping techniques now aims to overcome this limit and achieve diffraction-limited control of light beyond one centimetre. By manipulating the spatial profile of an optical field before it enters a scattering medium, it is possible to create a micrometre-scale focal spot deep within tissue. To successfully operate in vivo, these wavefront-shaping techniques typically require feedback from within the biological sample. This Review summarizes recently developed 'guidestar' mechanisms that provide feedback for intra-tissue focusing. Potential applications of guidestar-assisted focusing include optogenetic control over neurons, targeted photodynamic therapy and deep tissue imaging.

Guidestar-assisted wavefront-shaping methods for focusing light into biological tissue

The main advantage of confocal microscopes over their conventional counterparts is their ability to optically “section” thick specimens; the thin image slices thus obtained can be used to reconstruct three-dimensional images, a capability which is particularly useful in biological applications. However, it is well known that the resolution and optical sectioning ability can be severely degraded by system or specimen-induced aberrations. The use of high aperture lenses further exacerbates the problem. Moreover, aberrations can considerably reduce the number of photons that reach the detector, leading to lower contrast. It is rather unfortunate, therefore, that in practical microscopy, aberration-free confocal imaging is rarely achieved. Adaptive optics systems, which have been used widely to correct aberrations in astronomy, offer a solution here but also present new challenges. The optical system and the source of aberrations in a confocal microscope are considerably different and require a novel approach to wavefront sensing. This method, based upon direct measurement of Zernike aberration modes, also exhibits an axial selectivity similar to that of a confocal microscope. We demonstrate an adaptive confocal fluorescence microscope incorporating this modal sensor together with a deformable membrane mirror for aberration correction. Aberration corrected images of biological specimens show considerable improvement in contrast and apparent restoration of axial resolution.
