Evidence for an alternative fatty acid desaturation pathway increasing cancer plasticity
Kim Vriens,Stefan Christen,Sweta Parik,Dorien Broekaert,Kazuaki Yoshinaga,Ali Talebi,Jonas Dehairs,Carmen Escalona-Noguero,Roberta Schmieder,Thomas Cornfield,Catriona Charlton,Laura Romero-Pérez,Matteo Rossi,Gianmarco Rinaldi,Martin F. Orth,Ruben Boon,Axelle Kerstens,Suet Ying Kwan,Brandon Faubert,Andrés Méndez-Lucas,Charlotte Kopitz,Ting Chen,Juan Fernández-García,Joao A.G. Duarte,Arndt Schmitz,Patrick Steigemann,Mustapha Najimi,Andrea Hägebarth,Jo A. Van Ginderachter,Etienne Sokal,Naohiro Gotoh,Kwok-Kin Wong,Catherine M. Verfaillie,Rita Derua,Sebastian Munck,Mariia Yuneva,Laura Beretta,Ralph J. DeBerardinis,Johannes V. Swinnen,Leanne Hodson,David Cassiman,Chris Verslype,Sven Christian,Sylvia Grünewald,Thomas G. P. Grunewald,Sarah-Maria Fendt +45 more
TL;DR: In several human and mouse cancer cell lines and carcinomas, a sapienate biosynthesis pathway underpins metabolic plasticity by allowing these cells to bypass stearoyl-CoA desaturase-dependent fatty acid desaturation.
Journal ArticleDOI
Lipids and cancer: Emerging roles in pathogenesis, diagnosis and therapeutic intervention
Lisa M. Butler,Ylenia Perone,Jonas Dehairs,Leslie E. Lupien,Vincent de Laat,Ali Talebi,Massimo Loda,William B. Kinlaw,Johannes V. Swinnen +8 more
TL;DR: Pre-clinical studies illustrate the translational promise of exploiting lipid metabolism in cancer, and critically, have shown context dependent actionable vulnerabilities that can be rationally targeted, particularly in combinatorial approaches.
Journal ArticleDOI
CRISP-ID: decoding CRISPR mediated indels by Sanger sequencing
TL;DR: CRISP-ID is presented, a web application which uses a unique algorithm for genotyping up to three alleles from a single Sanger sequencing trace, providing a robust and readily accessible platform to directly identify indels and significantly speed up the characterization of clones.
Journal ArticleDOI
Non-small cell lung cancer is characterized by dramatic changes in phospholipid profiles
Eyra Marien,Michael Meister,Thomas Muley,Steffen Fieuws,Sergio Bordel,Rita Derua,Jeffrey M. Spraggins,Raf Van de Plas,Raf Van de Plas,Jonas Dehairs,Jens Wouters,Muralidhararao Bagadi,Hendrik Dienemann,Michael Thomas,Philipp A. Schnabel,Richard M. Caprioli,Etienne Waelkens,Johannes V. Swinnen +17 more
TL;DR: Using both shotgun and 2D‐imaging lipidomics analysis, a hitherto unrecognized alteration in phospholipid profiles in NSCLC is uncovered, which may have important biological implications and may have significant potential for biomarker development.
Journal ArticleDOI
Loss of Chromosome 8p Governs Tumor Progression and Drug Response by Altering Lipid Metabolism
Yanyan Cai,Jonathan Crowther,Tibor J. Pastor,Layka Abbasi Asbagh,Maria Francesca Baietti,Magdalena De Troyer,Iria Vazquez,Ali Talebi,Fabrizio Renzi,Jonas Dehairs,Johannes V. Swinnen,Anna Sablina +11 more
TL;DR: To delineate the functional outcome of chromosome 8p loss of heterozygosity (LOH), a common aberration in breast cancer, this work modeled 8p LOH using TALEN-based genomic engineering and found the shift in lipid metabolism triggers invasiveness and confers tumor growth under stress conditions due to increased autophagy.
