Abstract: Rolliniamucosa (Annonaceae), known in primitive medical practices of Indonesia and the West Indies as a treatment for tumors, has been investigated employing the seeds from a South American variety and the murine P388 lymphocytic leukemia (PS) for bioassay. The principal antineoplastic constituent (PS, 28% life extension, at 0.25 mg/kg and ED504.5 × 10−5 μg/mL) was found to be a new bis-tetrahydrofuran designated rolliniastatin 1. Structural elucidation of rolliniastatin 1 was accomplished by a combination of high resolution nuclear magnetic resonance (300 MHz), mass spectral, and X-ray crystal structure techniques.

Abstract: A structure/activity study of selected limonoids from two plant families (Meliaceae and Rutaceae) of the Rutales order against the murine P-388 lymphocytic leukemia system was undertaken. The presence of both a 19 leads to 28 lactol and a 14,15 beta-epoxide group was found especially important for pronounced inhibition of the PS in vitro cell line. Substitution of an A-ring alpha,beta-unsaturated ketone (3-oxo-1-ene) for the lactol led to diminished activity, while reduction of the olefin caused complete loss of activity. At the dose levels employed, even very good PS in vitro inhibition was not translated into PS in vivo antineoplastic effects.

Abstract: 2 new limonoid-type terpenes have been isolated from an aqueous extract of seeds produced by the Eastern Himalayan (India) plant Aphanamixis grandifolia B1. By interpreting principally mass spectral and nuclear magnetic resonance data, the structures of 12-hydroxyamoorastatin (2b) and amoorastatone (3) were elucidated. Unequivocal evidence for the 12-hydroxyamoorastatin structural assignment was obtained by chemical conversion to sendanin (4). Amoorastatin derivative 2b was found to significantly inhibit growth of the murine P388 lymphocytic leukemia cell lines but amoorastatone in the same system was inactive. In a comparative biological study, sendanin (4) and anthothecol (7) were also found significantly to inhibit growth of the P388 cell line, while rohitukin (8) and limonin (9) were found to be inactive.

Abstract: A new cell growth inhibitory acetogenin, designated rolliniastatin 2, has been isolated from seeds of the South American Rollinia mucosa (Annonaceae) and its structure determined. A diastereomeric relationship for rolliniastatin 1, rolliniastatin 2, and asimicin was proposed. The Rollinia genus (Annonaceae) is composed of some 65 species occurring principally from Central America to Argentina. Rollinia, and Uvaria genera have been found to contain members of a new class of biologically active bis- tetrahydrofurans. Our crystal smcture determination of rolliniastatin 12 provided the first definitive smcture in this new series.3-10 By x-ray crystallography, the relative stereochemistry of rolliniastatin 1 was established as 4S*, 15S*, 16S*, 19R*, 20R*, 23S*, 24R*, and 36R*.2 Asimicin, recently isolated from Asimina triloba,8 apparently differs from rolliniastatin 1 in its configuration at one or more of the chiral centers associated with the bis-tetrahydrofuran system. Further detailed fractionation of a hexane extract of Rolliniu mucosa seeds' has led us to uncover companions of rolliniastatin 1, including asimicin and a new diastereomer of rolliniastatin 1, named rolliniastatin 2. Isolation of rolliniastatin 2 flash chromatography on silica gel, high-speed countercurrent distribution (J!ISCCD)11 with a multilayer coil planet centrifuge,lz column chromatography (LiChrom Sia), and finally hplc (semipreparative with Partisil 10 silica gel followed by analytical with Partisil 5 silica gel) with the solvent system hexane - ethyl ether - methanol. The structure assigned by spectral techniques was confirmed by synthesis.
