Apoptotic Caspases Suppress mtDNA-Induced STING-Mediated Type I IFN Production
Michael J. White,Michael J. White,Kate McArthur,Kate McArthur,Donald Metcalf,Donald Metcalf,Rachael M. Lane,John C. Cambier,Marco J Herold,Marco J Herold,Mark F. van Delft,Mark F. van Delft,Sammy Bedoui,Guillaume Lessene,Guillaume Lessene,Matthew E. Ritchie,Matthew E. Ritchie,David C.S. Huang,David C.S. Huang,Benjamin T. Kile,Benjamin T. Kile +20 more
TL;DR: The apoptotic caspase cascade functions to render mitochondrial apoptosis immunologically silent, and suppresses type I interferon (IFN) production by cells undergoing Bak/Bax-mediated apoptosis.
Journal ArticleDOI
BAK/BAX macropores facilitate mitochondrial herniation and mtDNA efflux during apoptosis
Kate McArthur,Kate McArthur,Kate McArthur,Lachlan Whitehead,Lachlan Whitehead,John M. Heddleston,Lucy Li,Benjamin S. Padman,Viola Oorschot,Niall D. Geoghegan,Niall D. Geoghegan,Stephane Chappaz,Stephane Chappaz,Stephane Chappaz,Sophia Davidson,Hui San Chin,Rachael M. Lane,Marija Dramicanin,Marija Dramicanin,Tahnee L. Saunders,Canny Sugiana,Romina Lessene,Romina Lessene,Laura D. Osellame,Teng Leong Chew,Grant Dewson,Grant Dewson,Michael Lazarou,Georg Ramm,Guillaume Lessene,Guillaume Lessene,Michael T. Ryan,Kelly L. Rogers,Kelly L. Rogers,Mark F. van Delft,Mark F. van Delft,Benjamin T. Kile,Benjamin T. Kile,Benjamin T. Kile +38 more
TL;DR: This study provides a mechanistic description of mtDNA release from mitochondria during apoptosis, and suggests that mtDNA is found outside the mitochondria—and, indeed, outside the cell—in a wide range of circumstances.
Journal ArticleDOI
TDP-43 Triggers Mitochondrial DNA Release via mPTP to Activate cGAS/STING in ALS
Chien-Hsiung Yu,Chien-Hsiung Yu,Sophia Davidson,Sophia Davidson,Cassandra R. Harapas,Cassandra R. Harapas,James B. Hilton,James B. Hilton,Michael J. Mlodzianoski,Michael J. Mlodzianoski,Pawat Laohamonthonkul,Pawat Laohamonthonkul,Cynthia Louis,Cynthia Louis,Ronnie Ren Jie Low,Ronnie Ren Jie Low,Jonas Moecking,Jonas Moecking,Jonas Moecking,Dominic De Nardo,Dominic De Nardo,Dominic De Nardo,Katherine R. Balka,Katherine R. Balka,Katherine R. Balka,Dale J. Calleja,Dale J. Calleja,Fiona Moghaddas,Fiona Moghaddas,Fiona Moghaddas,Erya Ni,Erya Ni,Catriona McLean,Andre L. Samson,Andre L. Rogers,Kelly L. Rogers,Kate McArthur,Peter J. Crouch,Peter J. Crouch,Seth L. Masters,Seth L. Masters +51 more
TL;DR: The results identify mtDNA release and cGAS/STING activation as critical determinants of TDP-43-associated pathology and demonstrate the potential for targeting this pathway in ALS.
Journal ArticleDOI
NLRP1 Inflammasome Activation Induces Pyroptosis of Hematopoietic Progenitor Cells
Seth L. Masters,Motti Gerlic,Motti Gerlic,Donald Metcalf,Donald Metcalf,Simon Preston,Simon Preston,Marc Pellegrini,Marc Pellegrini,Joanne A. O’Donnell,Joanne A. O’Donnell,Kate McArthur,Kate McArthur,Tracey M. Baldwin,Stéphane Chevrier,Cameron J. Nowell,Louise H. Cengia,Katya J. Henley,Janelle E. Collinge,Daniel L. Kastner,Lionel Feigenbaum,Douglas J. Hilton,Douglas J. Hilton,Warren S. Alexander,Warren S. Alexander,Benjamin T. Kile,Benjamin T. Kile,Ben A. Croker,Ben A. Croker +28 more
TL;DR: It is shown that NLRP1 acts as a cellular sentinel to alert Caspase-1 to hematopoietic and infectious stress and enhanced recovery from chemotherapy and LCMV infection is shown.
Journal ArticleDOI
The Mitochondrial Apoptotic Effectors BAX/BAK Activate Caspase-3 and -7 to Trigger NLRP3 Inflammasome and Caspase-8 Driven IL-1β Activation
James E Vince,James E Vince,Dominic De Nardo,Dominic De Nardo,Wenqing Gao,Angelina J. Vince,Cathrine Hall,Kate McArthur,Kate McArthur,Kate McArthur,Daniel S Simpson,Daniel S Simpson,Swarna L Vijayaraj,Swarna L Vijayaraj,Lisa M Lindqvist,Lisa M Lindqvist,Philippe Bouillet,Philippe Bouillet,Mark A. Rizzacasa,Si Ming Man,John Silke,John Silke,Seth L. Masters,Seth L. Masters,Guillaume Lessene,Guillaume Lessene,David C.S. Huang,David C.S. Huang,Daniel H.D. Gray,Daniel H.D. Gray,Benjamin T. Kile,Benjamin T. Kile,Benjamin T. Kile,Feng Shao,Kate E. Lawlor,Kate E. Lawlor +35 more
TL;DR: It is shown that innate immune cells undergoing BAX/BAK-mediated apoptosis have the capacity to generate pro-inflammatory signals and provide an explanation as to why IL-1β activation is often associated with cellular stress, such as during chemotherapy.
