Journal ArticleDOI
Single-molecule visualization of DNA G-quadruplex formation in live cells
Marco Di Antonio,Marco Di Antonio,Aleks Ponjavic,Aleks Ponjavic,Antanas Radzevičius,Rohan T. Ranasinghe,Marco Catalano,Xiaoyun Zhang,Jiazhen Shen,Lisa-Maria Needham,Steven F. Lee,David Klenerman,Shankar Balasubramanian +12 more
TL;DR: A G4-specific fluorescent probe (SiR-PyPDS) has been developed that enables single-molecule and real-time detection of individual G4 structures in living cells without perturbing G4 formation and dynamics.
Journal ArticleDOI
Poly(ADP-Ribose) Links the DNA Damage Response and Biomineralization
Karin H. Müller,Robert Hayward,Rakesh Rajan,Meredith Whitehead,Andrew M. Cobb,Sadia Ahmad,Mengxi Sun,Ieva Goldberga,Rui Li,Uliana Bashtanova,Anna M. Puszkarska,David G. Reid,Roger A. Brooks,Jeremy N. Skepper,Jayanta Bordoloi,Wing Ying Chow,Hartmut Oschkinat,Alex Groombridge,Oren A. Scherman,James A. Harrison,Anja Verhulst,Patrick C. D'Haese,Ellen Neven,Lisa-Maria Needham,Steven F. Lee,Catherine M. Shanahan,Melinda J. Duer +26 more
TL;DR: It is concluded that poly(ADP-ribose) plays a central chemical role in both pathological and physiological extracellular matrix calcification, suggesting a therapeutic route for the treatment of vascular calcifications.
Journal ArticleDOI
Spectrally Resolved Photodynamics of Individual Emitters in Large-Area Monolayers of Hexagonal Boron Nitride.
Hannah L. Stern,Ruizhi Wang,Ye Fan,Ryo Mizuta,James C. Stewart,Lisa-Maria Needham,Trevor D. Roberts,Rebecca B. Wai,Naomi S. Ginsberg,David Klenerman,Stephan Hofmann,Steven F. Lee +11 more
TL;DR: These results show that high control over h-BN growth and treatment can produce a narrow distribution of emitter type and that surface interactions heavily influence the photodynamics, and highlight the utility of spectrally resolved wide-field microscopy in the study of optically active excitations in atomically thin two-dimensional materials.
Journal ArticleDOI
Rational design of a conformation-specific antibody for the quantification of Aβ oligomers
Francesco A. Aprile,Pietro Sormanni,Marina Podpolny,Shianne Chhangur,Lisa-Maria Needham,Francesco Simone Ruggeri,Michele Perni,Ryan Limbocker,Gabriella T. Heller,Tomas Sneideris,Tom Scheidt,Benedetta Mannini,Johnny Habchi,Steven F. Lee,Patricia C. Salinas,Tuomas P. J. Knowles,Christopher M. Dobson,Michele Vendruscolo +17 more
TL;DR: A two-step rational design method for the discovery of oligomer-specific antibodies that enables the accurate detection and quantification of Aβ oligomers in vitro, and in Caenorhabditis elegans and mouse hippocampal tissues.
Journal ArticleDOI
ThX – a next-generation probe for the early detection of amyloid aggregates
Lisa-Maria Needham,Judith Weber,Juan A. Varela,James W. B. Fyfe,Dung T. Do,Catherine K. Xu,Luke Tutton,Rachel Cliffe,Benjamin Keenlyside,David Klenerman,Christopher M. Dobson,Christopher A. Hunter,Karin H. Müller,Kevin O'Holleran,Sarah E. Bohndiek,Thomas N. Snaddon,Steven F. Lee +16 more
TL;DR: Thioflavin X, (ThX), a next-generation fluorescent probe which displays superior properties; including a 5-fold increase in brightness and 7-fold increased in binding affinity to amyloidogenic proteins, is designed.
