Journal ArticleDOI
Epigenetic differences arise during the lifetime of monozygotic twins
Mario F. Fraga,Esteban Ballestar,Maria F. Paz,Santiago Ropero,Fernando Setien,Maria Luisa Ballestar,Damia Heine-Suñer,Juan C. Cigudosa,Miguel Urioste,Javier Benitez,Manuel Boix-Chornet,Abel Sánchez-Aguilera,Charlotte Ling,Emma Carlsson,Pernille Poulsen,Allan Vaag,Zarko Stephan,Tim D. Spector,Yue Zhong Wu,Christoph Plass,Manel Esteller +20 more
TL;DR: Older monozygous twins exhibited remarkable differences in their overall content and genomic distribution of 5-methylcytosine DNA and histone acetylation, affecting their gene-expression portrait, indicating how an appreciation of epigenetics is missing from the understanding of how different phenotypes can be originated from the same genotype.
Journal ArticleDOI
Loss of acetylation at Lys16 and trimethylation at Lys20 of histone H4 is a common hallmark of human cancer
Mario F. Fraga,Esteban Ballestar,Ana Villar-Garea,Manuel Boix-Chornet,Jesús Espada,Gunnar Schotta,Tiziana Bonaldi,Claire E. Haydon,Santiago Ropero,Kevin Petrie,N. Gopalakrishna Iyer,Alberto Pérez-Rosado,Enrique Calvo,Juan Antonio López,Amparo Cano,María José Calasanz,Dolors Colomer,Miguel A. Piris,Natalie G. Ahn,Natalie G. Ahn,Axel Imhof,Carlos Caldas,Thomas Jenuwein,Manel Esteller +23 more
TL;DR: It is found that cancer cells had a loss of monoacetylated and trimethylated forms of histone H4 early and accumulated during the tumorigenic process, which is a common hallmark of human tumor cells.
Journal ArticleDOI
A truncating mutation of HDAC2 in human cancers confers resistance to histone deacetylase inhibition
Santiago Ropero,Mario F. Fraga,Esteban Ballestar,Richard Hamelin,Hiroyuki Yamamoto,Manuel Boix-Chornet,Rosalia Caballero,Miguel Alaminos,Fernando Setien,Maria F. Paz,Michel Herranz,José Palacios,Diego Arango,Torben F. Ørntoft,Lauri A. Aaltonen,Simó Schwartz,Manel Esteller +16 more
TL;DR: The identified truncating mutations in one of the primary human histone deacetylases, HDAC2, are identified in sporadic carcinomas with microsatellite instability and in tumors arising in individuals with hereditary nonpolyposis colorectal cancer syndrome and support the use ofHDAC2 mutational status in future pharmacogenetic treatment of these individuals.
Journal ArticleDOI
DNA methylation-independent loss of RARA gene expression in acute myeloid leukemia
Annegret Glasow,Annegret Glasow,Angela N. Barrett,Kevin Petrie,Rajeev Gupta,Manuel Boix-Chornet,Da Cheng Zhou,David Grimwade,Robert E. Gallagher,Marieke von Lindern,Samuel Waxman,Tariq Enver,Guido Hildebrandt,Arthur Zelent +13 more
TL;DR: It is demonstrated that relative to normal myeloid stem/progenitor cells, RARalpha2 expression is dramatically reduced in primary AML blasts and the notion that diminished RARA expression contributes to leukemogenesis is supported.
Journal ArticleDOI
Combinatorial effects of splice variants modulate function of Aiolos
Rosalia Caballero,Fernando Setien,Lidia Lopez-Serra,Manuel Boix-Chornet,Mario F. Fraga,Santiago Ropero,Diego Megías,Miguel Alaminos,Eva María Sánchez-Tapia,María C. Montoya,Manel Esteller,Rogelio González-Sarmiento,Esteban Ballestar +12 more
TL;DR: It is demonstrated that the cellular activities of Aiolos isoforms are dependent on combinations of various functional domains arising from the differential splicing of mRNA transcripts, and these data support the general principle that the function of an individual protein is modulated through alternative splicing.
