Journal ArticleDOI
Delineating the role of cooperativity in the design of potent PROTACs for BTK
Adelajda Zorba,Chuong Nguyen,Yingrong Xu,Jeremy T. Starr,Kris A. Borzilleri,James F. Smith,Hongyao Zhu,Kathleen A. Farley,WeiDong Ding,James Schiemer,Xidong Feng,Jeanne S. Chang,Daniel P. Uccello,Jennifer A. Young,Carmen N. Garcia-Irrizary,Lara C. Czabaniuk,Brandon P. Schuff,Robert M. Oliver,Justin I. Montgomery,Matthew Merrill Hayward,Jotham Wadsworth Coe,Jinshan Chen,Mark Niosi,Suman Luthra,Jaymin C. Shah,Ayman El-Kattan,Xiayang Qiu,Graham M. West,Mark C. Noe,Veerabahu Shanmugasundaram,Adam M. Gilbert,Matthew Frank Brown,Matthew F. Calabrese +32 more
TL;DR: It is shown that within a Bruton’s tyrosine kinase/cereblon PROTAC system, potent knockdown correlates with alleviation of steric clashes in the absence of thermodynamic cooperativity, which broadens the scope of PROTAC applications and affects fundamental design criteria across the field.
Journal ArticleDOI
Chemical and Computational Methods for the Characterization of Covalent Reactive Groups for the Prospective Design of Irreversible Inhibitors
Mark Edward Flanagan,Joseph A. Abramite,Dennis P. Anderson,Ann Aulabaugh,Upendra P. Dahal,Adam M. Gilbert,Chao Li,Justin I. Montgomery,Stacey R. Oppenheimer,Tim F. Ryder,Brandon P. Schuff,Daniel P. Uccello,Gregory S. Walker,Yan Wu,Matthew Frank Brown,Jinshan M. Chen,Matthew Merrill Hayward,Mark C. Noe,R. Scott Obach,Laurence Philippe,Veerabahu Shanmugasundaram,Michael J. Shapiro,Jeremy T. Starr,Justin G. Stroh,Ye Che +24 more
TL;DR: Methods for measuring the reactivity of covalent reactive groups (CRGs) with a biologically relevant nucleophile, glutathione (GSH), along with kinetic data for a broad array of electrophiles are described.
Journal ArticleDOI
Highly enantioselective synthesis of cyclic and functionalized α-amino acids by means of a chiral phase transfer catalyst
E. J. Corey,Mark C. Noe,Feng Xu +2 more
TL;DR: The chiral quaternary ammonium salt as mentioned in this paper serves as phase transfer catalyst for the enantioselective conversion of the glycine derivative 2 to a variety of cyclic and acyclic chiral α-amino acids with enanti-lectivities as high as 200:1 in alkylation and Michael addition reactions.

Pharmacological Characterization of CP-547,632, a Novel Vascular Endothelial Growth Factor Receptor-2 Tyrosine Kinase Inhibitor for Cancer Therapy
Jean S. Beebe,Jitesh P. Jani,Elisabeth Knauth,Peter Goodwin,Carla Higdon,Ann Marie Rossi,Erling Emerson,Martin Finkelstein,Eugenia Floyd,Shawn Harriman,Jim Atherton,Steve Hillerman,Cathy Soderstrom,Kou Kou,Tom Gant,Mark C. Noe,Barb Foster,Farzan Rastinejad,Matthew A. Marx,Tracey Lee Schaeffer,Pamela Whalen,W. Gregory Roberts +21 more
TL;DR: CP-547,632 is a well-tolerated, orally-bioavailable inhibitor presently under clinical investigation for the treatment of human malignancies and is selective relative to epidermal growth factor receptor, platelet-derived growth factor beta, and other related TKs.
