Abstract: Secreted protein acidic and rich in cysteine (SPARC) is a secreted glycoprotein protein which modulates cell shape and cell-matrix interactions and has been implicated in the regulation of angiogenesis, vascular permeability and cataract formation. In situ hybridization and immunohistochemical studies for SPARC were performed to determine the cell and tissue distribution of SPARC in rat eye. Studies demonstrated SPARC mRNA and protein co-localization at all sites. In the retina SPARC mRNA and protein were localized predominantly to the Muller and ganglion cells. Within the choroid, SPARC was found in vascular endothelial cells and fibroblasts; in the sclera SPARC was present in blood vessels and fibroblasts. SPARC was also present in the non-pigmented epithelial cells of the ciliary body, and in the epithelium of the lens capsule and cornea. The demonstrated anatomical distribution of SPARC in the rat eye is consistent with several of the biological functions ascribed to this matricellular protein and provides a rational basis for its examination in pathological conditions.

Abstract: The impact of under-acylation of lipid A on the interaction between Klebsiella pneumoniae LPS and polymyxins B and E was examined with fluorometric and calorimetric methods, and by (1)H NMR, using a paired wild type (WT) and the ΔlpxM mutant strains B5055 and B5055ΔlpxM, which predominantly express LPS with hexa- and penta-acylated lipid A structures respectively. LPS from B5055ΔlpxM displayed a fourfold increased binding affinity for polymyxins B and E compared with the B5055 WT LPS. EC50 values were consistent with polymyxin minimum inhibitory concentration (MIC) values for each strain. Accordingly, polymyxin exposure considerably enhanced the permeability of the B5055ΔlpxM OM. Analysis of the melting profiles of isolated LPS aggregates suggested that bactericidal polymyxin activity may relate to the acyl chains' phase of the outer membrane (OM). The enhanced polymyxin susceptibility of B5055ΔlpxM may be attributable to the favorable insertion of polymyxins into the more fluid OM compared with B5055. Molecular models of the polymyxin B-lipid A complex illuminate the key role of the lipid A acyl chains for complexation of polymyxin. The data provide important insight into the molecular basis for the increased polymyxin susceptibility of K. pneumoniae strains with under-acylated lipid A. Under-acylation appears to facilitate the integration of the N-terminal fatty-acyl chain of polymyxin into the OM resulting in an increased susceptibility to its antimicrobial activity/activities.

Abstract: A successful strategy for prediction of crop yield that accounts for the effects of genotype and environment will open up many opportunities for enhancing the productivity of agricultural systems. Crop growth models (CGMs) have a history of application for crop management decision support. Recently whole genome prediction (WGP) methodologies have been developed and applied in breeding to enable prediction of crop traits for new genotypes and thus increase the size of plant breeding programs without the need to expand expensive field testing. The presence of Genotype-by-Environment-by-Management (G×E×M) interactions for yield presents a significant challenge for the development of prediction technologies for both product development by breeding and product placement within agricultural production systems. The integration of a CGM into the algorithm for whole genome prediction WGP, referred to as CGM-WGP, has opened up the potential for prediction of G×E×M interactions for breeding and product placement applications. Here a combination of simulation and empirical studies are used to explain how the CGM-WGP methodology works and to demonstrate successful reduction to practice for applications to maize breeding and product placement recommendation in the US corn belt.

Abstract: Water availability can limit maize (Zea mays L.) yields, and root traits may enhance drought adaptation if they can moderate temporal patterns of soil water extraction to favour grain filling. Root system efficiency (RSE), defined as transpiration per unit leaf area per unit of root mass, represents the functional mass allocation to roots to support water capture relative to the allocation to aerial mass that determines water demand. The aims of this study were to identify the presence of hybrid variation for RSE in maize, determine plant attributes that drive these differences and illustrate possible links of RSE to drought adaptation via associations with water extraction patterns. Individual plants for a range of maize hybrids were grown in large containers in shadehouses in Queensland, Australia. Leaf area, shoot and root mass, transpiration, root distribution and soil water were measured in all or selected experiments. Significant hybrid differences in RSE existed. High RSE was associated with reduced dry mass allocation to roots and more efficient water capture per unit of root mass. It was also weakly negatively associated with total plant dry mass, reducing preanthesis water use. This could increase grain yield under drought. RSE provides a conceptual physiological framework to identify traits for high-throughput phenotyping in breeding programs.

Abstract: Two outlines for mixed model based approaches to quantitative trait locus (QTL) mapping in existing maize hybrid selection programs are presented: a restricted maximum likelihood (REML) and a Bayesian Markov Chain Monte Carlo (MCMC) approach. The methods use the in-silico-mapping procedure developed by Parisseaux and Bernardo (2004) as a starting point. The original single-point approach is extended to a multi-point approach that facilitates interval mapping procedures. For computational and conceptual reasons, we partition the full set of relationships from founders to parents of hybrids into two types of relations by defining so-called intermediate founders. QTL effects are defined in terms of those intermediate founders. Marker based identity by descent relationships between intermediate founders define structuring matrices for the QTL effects that change along the genome. The dimension of the vector of QTL effects is reduced by the fact that there are fewer intermediate founders than parents. Furthermore, additional reduction in the number of QTL effects follows from the identification of founder groups by various algorithms. As a result, we obtain a powerful mixed model based statistical framework to identify QTLs in genetic backgrounds relevant to the elite germplasm of a commercial breeding program. The identification of such QTLs will provide the foundation for effective marker assisted and genome wide selection strategies. Analyses of an example data set show that QTLs are primarily identified in different heterotic groups and point to complementation of additive QTL effects as an important factor in hybrid performance.

Abstract: "The Seventh Report of the Joint National Committee on Prevention, Detection, Evaluation, and Treatment of High Blood Pressure" provides a new guideline for hypertension prevention and management. The following are the key messages(1) In persons older than 50 years, systolic blood pressure (BP) of more than 140 mm Hg is a much more important cardiovascular disease (CVD) risk factor than diastolic BP; (2) The risk of CVD, beginning at 115/75 mm Hg, doubles with each increment of 20/10 mm Hg; individuals who are normotensive at 55 years of age have a 90% lifetime risk for developing hypertension; (3) Individuals with a systolic BP of 120 to 139 mm Hg or a diastolic BP of 80 to 89 mm Hg should be considered as prehypertensive and require health-promoting lifestyle modifications to prevent CVD; (4) Thiazide-type diuretics should be used in drug treatment for most patients with uncomplicated hypertension, either alone or combined with drugs from other classes. Certain high-risk conditions are compelling indications for the initial use of other antihypertensive drug classes (angiotensin-converting enzyme inhibitors, angiotensin-receptor blockers, β-blockers, calcium channel blockers); (5) Most patients with hypertension will require 2 or more antihypertensive medications to achieve goal BP (<140/90 mm Hg, or <130/80 mm Hg for patients with diabetes or chronic kidney disease); (6) If BP is more than 20/10 mm Hg above goal BP, consideration should be given to initiating therapy with 2 agents, 1 of which usually should be a thiazide-type diuretic; and (7) The most effective therapy prescribed by the most careful clinician will control hypertension only if patients are motivated. Motivation improves when patients have positive experiences with and trust in the clinician. Empathy builds trust and is a potent motivator. Finally, in presenting these guidelines, the committee recognizes that the responsible physician's judgment remains paramount.

Abstract: The National High Blood Pressure Education Program presents the complete Seventh Report of the Joint National Committee on Prevention, Detection, Evaluation, and Treatment of High Blood Pressure. Like its predecessors, the purpose is to provide an evidence-based approach to the prevention and management of hypertension. The key messages of this report are these: in those older than age 50, systolic blood pressure (BP) of greater than 140 mm Hg is a more important cardiovascular disease (CVD) risk factor than diastolic BP; beginning at 115/75 mm Hg, CVD risk doubles for each increment of 20/10 mm Hg; those who are normotensive at 55 years of age will have a 90% lifetime risk of developing hypertension; prehypertensive individuals (systolic BP 120-139 mm Hg or diastolic BP 80-89 mm Hg) require health-promoting lifestyle modifications to prevent the progressive rise in blood pressure and CVD; for uncomplicated hypertension, thiazide diuretic should be used in drug treatment for most, either alone or combined with drugs from other classes; this report delineates specific high-risk conditions that are compelling indications for the use of other antihypertensive drug classes (angiotensin-converting enzyme inhibitors, angiotensin-receptor blockers, beta-blockers, calcium channel blockers); two or more antihypertensive medications will be required to achieve goal BP (<140/90 mm Hg, or <130/80 mm Hg) for patients with diabetes and chronic kidney disease; for patients whose BP is more than 20 mm Hg above the systolic BP goal or more than 10 mm Hg above the diastolic BP goal, initiation of therapy using two agents, one of which usually will be a thiazide diuretic, should be considered; regardless of therapy or care, hypertension will be controlled only if patients are motivated to stay on their treatment plan.
