Abstract: Osteoporosis is a common disease in which the risk of fracture increases due to decreased bone mass and qualitative skeletal changes. Selective androgen receptor modulators (SARMs) are agonists with tissue selectivity, which act as partial or weak androgen receptor (AR) agonists in androgenic tissues, but mainly as complete AR agonists in synthetic metabolic tissues. In the recent 20 years, many scaffolds of SARMs have been reported, among which several molecules are promising and are undergoing clinical trial evaluation. However, it is still a challenge to discover SARMs with high activity and reduced side effects. In this review, not only are structure of SARMs reported in the literatures systematically collected and classified but also the structure-activity relationships (SAR) are systematically summarized. Furthermore, the advances in SARMs as potential treatment for osteoporosis are also updated.

Abstract: Simple Summary Anti-cancer treatments induced an increase in the childhood cancer survival rate. However, they are responsible for several long-term side effects in childhood cancer survivors, including osteoporosis. Cancer itself, a sedentary lifestyle, and an unhealthy diet might adversely affect bone health. Early identification and adequate management of bone fragility in childhood cancer survivors could be useful to prevent osteoporosis onset and consequently fragility fractures. Abstract The improvement of chemotherapy, radiotherapy, and surgical interventions, together with hematopoietic stem cell transplantation, increased childhood cancer survival rate in the last decades, reaching 80% in Europe. Nevertheless, anti-cancer treatments are mainly responsible for the onset of long-term side effects in childhood cancer survivors (CCS), including alterations of the endocrine system function and activity. In particular, the most frequent dysfunction in CCS is a metabolic bone disorder characterized by low bone mineral density (BMD) with increased skeletal fragility. BMD loss is also a consequence of a sedentary lifestyle, malnutrition, and cancer itself could affect BMD, thus inducing osteopenia and osteoporosis. In this paper, we provide an overview of possible causes of bone impairment in CCS in order to propose management strategies for early identification and treatment of skeletal fragility in this population.

Abstract: Selective serotonin reuptake inhibitors (SSRIs) are used to treat major depressive disorder (MDD) and other psychiatric disorders (e.g., obsessive compulsive disorder, social anxiety disorder, and panic disorder). In MDD treatment, SSRIs do not show remission in approximately 30% of patients, indicating a need for a better treatment option. Forced swimming test (FST) is a behavioral assay to evaluate depression‐like behavior and antidepressant efficacy in rodents. In the present study, we evaluated the combination effect of brexpiprazole with SSRIs on FST in mice, in order to investigate their synergistic effect.

Abstract: Abstract Background The lateral maxillary sinus augmentation (MSA) procedure has good predictability in terms of the success of bone regeneration with a low incidence of postoperative infections, estimated between 2% and 5.6%. Although the use of antibiotics is an established and standardized prophylactic measure for MSA procedures, the addition of corticosteroids still varies among clinician preference and clinical judgment. Purpose The aim of this systematic review was to identify whether the administration of corticosteroids during the MSA surgical procedure affects postoperative symptoms including swelling, pain, and infection rate. Materials and methods A literature search through PubMed, EMBASE, Ovid MEDLINE, and Web of Science indices, according to PICO criteria, was conducted to identify whether MSA peri‐operative use of corticosteroids reduces the incidence of complications and patient morbidity. A single arm meta‐analysis was performed due to the lack of randomized controlled trials (RCTs) comparing groups treated with or without peri‐operative corticosteroids. The intracluster correlation co‐efficient (ICC) and design effect were calculated to adjust for the clustering design. Results In the 37 studies included, a total of 1599 patients (378 Cort, 1221 No‐Cort) were analyzed. Before and after taking account of clustering, there was statistically significant effect of corticosteroids on swelling, pain, wound dehiscence, trismus, and hematoma. The complication rates postoperatively were comparable between the two study groups, however slight differences existed in the incidence of active suppuration (1.7% [95% CI 0.7–3.9] Cort vs. 3.2% [2.2–4.5] No‐Cort), wound dehiscence (3.9% [1.3–11.2] Cort vs. 2.1% [1.0–4.1] No‐Cort) and trismus (2.7% [0.8–8.4] Cort vs. 1.4% [0.8–2.5] No‐Cort). Conclusions Although the event rate of the 1‐to‐2‐week postoperative complications did not differ between the two groups, the lack of conclusive data and research comparing peri‐operative corticosteroid use makes it impossible to draw definitive conclusions and more evidence and studies designed for this specific purpose are needed.

Abstract: There are many factors to consider when clinicians prepare the alveolus ridge for optimal implant placement for form and function. From single tooth sites to full arch reconstruction, understanding bone and membrane properties, physiology of healing, and types of defects to reconstruct will allow clinicians to choose the proper regenerative technique. This chapter discusses common commercially available materials and literature reviews to seek out the best current choice for each type of augmentation. A xenograft is defined as a graft from genetically different species other than human. The idea of ridge preservation is simply placing bone-grafting material within the socket after tooth extraction. Guided bone regeneration can be performed simultaneously during implant placement, for ridge development or for treatment of peri-implantitis. Whether the alveolus ridge lacks width and/or height, onlay grafts can be used to augment these deficiencies. Three to four wall defects with an inferior stop can be remedied with particulate onlay graft.
