Regulatory T-cell Depletion Alters the Tumor Microenvironment and Accelerates Pancreatic Carcinogenesis.
Yaqing Zhang,Jenny Lazarus,Nina Steele,Wei Yan,Ho-Joon Lee,Zeribe C. Nwosu,Christopher J. Halbrook,Rosa E. Menjivar,Samantha Kemp,Veerin Sirihorachai,Ashley Velez-Delgado,Katelyn Donahue,Eileen S. Carpenter,Kristee Brown,Valerie Irizarry-Negron,Anna C. Nevison,Alekya Vinta,Michelle A. Anderson,Howard C. Crawford,Costas A. Lyssiotis,Timothy L. Frankel,Filip Bednar,Marina Pasca di Magliano +22 more
TL;DR: It is shown that Tregs are a key source of TGFβ ligands and, accordingly, their depletion reprogramed the fibroblast population, with loss of tumor-restraining, smooth muscle actin-expressing fibroblasts (myCAFs), and supports the notion that fibro Blasts are a heterogeneous population with different and opposing functions in pancreatic carcinogenesis.
Journal ArticleDOI
Multimodal mapping of the tumor and peripheral blood immune landscape in human pancreatic cancer
Nina Steele,Eileen S. Carpenter,Samantha Kemp,Veerin Sirihorachai,Lawrence Delrosario,Jenny Lazarus,El-ad David Amir,Valerie Gunchick,Carlos Espinoza,Samantha Bell,Lindsey Harris,Fatima Lima,Valerie Irizarry-Negron,Daniel Paglia,Justin Macchia,Angel Ka Yan Chu,Heather K. Schofield,Erik Jan Wamsteker,Richard S. Kwon,Allison R. Schulman,Anoop Prabhu,Ryan Law,Arjun R. Sondhi,Jessica Yu,Arpan Patel,Katelyn Donahue,Hari Nathan,Clifford S. Cho,Michelle A. Anderson,Vaibhav Sahai,Costas A. Lyssiotis,Weiping Zou,Benjamin L. Allen,Arvind Rao,Howard C. Crawford,Filip Bednar,Timothy L. Frankel,Marina Pasca di Magliano +37 more
TL;DR: Tumor-infiltrating CD8+ T cells had an increased proportion of cells expressing an exhausted expression profile that included upregulation of the immune checkpoint TIGIT, a finding that was validated at the protein level.
Journal ArticleDOI
The role of endoscopy in the management of choledocholithiasis
John T. Maple,Laurel Fisher,Norio Fukami,Joo Ha Hwang,Rajeev Jain,Terry L. Jue,Khalid Khan,Mary L. Krinsky,Phyllis M. Malpas,Tamir Ben-Menachem,Ravi Sharaf,Jason A. Dominitz,Steven O. Ikenberry,Michelle A. Anderson,Vasundhara Appalaneni,G. Anton Decker,Dayna S. Early,John A. Evans,Robert D. Fanelli,Deborah A. Fisher +19 more
TL;DR: This guideline is intended to be an educational device o provide information that may assist endoscopists in roviding care to patients that is based on both the aggregate evidence quality and an assessment of the anticipated benefits and harms.
Journal ArticleDOI
Role of endoscopy in the management of GERD.
David R. Lichtenstein,Brooks D. Cash,Raquel E. Davila,Todd H. Baron,Douglas G. Adler,Michelle A. Anderson,Jason A. Dominitz,Seng Ian Gan,M. Edwyn Harrison,Steven O. Ikenberry,Waqar A. Qureshi,Elizabeth Rajan,Bo Shen,Marc J. Zuckerman,Robert D. Fanelli,Trina VanGuilder +15 more
TL;DR: In this paper, the authors recommend that endoscopic findings of reflux esophagitis be classified according to an accepted grading scale or described in detail, without the use of diagnostic testing, including EGD.
Journal ArticleDOI
Mechanisms of CFTR functional variants that impair regulated bicarbonate permeation and increase risk for pancreatitis but not for cystic fibrosis.
Jessica LaRusch,Jinsei Jung,Ignacio J. General,Michele D. Lewis,Hyun Woo Park,Randall E. Brand,Andres Gelrud,Michelle A. Anderson,Peter A. Banks,Darwin L. Conwell,Christopher Lawrence,Joseph Romagnuolo,John Baillie,Samer Alkaade,Gregory A. Cote,Timothy B. Gardner,Stephen T. Amann,Adam Slivka,Bimaljit S. Sandhu,Amy Aloe,Michelle L. Kienholz,Dhiraj Yadav,M. Michael Barmada,Ivet Bahar,Min Goo Lee,David C. Whitcomb +25 more
TL;DR: WNK1-SPAK pathway-activated increases in CFTR bicarbonate permeability are altered by CFTRBD variants through multiple mechanisms, and molecular dynamics simulations suggest physical restriction of the CFTR channel and altered dynamic channel regulation.
