Journal ArticleDOI
Integrating predicted transcriptome from multiple tissues improves association detection.
Alvaro N. Barbeira,Milton Pividori,Jiamao Zheng,Heather E. Wheeler,Dan L. Nicolae,Hae Kyung Im +5 more
TL;DR: This work proposes an efficient statistical method (MultiXcan) that leverages the substantial sharing of eQTLs across tissues and contexts to improve the ability to identify potential target genes and develops a robust implementation of the summary-based extension.
Journal ArticleDOI
Shared and Distinct Genetic Risk Factors for Childhood-Onset and Adult-Onset Asthma: Genome-Wide and Transcriptome-Wide Studies
TL;DR: It is suggested that the establishment of disease in children is driven more by dysregulated allergy and epithelial barrier function genes, whereas the cause of adult-onset asthma is more lung-centred and environmentally determined, but with immune-mediated mechanisms driving disease progression in both children and adults.
Journal ArticleDOI
Exploiting the GTEx resources to decipher the mechanisms at GWAS loci.
Alvaro N. Barbeira,Rodrigo Bonazzola,Eric R. Gamazon,Yanyu Liang,YoSon Park,Sarah Kim-Hellmuth,Sarah Kim-Hellmuth,Gao Wang,Zhuoxun Jiang,Dan Zhou,Farhad Hormozdiari,Farhad Hormozdiari,Boxiang Liu,Abhiram Rao,Andrew R. Hamel,Andrew R. Hamel,Milton Pividori,François Aguet,Lisa Bastarache,Daniel M. Jordan,Marie Verbanck,Ron Do,Matthew Stephens,Kristin G. Ardlie,Mark I. McCarthy,Stephen B. Montgomery,Ayellet V. Segrè,Ayellet V. Segrè,Christopher D. Brown,Tuuli Lappalainen,Xiaoquan Wen,Hae Kyung Im +31 more
TL;DR: This article presented an in-depth examination of the phenotypic consequences of transcriptome regulation and a blueprint for the functional interpretation of genome-wide association study-discovered loci.

TL;DR: An in-depth examination of the phenotypic consequences of transcriptome regulation and a blueprint for the functional interpretation of genome-wide association study-discovered loci are presented and widespread dose-dependent effects of RNA expression and splicing are demonstrated.

Integrating Predicted Transcriptome From Multiple Tissues Improves Association Detection
Alvaro N. Barbeira,Milton Pividori,Jiamao Zheng,Heather E. Wheeler,Dan L. Nicolae,Hae Kyung Im +5 more
TL;DR: A new method is developed to test the effect of gene expression regulation on complex traits, integrating information available across multiple tissue studies, and it is shown this approach has higher power than traditional single-tissue methods.
