TL;DR: The dissection of the alcoholism phenotype, to disentangle the influence of comorbid substance-use disorders, will be a next step in identifying genetic variants associated with alcohol dependence.
Journal ArticleDOI
A genome-wide association study confirms PNPLA3 and identifies TM6SF2 and MBOAT7 as risk loci for alcohol-related cirrhosis
Stephan Buch,Felix Stickel,Eric Trepo,Michael J. Way,A Herrmann,Hans Dieter Nischalke,Mario Brosch,Jonas Rosendahl,Thomas Berg,Monika Ridinger,Marcella Rietschel,Andrew McQuillin,Josef Frank,Falk Kiefer,Stefan Schreiber,Wolfgang Lieb,Michael Soyka,Nasser Semmo,Elmar Aigner,Christian Datz,Renate Schmelz,Stefan Brückner,Sebastian Zeissig,Anna-Magdalena Stephan,Norbert Wodarz,Jacques Devière,Nicolas Clumeck,Christoph Sarrazin,Frank Lammert,Thierry Gustot,Pierre Deltenre,Pierre Deltenre,Henry Völzke,Markus M. TL;DR: A genome-wide association study for alcohol-related cirrhosis in individuals of European descent with subsequent validation in two independent European cohorts is performed, suggesting that lipid turnover is important in the pathogenesis of alcohol- related Cirrhosis.
Journal ArticleDOI
Genome-wide association study of alcohol dependence.
Jens Treutlein,Sven Cichon,Monika Ridinger,Norbert Wodarz,Michael Soyka,Peter Zill,Wolfgang Maier,Rainald Moessner,Wolfgang Gaebel,Norbert Dahmen,Christoph Fehr,Norbert Scherbaum,Michael Steffens,Kerstin U. Sullivan,Markus M. Nöthen,Falk Kiefer,Rainer Spanagel,Karl Mann,Marcella Rietschel +28 more
TL;DR: This is the first GWAS and follow-up study to identify a genome-wide significant association in alcohol dependence and nine SNPs were located in genes that have been reported to be associated with alcohol dependence, including the CDH13 and ADH1C genes.
Journal ArticleDOI
Genome-wide association study of alcohol dependence:significant findings in African- and European-Americans including novel risk loci
Joel Gelernter,Henry R. Kranzler,Richard Sherva,Laura Almasy,Ryan Koesterer,Andrew H. Smith,Raymond F. Anton,Ulrich W. Preuss,Monika Ridinger,Dan Rujescu,Norbert Wodarz,Peter Zill,Hongyu Zhao,Lindsay A. Farrer +13 more
TL;DR: A GWAS of alcohol dependence in European-American and African-American populations, with replication in independent samples of EAs, AAs and Germans, finds a convergence of risk genes between populations, and, to a lesser extent, between psychiatric traits.
Journal ArticleDOI
Genetic variation in the PNPLA3 gene is associated with alcoholic liver injury in caucasians

TL;DR: Genotype PNPLA3 rs738409(GG) is associated with alcoholic liver cirrhosis and elevated aminotransferase levels in alcoholic Caucasians.
