HDAC2 negatively regulates memory formation and synaptic plasticity
Ji-Song Guan,Stephen J. Haggarty,Emanuela Giacometti,Jan Hermen Dannenberg,Jan Hermen Dannenberg,Nadine F. Joseph,Nadine F. Joseph,Jun Gao,Thomas J.F. Nieland,Ying Zhou,X. L. Wang,Ralph Mazitschek,James E. Bradner,Ronald A. DePinho,Rudolf Jaenisch,Li-Huei Tsai,Li-Huei Tsai +16 more
TL;DR: It is suggested that HDAC2 functions in modulating synaptic plasticity and long-lasting changes of neural circuits, which in turn negatively regulates learning and memory.
Journal ArticleDOI
An epigenetic blockade of cognitive functions in the neurodegenerating brain
Johannes Gräff,Damien Rei,Damien Rei,Ji-Song Guan,Ji-Song Guan,Wenyuan Wang,Wenyuan Wang,Jinsoo Seo,Jinsoo Seo,Krista M. Hennig,Krista M. Hennig,Thomas J.F. Nieland,Daniel M. Fass,Daniel M. Fass,Patricia F. Kao,M C Kahn,Susan C. Su,Susan C. Su,Alireza Samiei,Nadine F. Joseph,Nadine F. Joseph,Stephen J. Haggarty,Stephen J. Haggarty,Ivana Delalle,Li-Huei Tsai,Li-Huei Tsai +25 more
TL;DR: Reversing the build-up of histone deacetylase 2 by short-hairpin-RNA-mediated knockdown unlocks the repression of genes, reinstates structural and synaptic plasticity, and abolishes neurodegeneration-associated memory impairments.

Journal ArticleDOI
Epigenetic Priming of Memory Updating during Reconsolidation to Attenuate Remote Fear Memories
Johannes Gräff,Nadine F. Joseph,Nadine F. Joseph,Meryl E. Horn,Alireza Samiei,Jia Meng,Jia Meng,Jinsoo Seo,Jinsoo Seo,Damien Rei,Damien Rei,Adam W. Bero,Adam W. Bero,Trongha X. Phan,Trongha X. Phan,Florence F. Wagner,Edward Holson,Jinbin Xu,Jianjun Sun,Rachael L. Neve,Robert H. Mach,Stephen J. Haggarty,Li-Huei Tsai,Li-Huei Tsai +23 more
TL;DR: In this paper, the authors show that, whereas recent memory recall induces a limited period of hippocampal neuroplasticity mediated, in part, by S-nitrosylation of HDAC2 and histone acetylation, such plasticity is absent for remote memories.
Epigenetic Priming of Memory Updating during Reconsolidation to Attenuate Remote Fear Memories
Johannes Gräff,Nadine F. Joseph,Nadine F. Joseph,Meryl E. Horn,Alireza Samiei,Jia Meng,Jia Meng,Jinsoo Seo,Jinsoo Seo,Damien Rei,Damien Rei,Adam W. Bero,Adam W. Bero,Trongha X. Phan,Trongha X. Phan,Florence F. Wagner,Edward Holson,Jinbin Xu,Jianjun Sun,Rachael L. Neve,Robert H. Mach,Stephen J. Haggarty,Li-Huei Tsai,Li-Huei Tsai +23 more
TL;DR: It is shown that, in mice, successful reconsolidation-updating paradigms for recent memories fail to attenuate remote memories, but by using an HDAC2-targeting inhibitor (HDACi) during reconsideration, even remote memories can be persistently attenuated.
