Abstract: Engineered metalloenzymes represent promising catalysts for stereoselective C-H functionalization reactions. Recently, P450 enzymes have been evolved to allow for new-to-nature intramolecular C(sp3)-H amination reactions via a nitrene transfer mechanism, giving rise to diamine derivatives with excellent enantiocontrol. To shed light on the origin of enantioselectivity, a combined computational and experimental study was carried out. Hybrid quantum mechanics/molecular mechanics calculations were performed to investigate the activation energies and enantioselectivities of both the hydrogen atom transfer (HAT) and the subsequent C-N bond forming radical rebound steps. Contrary to previously hypothesized enantioinduction mechanisms, our calculations show that the radical rebound step is enantioselectivity-determining, whereas the preceding HAT step is only moderately stereoselective. Furthermore, the selectivity in the initial HAT is ablated by rapid conformational change of the radical intermediate prior to C-N bond formation. This finding is corroborated by our experimental study using a set of enantiomerically pure, monodeuterated substrates. Furthermore, classical and ab initio molecular dynamics simulations were carried out to investigate the conformational flexibility of the carbon-centered radical intermediate. This key radical species undergoes a facile conformational change in the enzyme active site from the pro-(R) to the pro-(S) configuration, whereas the radical rebound is slower due to the spin-state change and ring strain of the cyclization process, thereby allowing stereoablative C-N bond formation. Together, these studies revealed an underappreciated enantioinduction mechanism in biocatalytic C(sp3)-H functionalizations involving radical intermediates, opening up new avenues for the development of other challenging asymmetric C(sp3)-H functionalizations.

Neste trabalho abordo especificamente a concepcao da autora acerca do uso da linguagem ou discurso dos direitos pelos grupos subalternizados em funcao da raca. Tal enfoque, alem de permitir a reflexao sobre o tema no âmbito da producao juridica brasileira, contribuira para que o trabalho cumpra dois objetivos: apresentar as principais caracteristicas do processo de teorizacao da autora e permitir que conhecamos um dos pontos fundamentais da controversia entre o Critical Race Theory e o Critical Legal Studies.

Abstract: RESUMO O objetivo deste artigo e analisar a forma como os mecanismos legais do antirracismo brasileiro tem funcionado e os principais obstaculos a sua aplicacao vis-a-vis a dinâmica social do estigma e insultos raciais. Nosso argumento e que existe uma dificuldade em conciliar as categorias da lei interpretadas pelos juizes com a forma real do racismo brasileiro no qual prevalece atos sutis de discriminacao assim como o uso de insultos raciais em situacoes cotidianas.

Abstract: Selective functionalization of aliphatic C-H bonds, ubiquitous in molecular structures, could allow ready access to diverse chemical products. While enzymatic oxygenation of C-H bonds is well established, the analogous enzymatic nitrogen functionalization is still unknown; nature is reliant on preoxidized compounds for nitrogen incorporation. Likewise, synthetic methods for selective nitrogen derivatization of unbiased C-H bonds remain elusive. In this work, new-to-nature heme-containing nitrene transferases were used as starting points for the directed evolution of enzymes to selectively aminate and amidate unactivated C(sp3)-H sites. The desymmetrization of methyl- and ethylcyclohexane with divergent site selectivity is offered as demonstration. The evolved enzymes in these lineages are highly promiscuous and show activity toward a wide array of substrates, providing a foundation for further evolution of nitrene transferase function. Computational studies and kinetic isotope effects (KIEs) are consistent with a stepwise radical pathway involving an irreversible, enantiodetermining hydrogen atom transfer (HAT), followed by a lower-barrier diastereoselectivity-determining radical rebound step. In-enzyme molecular dynamics (MD) simulations reveal a predominantly hydrophobic pocket with favorable dispersion interactions with the substrate. By offering a direct path from saturated precursors, these enzymes present a new biochemical logic for accessing nitrogen-containing compounds.

Abstract: Abstract The conversion of C‒H bonds to C‒N bonds offers a sustainable and economical strategy for the synthesis of nitrogen-containing compounds. However, challenges regarding the control of regio- and stereoselectivity currently limit the broad applicability of intermolecular C( sp 3 )‒H amination reactions. We address these restrictions by directed nitrene-mediated C‒H insertion using a metal-coordinating functional group. We report a highly stereocontrolled, iron-catalysed direct α-amination of abundant carboxylic acid feedstock molecules. The method provides in a single step high-value N -Boc-protected α-monosubstituted and α,α-disubstituted α-amino acids, which can then be immediately used for applications including solution- and solid-phase peptide synthesis. This method fulfils important aspects of sustainability by being highly step efficient and utilizing non-toxic, Earth-abundant iron as the catalytic metal.

Abstract: Resumo Este artigo se situa na intersecao dos campos disciplinares do direito e das ciencias sociais e sonda o tratamento institucional da intolerância religiosa no Brasil, particularmente no estado de Minas Gerais. Trata-se de pesquisa empirica que privilegia a analise qualitativa e quantitativa de dados primarios, retirados de boletins de ocorrencia registrados entre 2016 e 2018, e recorre, principalmente, aos procedimentos da coleta de dados e analise de conteudo. O trabalho restitui os principais resultados de uma pesquisa de pos-doutorado que visou, inicialmente, identificar o modo pelo qual se da a traducao das demandas baseadas em intolerância religiosa do codigo social para o codigo especializado do direito. Objetivo acrescido no processo de investigacao foi o de analisar a possibilidade de emergencia da intolerância religiosa como um problema publico, nos termos desenvolvidos por Daniel Cefai, em face do tratamento que lhe e dispensado pelas instituicoes de seguranca publica. Os resultados apontam para dificuldades na referida traducao, suscetiveis de inviabilizar a emergencia da intolerância religiosa como um problema publico, bem como para a oportunidade de reflexao acerca de outras tecnologias sociais, nao juridicas, para abordar a questao da intolerância.

Neumann centraliza a pessoa como principio fundamental do Estado de Direito, em razao da possibilidade de responsabilizacao sobre a dominacao. O sujeito de direitos e compreendido entao como centro de imputacao de direitos e deveres e autor das normas que regulam sua vida. Foucault analisa o poder como forma dinâmica e circular nas relacoes sociais. Judith Butler contribui com seu conceito de resistencia e analisa a pratica da critica como uma relacao social do sujeito com as normas. No interior do direito, podemos compreende-la como uma reivindicacao das normas com o objetivo de transforma-las, a fim de possibilitar a expressao de diferentes formas de vida. Sugerimos analisar, em suma, a possibilidade de uma gramatica da regulacao social e a ideia de pessoa no direito como propulsora de um processo de construcao autonoma e constante de criterios normativos, com base na formulacao de igualdade dos sujeitos perante o direito. Abstract This paper examines the effects of the idea of person on Law, based on a dialogue between the critical view of Evguieni Pachukanis, for whom legal subject is merely a way of enabling capital domination, and the critical contributions of Franz Neumann, Michel Foucault and Judith Butler. Neumann centralizes the person as a fundamental principle of rule of law because the possibility of accountability over domination. Legal subject is then understood as the centre of imputation of rights and duties and author of norms that regulates his life. Foucault analyzes power as a dynamic and circular form in social relations. Judith Butler contributes with her concept of resistance and analyzes the practice of criticism as a social relation of the subject to the norms. Within Law, we can understand it as a claim to norms with the aim of transforming them, in order enable the expression of different life forms. We suggest to analyze, in short, the possibility of a social regulation grammar as well the idea of person as propellant of an autonomous and constant process of normative criteria construction, based on the formulation of equality of subjects before Law.
