TL;DR: In this article, the authors present characteristics and subgroup analyses from the first 1257 patients enrolled in the study, and conclude that there are no differences in outcomes of patients with short vs long latency of DILI.
Journal ArticleDOI
The human pregnane X receptor: genomic structure and identification and functional characterization of natural allelic variants.
Jiong Zhang,Peter M. Kuehl,Eric D. Green,Jeffery W. Touchman,Paul B. Watkins,Ann K. Daly,Stephen D. Hall,Patrick Maurel,M. V. Relling,Cynthia Brimer,Kazuto Yasuda,Steven A. Wrighton,Michael L. Hancock,Richard B. Kim,Stephen C. Strom,Kenneth E. Thummel,Christopher G. Russell,James R. Hudson,Erin G. Schuetz,Mark S. Boguski +19 more
TL;DR: The relevance of each of the 38 PXR SNPs identified in DNA of individuals whose CYP3A basal and rifampin-inducible CYP 3A4 expression was determined in vivo and/or in vitro was demonstrated by univariate statistical analysis.
Journal ArticleDOI
Minocycline hepatotoxicity: Clinical characterization and identification of HLA- B*35:02 as a risk factor
Thomas J. Urban,Thomas J. Urban,Paola Nicoletti,Naga Chalasani,Jose Serrano,Andrew Stolz,Ann K. Daly,Guruprasad P. Aithal,John F. Dillon,Víctor M. Navarro,Joseph A. Odin,Huiman X. Barnhart,David A. Ostrov,Nanye Long,Nanye Long,Elizabeth T. Cirulli,Paul B. Watkins,Paul B. Watkins,Robert J. Fontana +18 more
TL;DR: It is demonstrated that individuals who are HLA-B∗35:02 carriers are at increased risk of developing minocycline related liver injury and these results may help doctors more rapidly and confidently diagnose affected patients and possibly reduce the risk of liver injury in individuals receiving minocyCline going forward.
Journal ArticleDOI
Sensitivity to hepatotoxicity due to epigallocatechin gallate is affected by genetic background in diversity outbred mice.
Rachel J. Church,Daniel M. Gatti,Thomas J. Urban,Nanye Long,Xi Yang,Qiang Shi,J. Scott Eaddy,Merrie Mosedale,Shawn Ballard,Gary A. Churchill,Victor Navarro,Paul B. Watkins,David W. Threadgill,Alison H. Harrill +13 more
TL;DR: The data indicate that the Diversity Outbred mice may provide a platform for informing risk of rare, adverse reactions that may occur in consumer populations upon ingestion of concentrated herbal products.
Minocycline Hepatotoxicity: Clinical characterization and identification of HLA-B* 35:02 as a risk factor
Thomas J. Urban,Thomas J. Urban,Paola Nicoletti,Naga Chalasani,Jose Serrano,Andrew Stolz,Ann K. Daly,Guruprasad P. Aithal,John F. Dillon,Víctor M. Navarro,Joseph A. Odin,Huiman X. Barnhart,David A. Ostrov,Nanye Long,Nanye Long,Elizabeth T. Cirulli,Paul B. Watkins,Paul B. Watkins,Robert J. Fontana +18 more
TL;DR: In this paper, the authors identify genetic determinants of minocycline DILI in a well-phenotyped cohort of patients using Auto-Dock Vina.
